    Notes: A software system for the automated computer simulation of two-dimensional elastostatic problems by the finite element method is described. This system consists of two parts, automated mesh generation and automated stress analysis. The mesh generation is based on a method in which equilateral triangles are generated successively in the unmeshed region. Automated mesh refinement is carried out in the latter part of the simulation process. The stress analysis is based on the assumed stress hybrid method and the successive over relaxation method. The computer program developed for this paper can generate a succession of increasingly refined triangular meshes until a certain mesh convergence criterion is achieved. The mesh convergence criterion is based on a comparison of nodal stresses in successive analyses until all the stress differences are within a specified tolerance.

    Notes: By the radiation-copolymerization method with polyethylene glycoldimethacrylate (PGD) as a main polymerizable reagent, microbial cells of Brevibacterium ammoniagenes were immobilized with high specific activity of NAD kinase and high mechanical strength. The reagents used for the immobilization such as PGD, polyvinyl alcohol (PVA), and N,N′-methylenebisacrylamide (Bis) did not inversely affect the enzyme activity. Freezing and irradiation treatment of the cell-reagent solution did not inactivate the enzyme either, but longer freezing time or a lower irradiation dose (less than 400 krad) resulted in the unsatisfactory mechanical strength of the immobilized cells. Almost all of NAD and ATP consumed were converted into NADP within three hours reaction time. The drum reactor was found to be ideal for the reaction of immobilized cells, since it gave little mechanical stress to the immobilized cells for the effective mixing of the cells and the substrates. The immobilized cells were subjected to three hours reaction repeatedly for 30 times without any activity loss.

    Notes: The cell cycle dependency of rice α-amylase production in Saccharomyces cerevisiae was investigated using synchronous and arrested cultures. The results of two separate synchronous cultures, using α-mating factor and a cdc28 mutant, indicated that the rice α-amylase-specific production rate is not constant during the cell cycle. The specific production rates during G1, S, and M phases were then ascertained by inhibiting the progression of the cell cycle using α-mating factor, hydroxyurea, and nocodazole, respectively. The specific production rate was found to be maximal during the M phase. The increase in the specific production rate during the M phase was confirmed from the accumulation of M-phase cells using a cdc15 mutant. The intracellular content of rice α-amylase was also measured during the cell cycle. Like the specific production rate, the intracellular content was found to fluctuate throughout the cell cycle, and to reach a maximum during M phase. © 1997 John Wiley & Sons, Inc.     Notes: The chemiluminescence of the Cypridina luciferin analogue, 2-methyl-6-(p-methoxyphenyl)-3,7-dihydroimidazo[1,2-a]pyrazin-3-one (MCLA) was observed at 462nm in the presence of horseradish peroxidase (HRP) and the total spectrum of light emitted was found to depend linearly on HRP concentration. Methods for the determination of HRP concentration using the chemiluminescence was investigated. HRP could be detected in the range from 100 pmol/L to 100nmol/L under the optimum condition, H2O2 (10mmol/L) and MCLA (10μmol/L) at pH 5.8.

    Notes: Unsymmetrical and symmetrical calix[4]arene esters carrying oligosiloxane moiety were designed for neutral carriers of silicone-rubber-membrane sodium ion-sensitive field-effect transistors (ISFETs). The incorporation of oligosiloxane moiety to calix[4]arene ionophores promotes their dispersibility in silicone rubber, thus bringing about high stability and low impedance of the ion-sensing membranes and thereby high sensitivity and fast response of the resulting sodium ISFETs.

    Notes: Using an automated cell electrophoresis system equipped with an image processor, we studied electrophoretic mobilites of erythrocytes of healthy donors and of patients suffering from systemic lupus erythematosus (SLE), rheumatoid arthritis (RA), hypergammaglobulinemia and diabetes mellitus (DM). On average, erythrocytes from SLE patients showed mean electrophoretic mobilities (EPM) which were significantly lower (p 〈 0.005) than the EPM of red blood cells of normal donors. Evaluation of mean EPM and standard deviations revealed that three groups of SLE patients could be distinguished regarding the electrophoretic behavior of their erythrocytes. Some patients had red blood cells with normal EPM, others had erythrocytes with significantly reduced EPM, and a third group appeared to have both kinds of erythrocytes. In addition, erythrocytes of various SLE patients showed enhanced resistance to lysis by glycerol and their membranes contained less quantities of band 3 proteins.
