    Notes: Continuous digitalized signals such as spectra, electrophoregrams or chromatograms generally have a large number of data points and contain redundant information. It is therefore troublesome performing discriminant analysis without any preliminary selection of variables. A procedure for the application of canonical discriminant analysis (CDA) on this kind of data is studied. CDA can be presented as a succession of two principal component analyses (PCAs). The first is performed directly on the raw data and gives PC scores. The second is applied on the gravity centres of each qualitative group assessed on the normalized PC scores. A stepwise procedure for selection of the relevant PC scores is presented. The method has been tested on an illustrative collection of 165 size-exclusion high-performance (SE-HPLC) chromatograms of proteins of wheat belonging to 55 genotypes and grown in three locations. The discrimination of the growing locations was performed using seven to nine PC scores and gave more than 86% accurate classifications of the samples both in the training sets and the verification sets. The genotypes were also rather well identified, with more than 85% of the samples correctly classified. The studied method gives a way of assessing relevant mathematical distances between digitalized signals according to qualitative knowledge of the samples.

    Notes: A computer-aided analysis based on soft-laser densitometry of electrophoretic patterns is described. The system has the potential to automatically identify wheat cultivars from normalized sodium dodecyl sulfate - polyacrylamide gel electropherograms. The principle emcompasses autocalibration of the relative mobilities of components of the unknown pattern using standards of Mr that bracket each pattern on the pictures, automatic identification of peaks and baseline correction, and simplified estimation of the peak intensities. The algorithm provides a normalized array that can be automatically compared with a library of standard cultivar patterns in order to determine those having the highest similarity percentage. The outputs of the program are (1) relative similarity plots which show the percentage of similarity between the unknown cultivar to that of each of the common standard cultivars put in a pattern library, (2) the names of cultivars in the library having the most similar patterns and (3) computer-generated electrophoretic graphics of these cultivars. This system is not intended to identify closely related cultivars but it is particularly recommended for cultivar identification without well-trained personnel, familiar with band location and nomenclature.

    Notes: An isoelectric focusing method capable of resolving all groups of storage protein of the wheat seed, including the most basic low molecular weight glutenin (LMWG), was developed. Ultrathin polyacrylamide gels were used after drying and rehydration with 8 m urea, 50 mm DTE and 2.4 % carrier ampholytes (pH 4-9). Densitometric scanning of the isoelectric focusing gels permitted a more accurate and specific quantitation of LMWG components among various cultivars than patterns based on sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The two main genetic types (i. e. ‘γ-42’ and ‘γ-45’) of durum wheats were separated on the basis of the proportion in LMWG in storage proteins, but no significant difference was found within these groups. Advantages of the system as regards reliability, high resolution ability to abolish protein oxidation and preventing reaggregation of LMWG were also discussed.
