    Notes: Laser diffraction spectrometry (LDS) is often claimed to operate on the principle of Fraunhofer diffraction. This is only true, however, if particles are large compared to the wavelength of light or if the ratio of the refractive indices of the disperse and continuous phases, m, is clearly different from unity. In this study it has been established that LDS, as applied to particle and droplet sizing in suspensions and emulsions, is based on Miescattering. Scattering patterns of single particles may be calculated if the refractive indices of both phases are known. Thus, a theoretical basis has been provided for the application of LDS to size-measurement in suspensions and emulsions, and for extension of this method to the lower size ranges and those cases in which the refractive indices of the disperse and continuous phases are similar. Extension of the work presented in this paper will enable the calculation of scattering matrices so that calibration of the apparatus with standard materials may be avoided.

    Notes: The practical performances of single and multi-angle photon correlation spectroscopy (PCS) for resolving bimodal distributions of industrial poly (methyl methacrylate) samples was investigated in a comparative study by several users affiliated to academic and industrial groups and by two suppliers of commercial equipment. The results for the harmonic intensity averaged diameters obtained by cumulants analysis reported by the different laboratories are in agreement, in addition to the results for the normalized second cumulants. The uncertainty on the latter quantities is large, however. For the bimodal samples with two populations with average diameters in a ratio of about 2.5 : 1, not all users were able to resolve the distribution in its components by single-angle PCS. Some slight improvement was obtained by multi-angle PCS. Other indirect techniques (polarization intensity differential scattering, static light scattering data and disc photosedimentometer) appeared to be superior for resolving the bimodal distributions.

    Notes: The objective of a continuing study by the Belgian Particle Technology Group involves difficulties in practical size determinations. This part describes the behaviour of silicas with different porosities during sizing by electrical sensing zone and laser diffraction methods. Anisotropy and porosity were identified to be important particle characteristics in understanding the differences between the two methods. Especially large pore diameters and pore volumes were found to be responsible for shifts in size distribution of 50 to 100%. The use of optical values and optical models was shown to influence these shifts considerably. In the case of spherical silica particles with moderate porosity, no significant differences could be found between the two sizing methods.

    Notes: Chromosomal DNAs from various yeast species were separated by orthogonal-field-alternation gel electrophoresis (OFAGE). To this end we developed a spheroplasting and lysis method to obtain intact DNA from both ascomycetous and basidiomycetous yeasts. The OFAGE banding patterns of 22 ascomycetous and four basidiomycetous yeast strains were compared. The strains represented species from the genera: Brettanomyces, Candida, Croococcus, Filobasidiella, Geotrichum, Hansenula, Kluyveromyces, Pachysolen, Pichia, Rhodosporidium, Rhodotorula, Saccharomyces, Saccharomycodes, Saccharomycopis, Schizosaccharomyces and Zygosaccharomyces. Variations occurred in the number of bands and their positions in the gel, not only among strains of different genera but also among species from the same genus and even between varieties of the same species. The ascomycetous yeasts, with the exeption of Saccharomyces cerevisiae, only showed one to five bands of DNA larger than 1000 kilobase pairs (kb) in general none smaller. The paterns of the four basidiomycetous yeasts revealed also a few large DNA bands but in addition one to six bands ranging in size from 500 to 1000 kb, with the exception of a single smaller chromosome in Rhodotorula mucilaginosa. From the OFAGE banding patterns of strains studied here it appears that in Saach. cerevisiae the partitioning of DNA over chromosomes in unique. But rather than the large number of chromosomes, the presence of four chromosomes with less than 500 kb of DNA is characteristic for Sacch. cerevisiae.

    Notes: In the yeast Saccharomyces cerevisiae a novel control exerted by TPS1 (=GGS1=FDP1=BYP1=CIF1=GLC6=TSS1)-encoded trehalose-6-phosphate synthase, is essential for restriction of glucose influx into glycolysis apparently by inhibiting hexokinase activity in vivo. We show that up to 50-fold overexpression of hexokinase does not noticeably affect growth on glucose or fructose in wild-type cells. However, it causes higher levels of glucose-6-phosphate, fructose-6-phosphate and also faster accumulation of fructose-1,6-bisphosphate during the initiation of fermentation. The levels of ATP and Pi correlated inversely with the higher sugar phosphate levels. In the first minutes after glucose addition, the metabolite pattern observed was intermediate between those of the tps1Δ mutant and the wild-type strain. Apparently, during the start-up of fermentation hexokinase is more rate-limiting in the first section of glycolysis than phosphofructokinase. We have developed a method to measure the free intracellular glucose level which is based on the simultaneous addition of d-glucose and an equal concentration of radiolabelled l-glucose. Since the latter is not transported, the free intracellular glucose level can be calculated as the difference between the total d-glucose measured (intracellular+periplasmic/extracellular) and the total l-glucose measured (periplasmic/extracellular). The intracellular glucose level rose in 5 min after addition of 100 mm-glucose to 0·5-2 mm in the wild-type strain, ±10 mm in a hxk1Δ hxk2Δ glk1Δ and 2-3 mm in a tps1Δ strain. In the strains overexpressing hexokinase PII the level of free intracellular glucose was not reduced. Overexpression of hexokinase PII never produced a strong effect on the rate of ethanol production and glucose consumption. Our results show that overexpression of hexokinase does not cause the same phenotype as deletion of Tps1. However, it mimics it transiently during the initiation of fermentation. Afterwards, the Tps1-dependent control system is apparently able to restrict properly up to 50-fold higher hexokinase activity. © 1998 John Wiley & Sons, Ltd.

