Notes: The on-line coupled LC-GC technique was applied to the analysis of several metal chelates of N,N-diethyldithiocarbamic acid. A 10-port valve interface was used to couple the LC and GC instruments. Conditions during sample transfer into the GC gave fully concurrent solvent evaporation. The chelates investigated were separated with a short apolar fused silica column. LC preseparation was made with cyano or amino phases using a hexane/dichloromethane mixture as eluent. On-line LC-GC combination seems to be very suitable for the separation of the metal chelates studied.

Notes: Rotational barriers and 15N chemical shifts have been measured in a number of ureas and thioureas. As anticipated on the basis of the 15N shifts, several previously unobserved rotational barriers could be detected by using lanthanide reagents or a high field spectrometer. Nearly constant effects on both the rotational activation energy and the 15N shift are produced on going from ureas to the corresponding thioureas, and correlations are found between the ΔG

Notes: The kinetics of the depolymerization of native dextran in dilute hydrochloric acid at different temperatures has been followed by measurements of M̄w, M̄n, [η], 〈ρ2〉1/2, and S. The data allow the correlation of the various parameters of the total hydrolyzate at various degrees of depolymerization. At a first approximation the results conform to a random splitting of a statistical, branched polymer.

Notes: Chemical shifts of base and H1′ protons of the single-stranded hexamers d(ATTACC) and d(GGTAAT), of the 1 : 1 mixtures of these complementary hexamers, and of the self-complementary dodecamer d(ATTACCGGTAAT) were measured at various temperatures in aqueous solution. Four different sample concentrations were used in the case of the dodecamer and of the mixture of the complementary hexamers; the individual hexamers were measured at two different DNA concentrations. Absorbance temperature profiles at five different NaCl concentrations were measured for the dodecamer in order to quantify the effect of the ionic strength on the duplex formation.Under suitable conditions of nucleotide concentration, temperature, and ionic strength, the dodecamer adopts either a B-DNA duplex or a hairpin-loop structure. Chemical shift vs temperature profiles, constructed for all samples, were used to obtain thermodynamic parameters either for the various stacking interactions in the single strands or for the duplex or the hairpin-loop formation. In the analysis of the duplex formation of the hexamers, a two-state approach appeared too simple, because systematic deviations were revealed. Therefore, a new three-state model (DUPSTAK) was developed. In order to investigate the magnitude of error arising from the use of the two-state approach in cases where the DUPSTAK model appears more appropriate, a series of test calculations was made. The magnitude of error in the enthalpy and in the entropy of duplex melting is found to depend linearly upon the actual melting temperature and not upon the individual ΔHOd and ΔSOd values.Thermodynamic analysis of the chemical shift vs temperature profiles in D2O solution (no added salt) yields an average Tmd value of 341 K (1M DNA) and ΔHOd of -121 kJ · mol-1 for the dimer/random-coil transition of the hexamer duplex d(ATTACC) · d(GGTAAT). For the duplex ⇄ random-coil transition of the 12-mer d(ATTACCGGTAAT) an average Tmd value of 336 K (1M DNA) and ΔHOd of -372 kJ · mol-1 are found. The hairpin/random-coil transition of d(ATTACCGGTAAT) is characterized by a rather large ΔHOh value, -130 kJ · mol-1, and an average Tmh value of 304 K.

Notes: To optimize antigen-antibody reactions, we have synthesized chemically well-defined hy-drophilic microspheres. Proteins or haptens were covalently linked to these carriers. When the microsphere conjugates were agglutinated by the corresponding antiserum, the size of the complex artificially increased during the immunological reaction. After optimizing var-ious parameters such as the hydrophilic character, repulsion charges, and amount of antigen coupled to the microspheres, we developed a rapid and sensitive immunoassay based on laser light scattering by the complexes.

Notes: The uv absorption of Na-hyaluronate (NaHA) films and the refractive indices, water content, and swelling of LiHA films have been measured as a function of relative humidity. Three peaks are observed in the uv absorption of NaHA (at about 250, 310, and 330 nm) for water content above 10 water molecules per disaccharide. The absorptivity of the 250, 310, and 330 nm peaks increase as the water content increases, indicating a change in the electronic properties of the HA molecule. The refractive indices, water content, and swelling of LiHA films are used to determine the optical polarizability via the Lorentz-Lorenz relation. The polarizability of LiHA is found to have a similar dependence on water content as NaHA, though the changes observed are larger in magnitude. © 1994 John Wiley & Sons, Inc.

Notes: Twelve well-characterized fractions of polypropylene and a set of polystyrene standard samples are used to establish the universal calibration in gel permeation chromatography for polypropylene in 1,2-dichlorobenzene at 135°C. The Mark-Houwink relation under those conditions and the unperturbed dimensions are determined for polypropylene. Comparisons are made with earlier work with less well characterized samples.

Notes: The influence of the degree of polymerization of oligomers (1 ≤ l ≤ 50) on the reactivity of the OH endgroups is exemplified by the reaction of oligoterephthalates of phenolphthalein with benzoyl chloride under ϑ-conditions. It is found that with increasing chain length l the hydroxyl group activity increases according to an S-form law. It is suggested that the ester groups of the macromolecule influence the activity of the OH endgroups due to solvation effects. This suggestion is confirmed theoretically and experimentally.

Notes: A series of well-characterized polyamic acids from pyromellitic dianhydride and 4,4′-diaminodiphenyl ether of systematically varied molecular weight and known molecular weight distribution were chemically converted to polyimide films under regulated conditions. The films were essentially noncrystalline, of moderate orientation, and soluble in concentrated sulfuric acid. The mechanical properties of the films varied sigmoidally with the solution properties of the precursor. Molecular weights were derived for the onset of mechanical strength and limiting property values are given. The relations between film properties and molecular structural parameters suggest that cyclization probably occurs here with little contribution from crosslinking, degradation, or other side reactions which would appreciably alter molecular weight and molecular weight distribution.
