    Notes: Newly metamorphosed Kenyan reed frogs, Hyperolius viridiflavus ferniquei, are able to regenerate amputated digits. The terminal digital pad is also completely reformed. Differentiation of the regenerating digital pad was studied by scanning electron microscopy. External differentiation of the digital pad began late in the second postamputational week with the appearance of small patches of specialized epidermal cells on the ventral surface of the regenerating digit. The differentiation of the pad spread out radially until late in the fourth week, when its overall shape approximated that of the normal digital pad. The appearance of patches of digital pad epidermis on the ends of spike regenerates arising from the forearm was also confirmed.

    Notes: The histochemistry and ultrastructure (SEM and TEM) of the spermatheca of Biomphalaria glabrata was investigated to elucidate the function of this organ and to compare its structure and function to similar organs found in other species. The spermatheca has a debris-filled lumen surrounded by a thin wall of tissue. The cells adjacent to the lumen are of three columnar epithelial cell types. Two cell types have abundant microvilli and mammalian cell-like organelle distribution and morphology. The above cell types differ in the electron density of their cytoplasms, nuclear morphologies, and organelle content. The third cell type differs from the other two in its cytoplasmic makeup. However, the most distinctive difference is the presence of large numbers of cilia at the apical surface with no evidence of microvilli. These columnar cells rest on a basal lamina adjacent to a two to three cell thick muscle layer. The entire organ is surrounded by an adventitia of unusual morphology. Histochemical investigation demonstrated that DNAase, RNAase, and protease are present in the lumen, alkaline phosphatase is associated primarily with the microvilli, small amounts of acid phosphatase are concentrated in the midcell area of the columnar epithelium, and ATPase activity is localized in the muscle cells and just below the absorptive surface of the microvillous cells. The luminal contents and adventitial areas are Sudan Black B positive, all areas of the lumen and organ wall are PAS positive, the cell nuclei and amorphous masses in the lumen showed Feulgen staining, and large vesicles in the columnar cells were Oil Red O positive. Apparently, the spermatheca of B. glabrata is both a digestive and absorptive structure. Although this organ shares functional similarities with those found in opisthobranchs and terrestrial pulmonates, the epithelia of the spermatheca differ dramatically in these groups.

    Notes: The bursa compulatrix of the Monarch butterfly was investigated utilizing light microscopy, histochemistry, and scanning and transmission electron microscopy in order to relate its morphology to the release of sperm from the spermatophore. The bursa has a row of large chitinous teeth on either side of the organ. The dorsal and ventral surfaces are covered with chitinous plates, the plates having bristles on one side. A single layer of cells lies under both the plates and teeth, one columnar cell under each plate, one cuboidal cell under each tooth. The toothed area has no muscle cells. However, the dorsal and ventral hemispheres of the bursa each have a crescent-shaped packet of muscle fibers that traverse the organ; there are no longitudinal fibers. Spermatophores with thick walls were found in the bursal lumen. Morphological evidence suggests that the presence of the spermatophores is sensed by the bristles and that the packets are opened by contraction of the muscles bringing the large teeth into contact with the spermatophore wall.

    Notes: To investigate the association of calmodulin (CaM) with microtubules (MTs) in the mitotic apparatus (MA), the distributions of CaM and tubulin were examined in cells in which the normal spindle organization had been altered. A fluorescent CaM conjugate with tetramethylrhodamine isothiocyanate (CaM-TRITC) and a dichlorotriazinyl aminofluorescein conjugate with tubulin (tubulin-DTAF) were injected into cells that had been treated with the MT inhibitor nocodazole. With moderate nocodazole concentration (0.3 μg/ml, 37°C, 4 h) in live cells, CaM-TRITC and tubulin-DTAF concentrated identically on or near the centrosomes and kinetochores. In serial sections of these cells, small MT segments were observed by transmission electron microscopy (TEM) in the regions where fluorescent protein had concentrated. When a higher drug concentration was used (3.0 μg/ml, 37°C, 4 h), no regions of CaM-TRITC or tubulin-DTAF localization were observed, and no MTs were observed when serial sections were examined by TEM. However, following release from the high-concentration nocodazole block, CaM-TRITC colocalized with newly formed MTs at the kinetochores and centrosomes. Later in the recovery period, when chromosome-to-pole fibers had formed, CaM association with kinetochores diminished, ultimately attaining its normal pole-proximal association with kinetochore MTs in cells that progressed through mitosis. We interpret these observations as supporting the hypothesis that in the MA, CaM attains a physical association with kinetochore MTs and suggest that CaM-associated MTs may be inherently more stable.