    Notes: At the nonpermissive temperature, somatic embryos of the temperature-sensitive (ts) carrot (Daucus carota L.) cell variant ts11 only proceed beyond the globular embryo stage in the presence of medium conditioned by wild-type cells. The causative component in the conditioned medium has been identified as an acidic 32 kD endochitinase. An antiserum raised against the 32 kD chitinase detected this protein in culture medium from ts11 embryo cultures grown at the permissive temperature as well as at the nonpermissive temperature. No difference in biochemical characteristics or in effect on ts11 embryo development could be detected between the 32 kD chitinase purified from wild-type cultures and the chitinase from ts11 cultures grown at the permissive or at the nonpermissive temperature. Compared to the amount present in a ts11 embryo culture at the permissive temperature, a reduction in the amount of 32 kD chitinase was observed during the temperature-sensitive period at the nonpermissive temperature. These results imply that the arrested embryo phenotype of ts11 is not the result of a structural difference in its 32 kD chitinase, but is the result of a transient decrease in the amount of 32 kD chitinase present. Morphological observations indicate that the ts11 phenotype is pleiotropic and also affects the cell wall of nonembryogenic cells. © 1995 Wiley-Liss, Inc.

    Notes: Experimental data are reported for ion exchange equilibria of binary systems on Amberlite IR-120 resin, at different temperatures and total ionic concentrations in aqueous solutions. These systems exhibit non-ideal behaviour in both phases, and the equilibrium characterization has been based on the law of mass action, using Wilson and Meisssner, and Kusik equations to correlate the activity coefficients in the resin and aqueous solutions, respectively. Equilibrium constants, standard thermodynamic properties (Gibbs free enthalpy changes, enthalpy and entropy) and Wilson binary parameters were obtained as functions of temperature.

    Notes: Computer-aided design of a packed column to remove ethanol from aqueous solution with dibutyl butylphosphonate (DBBP) was accomplished with a programme based on the Pratt method and on the slip velocity equation. Reliable data for successful computation were obtained from literature correlations (drop size, hold-up and axial mixing coefficients for different types of packing) and from experimental work (equilibria and kinetics). Equilibrium studies on the H2O-ethanol-dibutyl butylphosphonate system were performed at different temperatures. A decrease in temperature increased the selectivity of the extractant while the distribution coefficient of ethanol, KD ≈ 1.0, remained constant. The behaviour of the system kinetics towards temperature is assessed in order to determine the initial mass transfer flux (892 × 10-3 kg m-2s-1, 30°C) and the activation energy, 22.9 kJ/kmol. The process is governed by the diffusion of the species involved. The influence of the type of packing of the column on its design is analyzed from the inventory volume of the solvent, the total volume of the column and the axial dispersion coefficients under normalised inlet and outlet conditions.

    Notes: The possibilities and limitations of photon correlation spectroscopy (PCS) as a practical tool for particle sizing are reviewed. In this part the most frequently used methods of data analysis are briefly surveyed. The practical performances for the determination of particle size distributions were evaluated by a comparative study with different kinds of users (industrial and academic research groups and manufacturers). The PCS results were also compared with electron microscopy (EM) and static light scattering (SLS) results. Thereby it was confirmed that the amount of reliable information that can be obtained by PCS about particle size distributions with measuring times of the order of minutes is limited by the ill-conditioning of the data inversion procedure, among other factors. Nevertheless, the information obtained from the PCS measurements agrees essentially with the EM and SLS results.

    Notes: The application of the singular value analysis and reconstruction method (SVR) and of the Contin method for the collective analysis of multi-angle noisy photon correlation data sets was investigated. Provided the data are sampled equidistant in time and by proper tuning of the sampling time to the scattering angle, a collective multi-angle SVR analysis is feasible. For homogeneous spherical particles such an analysis does not require any prior knowledge of the angular dependence of particle scattering power (e.g. Mie scattering). SVR allows the information content to be separated from noisy intensity autocorrelation data. It is illustrated that the multi-angle SVR analysis enhances the recovery of the information content. Moreover, SVR can be used as a fast and accurate preprocessor for extracting the field autocorrelation function for a subsequent Contin analysis whereby prior knowledge of particle scattering power as a function of scattering angle is used as a constraint. Compared with the data analysis of multi-angle time-averaged scattered intensity measurements, the information that can be extracted from multi-angle PCS data in comparable accumulation times is poorer.
