    Notizen: The total (elastic plus inelastic) intensities of 51 keV electrons scattered by water molecules have been measured over a range of 1 ≦ K = (4π/λ) sin(θ/2) ≦ 12 Å-1. A computer program, ELIC, has been written for calculating the total intensities of electrons scattered by free molecules. The intensities can be calculated with self-consistent field and configuration interaction wavefunctions. The theoretical intensities based on a CI wavefunction are in good agreement with the observed intensities.

    Notizen: A free radical was produced enzymatically by incubating benzo(a)pyrene with liver microsomes. This radical, identified as the 6-oxybenzo(a) pyrene radical, was sufficiently reactive to bind covalently with nucleic acid bases. Similar reactive free radicals were produced enzymatically from anthanthrene and 10-aza-benzo(a)pyrene, which are carcinogenic in spite of lacking so-called “bay regions.” Hepatocarcinogens such as 3′-methyl-4-dimethylaminoazobenzene and related compounds, naphthylamines, and 2-acetylaminofluorene yield free radicals after being incubated with liver microsomes. Thus, various kinds of chemical carcinogens are found to be converted to free radicals, suggesting causal significance for the formation of free radicals in chemical carcinogenesis.

    Notizen: The total (elastic and inelastic) intensity of electrons scattered by CO2 was measured in the s range of 1 to 12 Å-1 and compared with the theoretical intensity calculated from the Hartree-Fock molecular wave function and those calculated for the independent-atom-model (IAM) molecule. In the range of s ≲ 4 Å-1 the electron correlation effect on the total scattered intensity was found to be represented by that for the IAM molecule.

    Notizen: Ionizing radiations generate in matter a large number of energetic electrons, which in turn collide with molecules in matter, produce ions and excited states, and thereby degrade in energy. The description of the consequences of many collision processes to the electrons and to matter is the goal of the electron degradation theory. We summarize the current understanding of this topic, which is important as a basis of radiation chemistry and biology. In addition, we present an initial report of our new work, namely, a generalization of the Spencer-Fano theory to time-dependent cases.

    Notizen: Adiabatic potential-energy surfaces in the lower excited states following the benzene⇌Dewar-benzene isomerization process were calculated by the INDO/S method postulating the concerted reaction mechanism which is proved to be valid in the ground state by the calculation of the intrinsic reaction coordinate. It was concluded that the benzene molecule in the 1B1u and 1E2u excited states readily isomerizes to Dewar-benzene in the condensed phase although the quantum yield may not be large. Bryce-Smith's proposal, that the isomerization to Dewar-benzene occurs only after the intersystem crossing to the 3B1u state in benzene molecule, will not be probable; for the 3B1u route is not favorable to the isomerization in comparison with these singlet routes. However, the rearomatization of Dewar-benzene in the ground state may produce the 3B1u benzene in small yield with higher yield of the ground-state benzene. The activation energy in the rearomatization is calculated to be 19.03 kcal/mol. These conclusions are consistent with the experimental facts. Molecular orbital correlations in the isomerization and the ionization potentials of Dewar-benzene were also discussed.

Significant discrepancies are found between the experimental intensities and the theoretical ones based on the SCF wave functions. Most of the chemical binding and electron correlation effects observed in the total scattered intensities are reproduced by the theoretical intensities based on the CI wave functions calculated with the basis set including polarization functions on all atoms. © 1992 John Wiley & Sons, Inc.

    Notizen: Animal cell bioreactors are often limited by the oxygen supply. The reduction in oxygen consumption per cell that occurs under hypoxic conditions may be exploited as a method for increasing reactor capacity if additional glucose is provided to offset increased glycolytic activity. The effects of oxygen deprivation on recombinant tPA (tissue-type plasminogen activator) production were investigated using midexponential and slowly growing CHO cells. The specific oxygen consumption rate can be reduced by at least 50% (mild hypoxic conditions) without affecting the cell growth rate, maximum cell concentration, tPA production rate, or tPA quality (as characterized by the tPA-specific activity and SDS-PAGE analysis). This suggests that mild-hypoxic conditions (with sufficient glucose) can be used to double the cell concentration and volumetric tPA production rate (at a constant volumetric oxygen supply rate) without sacrificing product quality. However, anoxic conditions should be avoided. When slowly growing cultures were exposed to anoxia, the tPA production rate decreased by 80% without affecting tPA quality. However, when midexponential cultures were exposed to anoxia, the drop in tPA production was accompanied by a decrease in tPA quality that ranged from a 40% decrease in tPA specific activity to extensive tPA degradation. © 1993 John Wiley & Sons, Inc.

    Notizen: Carbon dioxide is a by-product of mammalian cell metabolism that will build up in culture if it is not removed from the medium. Increased carbon dioxide levels are generally not a problem in bench-top bioreactors, but inhibitory levels can easily be reached in large-scale vessels, especially if the aeration gas is enriched in oxygen. Due to the equilibrium attained between dissolved CO2 and bicarbonate, increased pCO2 is associated with increased osmolality in bioreactors with pH control. While a few preliminary reports indicate that elevated pCO2 levels can inhibit cell growth and/or recombinant protein production, no comprehensive analysis of the interrelated effects of elevated pCO2 and osmolality has been published. We have examined the effects of 140, 195, and 250 mm Hg (187, 260, and 333 mbar, respectively) pCO2 (with and without osmolality control) on the growth of and recombinant tPA production by MT2-1-8 Chinese hamster ovary (CHO) cells. The effects of elevated osmolality were also investigated at the control pCO2 of 36 mm Hg. Elevated pCO2 at 310 mOsm/kg osmolality inhibited cell growth in a dose-dependent fashion, with a maximum decrease of 30% in the specific growth rate (μ) at 250 mm Hg. Osmolality alone had no effect on μ, but the combination of elevated pCO2 and osmolality increased the maximal reduction in μ to 45%. Elevated pCO2 at 310 mOsm/kg osmolality decreased the specific tPA production rate (qtPA) by up to 40% at 250 mm Hg. Interestingly, while increased osmolality decreased qtPA significantly at 140 mm Hg pCO2, it had no effect or even increased qtPA at 195 and 250 mm Hg. © 1996 John Wiley & Sons, Inc.

    Notizen: The accuracy and applicability of the finite-element method of the higher order interpolation functions to the one-dimensional Schrödinger equation were examined. When the fifth-order Lagrange and Hermite interpolation functions were used as the basis functions, practically exact solutions were obtained for all eigenvalues of several model potential energy functions. It was demonstrated that the appropriate analytical integration over the potential energy function within each element is important in the matrix element evaluation. The accuracy of the method was examined for the potential functions with a double minimum, which has a large classically forbidden region. The method was also applied to evaluate the Franck-Condon factors of the transitions between the 1 1Σg+ and 2 1Σu+ states of Na2; the latter state having a double minimum in its potential energy function.