    Notes: A new surface pretreatment for the preparation of wall-coated open tubular (WCOT) glass capillary columns has been evaluated. This technique involves the application of a non-extractable layer of Superox™-4, a 4,000,000 MW polyethylene glycol, to the glass surface as a pretreatment and deactivation agent. Unlike other polyethylene glycols, Superox-4 is stable at high temperatures (〉 300°C) in the absence of oxygen, coats smoothly onto a bare glass surface and resists droplet formation. WCOT columns (SE-54, Carbowax 20M, and SP-2250) prepared using this technique are compared to columns prepared using a modified Grob [2,3] BaCO3 procedure. The Superox-4 pretreated columns were equal or superior in quality to the BaCO3 pretreated columns, based on the appearance of an activity standard and the effective theoretical plates (Neff) per meter. Chromatograms showing practical application of the WCOT columns prepared using both methods is given.

    Notes: Nine organosulfur compounds present in an aged garlic extract and two isoflavonoids and one triterpenoid present in a licorice root extract powder have been identified and quantified. Quantification involved solvent extraction and gas chromatographic - mass spectrometric analysis (garlic extract) or hydrolysis, solvent extraction, and liquid chromatographic analysis (licorice root extract powder).Although the garlic extract proved to be unstable and the concentration of the organosulfur compounds varied with time, one analysis of the extract gave the following results: methyl disulfide (0.607 μg/g), methyl trisulfide (0.181 μg/g), allyl sulfide (2.02 μg/g), allyl disulfide (0.784 μg/g), allyl trisulfide (0.795 μg/g), allyl methyl sulfide (1.64 μg/g), allyl methyl disulfide (0.411 μg/g), allyl methyl trisulfide (0.695 μg/g), and ethyl 2-propenesulfinate (11.4 μg/g). The analysis of the licorice root extract powder gave the following results: formononetin (1.92 mg/g), isoliquiritigenin (9.61 mg/g), and 18β-glycyrrhetinic acid (43.9 mg/g).Methods were successfully developed to quantify the same compounds in the serum of test animals which had consumed feed spiked with garlic extract or licorice root extract powder. Only the 18β-glycyrrhetinic acid could be detected in the sera of such animals, however. An effort was also made to determine serum levels of prostaglandin E2 to correlate its inhibition with levels of the dietary components, but the prostaglandin E2 levels were too low to measure.

    Notes: The origin of the internal carotid artery in the lizards Tiliqua occipitalis and Trachysaurus rugosus has been shown to be both extremely variable and complex. In the dense connective tissue investing the junction of the internal carotid artery and the summit of the carotid arch are groups of cells associated with sinusoids derved from arterioles arising from the neighboring great vessels. The conspicuous epithelioid cells in these groups resemble the mammalian carotid body glomus and amphibian α cells. Nerve fibers showing no obvious synaptic specializations pass to and over these cell groups. It is suggested that these cell groups and sinusoids are comparable to mammalian carotid body glomeruli. A similar group of cells has been described in the concavity of the summit of the fourth aortic arch. This cell group, however, gives a distinct chromaffin reaction.

The sequence of morphological events is very similar to that seen in the typical epimorphic regeneration of amphibian extremities. Amputated forearms typically produce hypomorphic spike regenerates. Despite the lack of underlying hand and digital structures, an apical “digital” pad commonly differentiates.

    Notes: The ultrastructure of the spermatheca of the reproductive tract in the pulmonate snail, Sonorella santaritana, was investigated. This organ has a debris-filled lumen and an outer wall which can be divided into three distinct layers. The cell layer adjacent to the lumen is comprised of two cell types, tall columnar epithelial cells with microvilli and cells lacking microvilli. The next layer also has two cell types, muscle cells and apparent pigment cells. The most distant layer is an adventitia of large glycogen-containing cells. The lumen of the spermatheca contains a core of partially digested sperm and related materials.The luminal contents and the cellular morphology of this organ suggest that the spermathecal functions are both digestive and absorptive. It is proposed that excess sperm and related materials are transported to the spermatheca, digested, and the usable products are reabsorbed.

    Notes: General cytological and ultrastructural features of cells found in the peripheral blood of three captive beluga whales (Delphinapterus leucas) are described. We noted all major peripheral cell types common to the circulation of mammalian species, including polymorphonuclear neutrophils, eosinophils, basophils, lymphocytes, and monocytes, as well as myelocytes, erythroblasts, and plasma cells. Platelets and two populations (normal and hypochromic) of red blood corpuscles were observed.
