    Notes: Synaptic organization of the nucleus rotundus was studied with the electron microscope in three teleost species belonging to the same order. In spite of the different histological organization (non-laminated, incompletely laminated, and laminated), the same kinds of axon terminals (S and F) are observed in all species. A fibrous layer which is clearly formed only in the laminated nucleus is composed of F1 terminals and dendrites from a layer of small cells. The same kind of synapses formed between F1 terminals and dendrites of small cells are also found among glomeruli in the non-laminated and incompletely laminated nuclei. The main constituents of glomeruli are S and F2 terminals and dendrites of large cells in the non-laminated and incompletely laminated nuclei, and are S terminals and star-like structures which correspond to the tips of the dendrites of large cells in the laminated nucleus. The star-like structure contains numerous mitochondria and clusters of small polymorphic vesicles. Some of the vesicles aggregate at thickened cell membranes of the structure as in presynaptic dendrites.

    Notes: The nucleus rotundus of 21 species of teleosts was studied by a modified Bodian and the Golgi method to clarify the histological organization, with special reference to the cell lamination and the glomerular formation.The common components of the nucleus in all species are as follows: a thick fiber bundle which comes from the commissura horizontalis and enters the nucleus from the dorsal surface, many small cells, large cells, glomeruli, and a surrounding fibrous capsule. The nuclei of all species studied are classified into three types mainly on the distribution of the small cells, and to a lesser degree on the location of the large cells and the glomeruli.The first type of nucleus has small cells. large cells and glomeruli throughout its extent. In the second type of nucleus, many small cells form a peripheral cell layer, while the large cells and glomeruli are found all over the nucleus.The third type of nucleus is clearly laminated. It is composed of four layers arranged concentrically around a central fiber net in the following order: a glomerular layer, a fibrous layer, a small-cell layer, and a peripheral fibrous capsule. In some species, the large cells are located in the fibrous capsule, and all glomeruli contain a star-like structure, which corresponds to the tips of the large cell dendrites.

    Notes: Fine structure of the carp torus longitudinalis was studied by electron and light microscopy and compared with the granular layer of the carp cerebellum. There are three types of cells in the torus longitudinalis, that is, small, medium-sized and large cells. From the small cells to the medium-sized cells, there is gradual transition in size and in the amounts of cell organelles. These cell profiles and the closely packed cell pattern in a part of the torus longitudinalis are quite similar to those in the granular layer of the cerebellum. Moreover, peculiar synaptic configurations of so-called “synaptic glomeruli” are found both in the torus longitudinalis and in the granular layer of the cerebellum. In the torus longitudinalis, unmyelinated nerve fibers are seen to have bulbous swellings along their course, most of which contain a mitochondrion, as do they in the carp tectum opticum.It is speculated that the torus longitudinalis may be partly related to the granular layer of the cerebellum and partly to the tectum opticum.

    Notes: Endocrine cells and brush cells at the bronchiolo-alveolar junctions of the lung of neonatal hamsters were studied by transmission electron microscopy. On both sides of the junctions (bronchiolar and alveolar), clusters of endocrine cells occur as neuroepithelial bodies (NEB). A few solitary endocrine cells are also present at the alveolar sides of the junctions. Some endocrine cells reach from the basement membrane to the air space but the area of apical cell membrane exposed to the airway is small as the cells are largely covered by Clara cells in the bronchioles and by thin attenuations of alveolar type 1 cells in the alveoli. Some Clara cells around NEB contain cytoplasmic lamellar bodies, similar to those characteristically associated with alveolar type 2 cells. A few brush cells are also seen at both sides of the junctions. Long wide microvilli with filamentous cores extend from the apices of the brush cells. Endoplasmic reticulum and Golgi apparatus are moderately developed. Well-developed bundles of intermediate filaments course throughout the cytoplasm of some of the brush cells. The functions of endocrine cells and brush cells are unknown. However, the presence of these cells at the bronchiolo-alveolar junctions of neonatal hamster lungs suggests a role in regulation of respiratory function.

    Notes: Recombinant tumor necrosis factor (TNF), epidermal growth factor (EGF), and transforming growth factor β (TGF-β) stimulated growth of confluent human diploid fibroblasts (FS-4 cells) in the presence of fetal calf serum. TGF-β synergistically enhanced both the TNF- and EGF-stimulated cell growth, whereas synergism between the mitogenic action of EGF and that of TNF was not observed. When indomethacin or acetylsalicylic acid, an inhibitor of prostaglandin production, was added to FS-4 cells, cell growth stimulated by EGF or TNF was increased, suggesting that prostaglandins induced by these mitogens antagonize their growth stimulatory actions. In contrast, neither indomethacin nor acetylsalicylic acid had a significant effect on the TGF-β-induced growth of FS-4 cells. Mitogenic responses of indomethacin-treated cells to EGF, TNF, and TGF-β were similarly suppressed by the addition of exogenous prostaglandin D2 (PGD2). Other prostaglandins such as PGE2 and PGF2α produced less inhibition of the cell growth.

    Notes: We have examined the effects of in vitro aging on the growth capacity of human umbilical vein endothelial cells (HUVECs) under the influence of tumor necrosis factor (TNF) with or without interferon-γ (IFN-γ). The growth and colony-forming abilities of control cells were impaired with advancing age in vitro, especially at later stages (more than 70-80% of life span completed). It was found that treatment with TNF inhibited growth and colony-forming efficiency at any in vitro age. The effects of TNF were shown to increase with increasing in vitro age, as reflected by a more pronounced increase in doubling times, a decrease in saturation density, and a reduction in colony-forming efficiency. However, the characteristics of TNF receptors, including the dissociation constant, and the number of TNF-binding sites per cell-surface area remained rather constant. The effect of TNF was augmented by IFN-γ at a dose that alone affected growth and colony formation only slightly. The augmentation by IFN-γ was also found to depend on in vitro age; the synergy with TNF in the deterioration of colony-forming ability was observed only in “aged” cells. These results suggest that the intrinsic responsiveness of HUVECs to growth-inhibiting factors, as well as to growth-stimulating factors, changes during aging in vitro.

    Notes: Multiply charged ions from electrospray ionization (ESI) were observed for ruthenium-bidentate ligand complexes, such as [RuL2B]X2 and [(RuL2)2B]X4, where L is 2,2′-bipyridine, B are tetradentate ligands of 2,2′-bis(2′-pyridyl)bibenzimidazole and 2,6-bis(2′-pyridyl)benzodiimidazole, bidentate ligand of 2-(2′-pyridyl)benzimidazole and related compounds and X is CIO4- or CI-. ESI mass spectra showed a simple mass pattern for easy structural assignment and detecting impurities. The mass spectra for binuclear complexes provide a charge state distribution ranging from 4+ to 2+ for Ru(II) - Ru(II) compounds and 5+ to 2+ for Ru(II) - Rh(III) compounds. It was found that different multiply charged ions are generated by loss of counterions and by protonation/deprotonation at the proton site of ligands B. The abundances of these ions are qualitatively explained in terms of the acidity of metal complexes depending on the bridging ligand structures and the charge of the metal ions. Ions produced by removal of ligands were hardly observed.
