Notes: Electro-optic measurements are reported for the polynucleotides poly A, poly U, poly G, and NaDNA, and their complexes with acridine orange (AO). Measurements were also made on the methylene blue (MB) complex with NaDNA. Poly U, poly G, and NaDNA complexes with AO as well as the NaDNA-MB complex were found to exhibit positive birefringence and perpendicular dichroism indicating the dye molecules are oriented with their long axes perpendicular to the applied electric field. The opposite was found for the AO complex with poly A, which showed positive birefringence and parallel dichroism, indicating that in this case the dye molecules are oriented with their long axes parallel to the field.

Notes: Poly(t-butyl methacrylate-b-isoprene-b-t-butyl methacrylate) block copolymers have been synthesized in a controlled manner by anionic techniques. The block copolymers demonstrated predictable chemical composition and stereochemistry, and narrow molecular weight distributions. The addition of a polar solvent was required after complete conversion of the diene block in order to preserve the high 1,4 microstructure of the diene phase and to facilitate the efficient crossover to the poly(t-butyl methacrylate) lithium enolate. Thermomechanical analysis (TMA) of the triblock copolymers demonstrated a microphase separated morphology. The glass transition temperatures of the diene phase and acrylic phase were approximately -70 and 120°C, respectively. Cast films of the block copolymers from polar and nonpolar solvents were optically clear and elastic.

Notes: Explicit expressions for the DK and RK associated with the excitation of regular oligomers from their ground states to the Kth exciton state as functions of N, P, μ, μ∥, μt, and μr have been derived on the assumption that the only perturbation Vij to the N singly excited states results from exciton transfer between adjacent residues, V12. The DK relation predicts the occurrence of a perpendicularly polarized absorption band arising from μ⊥ and a parallel polarized band from μ∥ with maxima at K = [2(N + 1)/P] and 1, which are shifted from the monomer band by 2V12 cos(2π/P) and 2V12cos[π/(N + 1)], respectively. Overlapping of the bands, which may occur in oligomers, decreases rapidly with N, leaving two perfectly polarized bands in the polymer. Within each band which includes only a few levels near the maxima, the most probable transition may be associated with all of the intensity for very short chains but only 40.2-50% (⊥) and 80.9% (∥) as N → ∞ (polymer case). A relation was derived by which V12 may be simply calculated from μ, μr, μt, μ∥, a, and z. The nearest neighbor theory was compared with experiment by calculating a reasonable set of the relevant parameters which exactly account for known spectral shifts.

Notes: The coulombic interaction between pairs of trinucleotides has been computed from quantum theory. The results are discussed in terms of their relation to hydrogen bonds and stacking in such systems and to the mechanism by which messenger RNA codons recognize transfer RNA nodocs.

Notes: We have studied cross-linking and thermal degradation of high-performance first-and second-generation PMR-15 polyimides, both thermoset and thermoplastic versions, by performing nonspectroscopic NMR solid echo T*2 relaxation measurements at temperatures up to 430°C using probes built for this purpose. We employ signal averaging and automated decomposition of the relaxation decays into two Gaussian components, the slower of which gradually appears above 300°C. Tracking the molecular mobility spectrum in terms of the relative intensity of the components and their relaxation times as temperature is cycled, we detect essentially no irreversible effects below the glass transition, measure permanent mobility reductions attributable to completion of cure, and find that exposure to temperatures above 380°C on the order of 1 h is required for substantial thermal degradation to occur. These results are closely supported by thermal and mechanical measurements on parallel specimens. Second-generation PMR resins appear to have higher microscopic rigidity and reduced viscous fraction at high temperatures. ©1995 John Wiley & Sons, Inc.

Notes: Intravascular trophoblast (IVT) cells, derived from the trophoblast of the developing hamster embryo, are known to migrate in retrograde fashion into the uterine arteries. There they migrate to a certain point, destroy and replace the endothelial lining, and modify the smooth muscle of the arteries. The dilated vessels that result presumably enhance the flow of blood to the placental exchange area. The morphology of IVT cells in the hamster placenta was investigated by scanning and transmission electron microscopy. Although occasional single migrating cells were observed, the IVT generally appear as sheets of large, contiguous, sometimes overlapping cells that spread over the endothelial surfaces of the uterine central terminal arteries and vascular knot arteries. This process seems to be aided by the appearance of filopodia, which make contact either with other intravascular trophoblast cells or the endothelium. After consolidation, the IVT cells act as a functional part of the vessel lining and are readily distinguished from the surrounding endothelium by their numerous microvilli. The final distribution of the IVT cells is patchy rather than uniform. © 1994 Wiley-Liss, Inc.

Notes: We have developed a simple and rapid method for the selective synthesis of tetra-tertiarybutyldimethylsilyl (TBDMS) glutamine, which allows the simultaneous quantitation of glutamine (2-15N) and (5-15N) isotopic enrichment by selected ion monitoring (SIM) gas chromatographic/mass spectrometric analysis. The tetra-TBDMS glutamine (4S-gln) has an electron impact mass fragment at m/z 258 that contains only the amino-N and larger fragments (e.g., at m/z 545) that have both nitrogens. Derivatization with acetonitrile (ACN) and N-methyl-N-(tert-butyldimethylsilyl)trifluoroacetamide yields primarily tri-TBDMS glutamine (3S-gln) and quantities of 4S-gln too small to allow accurate SIM and tracer/tracee ratio determinations. However, when N,N-dimethylformamide, a more polar aprotic solvent, was substituted for ACN and the sample was heated for 30 min at 125°C, greater than 80% of derivatized glutamine appeared as 4S-gln. Derivatized plasma samples that had been mixed with amide- and/or amino-15N glutamine and analyzed by SIM demonstrated strong agreement (r ≥ 0.998, p = 0.0001) between theoretical and observed enrichment values for the 4S-gln fragments at m/z 258 and 545. Deamidation of glutamine to glutamate is negligible during sample processing and analysis. This procedure will facilitate the investigation of the specific sources and fates of glutamine amide and amino nitrogen as well as stable isotope studies involving amino acid transamination, ammonia clearance, urea production and other areas of nitrogen metabolism.

Notes: The utility of anisotropic ultrafiltration membranes in gas separation has been examined. These membranes consist of a thin (0.2-2μ) barrier layer supported upon a more porous supporting substructure. Gases permeate these membranes by a Knudsen diffusion mechanism. Permeabilities from 0.1 to 0.4 cm sec-1 cm Hg-1 were obtained. These values are 104 to 105 higher than most conventional gas separation membranes. These membranes may be useful in several gas separation processes.
