    Description: We aimed to elucidate the effects of hepatoma-derived growth factor ( HDGF ) on growth and metastasis of hepatocellular carcinoma (HCC) cells. Tissue microarrays with 236 HCC specimens and 18 extrahepatic metastases were utilized to detect the HDGF expression by immunohistochemistry. Meanwhile, HDGF expressions in HCC cell lines with different metastatic potentials were examined using immunofluorescence staining, real-time PCR and western blotting. After HDGF silencing, the growth and metastatic potentials of HCC cells were evaluated by soft agar assay, invasion assay, together with tumorigenicity assay in nude mice. The gelatin zymography was performed by detecting MMP-2 and MMP-9 levels. Additionally, western blotting was conducted to determine the levels of total and phosphorylated ERK1/2, JNK, p38 and Akt. The results showed that HDGF was overexpressed in HCC metastasis tumour, and the expression increased with the differentiation degree of tumours (Grade I 44.0%, Grade II 48.4% and Grade III 65.6%). Consistently, HDGF levels were positively associated with the metastatic capability of HCC cells (MHCC97L 〈 MHCC97H 〈 HCCLM3). The growth and metastasis were suppressed by HDGF-siRNA. Gelatinolytic activities were enhanced in the three metastatic HCC cell lines, but had no significant difference among them. The tumourigenicity and metastatic capability of HCCLM3 cells in nude mice were inhibited after silencing HDGF . Meanwhile, HDGF-siRNA specifically suppressed the total and phosphorylated protein levels of ERK1/2, while not JNK, p38 and Akt. In conclusion, HDGF was overexpressed in HCC patients and cells, and HDGF might be closely correlated with HCC metastasis via regulating ERK signalling pathway.     Description: Two new steroidal alkaloids, 23-methoxycyclopamine 3- O - β- D -glucopyranoside ( 1 ) and isoecliptalbine ( 2 ), were isolated from the root and rhizoma of Veratrum maackii Regel , together with five known compounds, i.e. , verussurine ( 3 ), verabenzoamine ( 4 ), verazine ( 5 ), isoverazine ( 6 ), and verazinine ( 7 ). Their structures were established by extensive analysis of spectroscopic data, as well as by comparison with literature data. Compounds 1 – 7 could cause DNA damage in the cerebellum and cerebral cortex of mice in a dose-dependent manner by using single-cell gel electrophoresis (comet assay).

    Description: Phospholipid hydroperoxide glutathione peroxidases (PHGPXs) are essential enzymes of the cellular antioxidant defense system during insect–plant interactions. However, little attention has been devoted to the functional characterization of PHGXPs in the whitefly Bemisia tabaci . Here, we report the identification and characterization of two PHGPX genes, designated as BtQ-PHGPX1 and BtQ-PHGPX2 from the Mediterranean species of the B. tabaci complex. Sequence analysis indicated that the length of BtQ-PHGPX1 is of 942 bp with a 729 bp open-reading frame (ORF) encoding 242 amino acids, and BtQ-PHGPX2 is of 699 bp with a 567 bp ORF encoding 188 amino acids. Sequence alignment analysis showed that BtQ-PHGPX1 and BtQ-PHGPX2 shared high similarity with other known PHGPXs. The NVASX C GXT, FPCN Q FXXQEPG, and IK W NFXKFLV surrounded the reactive cysteine, glutamine, and tryptophan residues, respectively. Recombinant BtQ-PHGPX1 and BtQ-PHGPX2 were overexpressed in Escherichia coli and purified. quantitative reverse transcription-polymerase chain reaction (qRT-PCR) analysis with whiteflies of different development stages showed that the mRNA levels of BtQ-PHGPX2 were significantly higher in larvae than in other stages. The mRNA levels of BtQ-PHGPX2 were significantly higher than BtQ-PHGPX1 during all the developmental stages. The mRNA levels of BtQ-PHGPX1 and BtQ-PHGPX2 in female adults were relatively higher than in male adults. The expression of BtQ-PHGPX1 and BtQ-PHGPX2 was induced by the insecticide imidacloprid. These results suggest that BtQ-PHGPX1 and BtQ-PHGPX2 may participate in detoxification of oxidative hazards in B. tabaci .

    Description: Two unusual rearranged flavan derivatives with a rare bicyclo[3.3.1]non-3-ene-2,9-dione ring, tazettone A ( 1 ) and tazettone B ( 2 ), together with five known flavans, 3 – 7 , were isolated from the bulbs of Narcissus tazetta var. chinensis Roem . The structures of two new compounds were elucidated by spectroscopic analyses, including 1D- and 2D-NMR spectroscopy. All of the isolated compounds were evaluated for their cytotoxicities against four human tumor cell lines A549, HCT116, SK-BR-3, and HepG2. Compounds 1 and 2 were almost inactive against all tested cell lines, while compounds 3 – 7 exhibited moderate or weak cytotoxicities against the tested cell lines.

