    Notes: The first practical total chemical synthesis of a plasmenylcholine (1-O-1'-(Z)-hexadecenyl-2-hexadecanoyl-sn-glycero-3-phosphocholine) with pure (Z) olefin stereochemistry is reported. Monopalmitin was doubly protected as the 3-TBDPS-2-TBDMS ethers (tert-butyldiphenylsilyl-, tert-butyldimethylsilyl-) and converted to the corresponding 1-O-1′-(Z)-hexadecenyl-2-TBDMS-3-TBDPS-glyceryl ether (by the method of ref. [43]). Clean deprotection with tetra-butylammonium fluoride in the presence of imidazole gave 1-O-1′-(Z)-hexadecenylglycerol in 〉90% yield. Resilylation with TBDPSCl followed by acylation of the sn-2 alcohol with palmitoyl chloride and deprotection of the resulting 3-TBDPS-2-hexadecanoyl-1-O-1′-(Z)-hexadecenylglycerol at -20°C with Bu4NF gave 2-hexadecanoyl-1-O-1′-(Z)-hexadecenylglycerol in 86% yield. The 3-phosphocholine group was attached by phosphorylating the free hydroxyl with 2-chloro-2-oxo-1,3,2-dioxaphospholane in the presence of pyridine, instead of Et3N, as base to avoid acyl migration; the dioxaphospholane triester intermediate was subsequently cleaved with Me3N to give 1-O-1′-(Z)-hexadecenyl-2-hexadecanoyl-sn-glycero-3-phosphocholine in 18% overall yield from monopalmitin. The efficiency and flexibility of this route makes it well-suited to the preparation of a wide variety of 1-, 2-, and 3-substituted as well as isotopically labeled plasmenylcholines for biophysical and biochemical studies.

    Notes: Epidermal growth factor stimulates phosphatidylinositol turnover in human foreskin fibroblasts. This is a primary cell culture with normal numbers of epidermal growth factor receptors that is stimulated to divide by epidermal growth factor. Increases are seen in the inositol phospholipids and inositol phosphates. Despite this activation of phosphatidylinositol turnover, there is no detectable activation of protein kinase C.

    Notes: The lignin peroxidases (LIP) and manganese peroxidases (MNP) of Phanerochaete chrysosporium catalyze a wide range of lignin depolymerization reactions with lignin models and synthetic lignins in solution. However, their ability to degrade insoluble natural lignin in aqueous media has not been demonstrated. Insoluble isolated poplar lignin similar to natural lignin was treated in vitro in aqueous media for 12 h with LIP, MNP, and both. Treatment with MNP alone slightly increased the solid mass and produced measurable amounts of lignin-derived 2,6-dimethoxyhydroquinone and 2-methoxyhydroquinone but did not appreciably decrease the total lignin content. Treatment with LIP alone did not decrease the mass but produced measurable amounts of lignin-derived p-hydroxybenzoic acid and slightly decreased the lignin content. Finally, treatment with LIP and MNP together decreased the solid mass by 11%, decreased the lignin content by 5%, and released low-concentration compounds with mass spectra containing the typical lignin-derived electron-impact fragments of mass 107, 137, 151, 167, and 181. These results suggest that MNP increases the effectiveness of LIP-mediated lignin degradation. © 1998 John Wiley & Sons, Inc. Biotechnol Bioeng 57: 704-717, 1998

    Notes: During long-term operation of a biofilter, the mandatory absence of net cell growth forces the cells into maintenance metabolism, which is of relatively low rate compared to substrate consumption during the active growth of the acclimation phase. A model based on this shift in metabolism can explain the postacclimation decrease in activity sometimes reported for biofilters. The cessation of growth can be caused by nutrient depletion in the bed. Postacclimation nutrient addition increases activity primarily by allowing a return to the high substrate consumption rate of active growth, and only secondarily helps raise bed activity because of the ultimately higher amount of biomass in the bed. Simulations incorporating the acclimation period and the role of maintenance metabolism predict about 4 logarithms of growth during acclimation of a hexane biofilter, which was confirmed experimentally. Changes in a biofilter's biomass during the acclimation phase can be estimated from substrate conversion data using two approximate methods. The first follows the cumulative amount of substrate converted and uses the estimated yield of cells from substrate during active growth to estimate the total biomass created.     Notes: The 1H and 31P NMR spectra of a series of compounds containing the PIII-N-PV skeleton including Cl2PNMeP(Z)Cl2 (Z=O or S), Cl2P·NMe·P(O)CINMe2, P4(NMe)6Zn (Z=S, n = 1-3; Z=Se, n = 1), XP(NBut)2P(Z)X (X=Cl, Z=O or S; X=OMe, Z=S or Se; X=NMe2, Z=S or Se; X=NEt2, Z=Se), (X=O or S), and Ph2PNRP(S)Ph2 (R=Me, Et) have been obtained. 1H-{31P} double resonance, and in selected cases, 31P-{1H, 77Se} and 31P-{1H, 31P} triple resonance experiments, indicate that 2J(PIII NPV) is positive in acyclic compounds, negative in most cyclic or cage compounds, and furthermore, is related to the conformation adopted by the PIII-N bond.

    Notes: The coupling constants nJ(77Se31P), n = 1-4, have been measured in the proton-decoupled 31P NMR spectra of a range of diphosphorus selenides and diselenides. 31P-{1H, 31P} and 31P-{1H, 77Se} triple resonance experiments have been used to establish the signs of the coupling constants, and it is found that both the magnitudes and signs depend upon the stereochemical relationship of the coupled nuclei.