    Description: The CuAlO 2 films were prepared on glass and quartz substrates by radio frequency magnetron sputtering with a sintered target composed of Al (OH) 3 and Cu 2 O powders. X-ray diffraction analyses reveal that the as-deposited films are amorphous structure and the as-annealed films are predominated by the CuAlO 2 phase along with the Al 2 O 3 phase. Scanning electron microscope (SEM) measurements indicate that grains grow up and surface becomes rougher as a result of increasing temperature. The optical studies show that annealing leads to the reduction of the average transmittance of the films from 60% to 40% for the visible range and the decrease of the fitted refractive index from 2.0 to 1.5. Direct band gaps confirmed by the transmittance and reflectance measurements are estimated to be 3.86 and 3.50 eV for as-deposited and as-annealed films, respectively. As increasing exposure time, water contact angles of the as-deposited films exposed under ultraviolet light source 365 nm or 254 nm decrease from 96.3° and 96.4° to 87.5° and 87.2°, respectively. The variation of contact angle can be attributed to the hydrophobic organic stains on the surface, surface roughness, the formation of Cu 2+ , and band gap variation of CuAlO 2 film. The as-annealed film becomes hydrophilic and its water contact angle is 31.1°. The wetting property of CuAlO 2 films is very important for the wide spread application of this material.

    Description: A systematic phytochemical investigation on Abies forrestii afforded two new and 20 known compounds. Abieseconordines A and B ( 1 and 2 ) are the first two examples of norditerpenes with a novel 18-nor-5,10 : 9,10-disecoabietane skeleton. Their structures were established mainly by analysis of 1D- and 2D-NMR spectroscopic data. In addition, electronic circular-dichroism calculations and molecular-orbital analysis were utilized to confirm the absolute configuration of 1 . Both compounds exhibited a potent effect in a bioassay inhibiting LPS-stimulated NO production in RAW264.7 macrophages.

    Description: Over 1400 electron probe and 700 ion probe microanalyses were performed on eleven mineral separates to evaluate their potential as reference materials for in situ Li isotopic determination. Our results suggest the homogenous distributions of major elements, Li and its isotopes for each sample. Hence these samples are suitable to be used as reference materials for in situ measurements of Li abundance and Li isotopes by secondary ion mass spectrometry (SIMS) or laser ablation-inductively coupled plasma-mass spectrometry (LA-ICP-MS). These samples have the advantage of mitigating probable matrix effects during calibration owing to the wide range of compositions. The effect of composition on the δ 7 Li of olivine measured by SIMS is a linear function of composition, with δ 7 Li increasing by 1.0‰ for each mole percent decrease in forsterite component.     Description: A composite time series of the merged satellite altimeters sea surface height anomaly (SSHA) data and satellite-observed sea surface temperature (SST) data were used to identify eddies in the Western North Pacific Ocean (WNPO), where there were numbers of intense typhoons. This study systematically investigated 15 super typhoons during the period of 2000-2008 in the WNPO to study their impacts on the pre-typhoon ocean features, e.g., the cyclonic ocean eddy (COE) feature (closed contours of SSHA 〈 -6 cm) and neutral condition (SSHA between -6 and 6 cm). Two new COEs are generated by two super typhoons, and 18 pre-existing COEs are intensified by 13 super typhoons. 5 of the 13 super typhoons each influenced two pre-exisiting COEs. Although the typhoon-induced maximum cooling centers had a right bias along the tracks due to wind conditions, pre-existing COEs also play a significant role in determining the strength and location of large SST cooling. Three possible factors (maximum wind speed, typhoon translation speed and the typhoon forcing time, T f ) are employed to explain the interactions. Above all, the changes of the COE geometric and physical parameters (e.g., effective radius, area, SST, SSHA, and eddy kinetic energy) were mostly related to the typhoon forcing time, T f . This is because T f is a parameter that is a combination of the typhoon’s translation speed, intensity and size. Although the typhoons may significantly impact COEs, such samples were not commonly observed. Thus, the impact of typhoon on the strength of COEs is generally inefficient.

    Description: Recently, PSEN1 has been reported to have mutations in dilated cardiomyopathy pedigrees. However, the function and mechanism of PSEN1 in cardiomyopathy remains unresolved. Here, we established 4 types of genetically modified mice to determine the function of PSEN1 in cardiac development and pathology. PSEN1 null mutation resulted in perinatal death, retardation of heart growth, ventricular dilatation, septum defects, and valvular thickening. PSEN1 knockout in adults led to decreased muscle fibers, widened sarcomere Z lines and reduced lengths of sarcomeres in cardiomyocytes. Cardiovascular loss of function of PSEN1 induced by Sm22a-Cre or Myh6-Cre/ER /tamoxifen also resulted in severe ultrastructural abnormalities, such as relaxed gap junctions between neighboring cardiomyocytes. Functionally, cardiovascular deletion of PSEN1 caused spontaneous mortality from birth to adulthood and led to diastolic heart dysfunction, including decreased volume of the left ventricle at the end-systolic and end-diastolic stages. Additionally, in a myocardial ischemia model, deletion of PSEN1 in the cardiovascular system first protected mice by inducing adaptive hypertrophy but ultimately resulted in severe heart failure. Furthermore, a collection of genes was abnormally expressed in the hearts of cardiac-specific PSEN1 knockout mice. They were enriched in cell proliferation, calcium regulation, and so on. Taken together, dynamic regulation and abnormal function of PSEN1 underlie the pathogenesis of cardiovascular diseases due to ultrastructural abnormality of cardiomyocytes.
