    Notes: Summary In order to determine how informative homologous donor DNA might be made available to propigment cells of the recipientXiphophorus helleri for transformation, labelled heterologous DNA fromE. coli was injected into the neural crest region or the yolk sac of embryos of the recipient. On the basis of the degradation rate of the donor DNA and the incorporation rate of radioactivity into the recipient DNA, it is concluded that injection into the neural crest region may be a suitable method to make available informative homologous donor DNA for transformation.

    Notes: Abstract. We report on the observation of a multiple-pattern stability region in a photorefractive single-feedback system. Whereas hexagonal patterns are predominant for feedback with positive diffraction length we show that a variety of stable non-hexagonal patterns are generated for certain negative diffraction lengths. For the same values of the control parameters square, rectangular, or squeezed hexagonal patterns are found alternating in time. Besides these pure states, we found a number of different mixed-pattern states. We review the linear stability analysis for this system and show that the special shape of the threshold curves in the investigated parameter region gives a first explanation for the occurrence of a multiple-pattern region.

    Notes: Zusammenfassung Elektrophoretische Untersuchungen an benignen und malignen Rückenflossenmelanomen sowie an normalen Rückenflossen lebendgebärender Zahnkarpfen zeigen, dass in den Geweben in erster Linie die für gut mit Sauerstoff versorgtes Gewebe typische Herz-LDH (B4-Isoenzym) vorkommt. Die für schlecht mit Sauerstoff versorgtes Gewebe typische Muskel-LDH (A4-Isoenzym) wird nur in manchen der untersuchten Gewebe gefunden, und ist auch dort nur in sehr geringer Menge vorhanden. Die Melanome scheinen also im Gegensatz zu vielen anderen Neoplasmen anderer Systeme aeroben Stoffwechsel durchzuführen.

    Notes: Abstract Tissue-specific esterases of the xiphophorine fishes Platypoecilus maculatus (platyfish), Xiphophorus helleri (swordtail), and their F1 hybrid have been analyzed using disc electrophoresis. Seven esterase zones (resolved into a maximum of nine bands) exist in these fishes, and these have been classified by employing specific inhibitors. Five of the seven zones, EST-1, EST-2, EST-5, EST-6, and EST-7, appeared to be carboxylesterases; while the two remaining zones, EST-3 and EST-4, were classified as cholinesterases. In the liver of the platyfish, all seven esterase zones were detected, while the liver of the swordtail exhibited only five esterase zones. EST-1 and EST-3 were lacking in the liver tissue of the swordtail. All seven esterase loci were expressed in the liver tissue of the F1 hybrid. The reciprocal crosses gave the same results. In the fin, skin, skeletal muscle, and eye tissues from all three genotypes, three major esterase zones, EST-2, EST-5, and EST-7, were detected. In addition, EST-1 was frequently detected in all these tissues of the platyfish and the F1, but was lacking in the swordtail. Serum from three genotypes showed one prominent esterase zone, EST-5; however, trace activity of EST-2 and EST-7 zones could also be detected. It seems that in all tissues of the F1 hybrid there is expression of all the esterase genes from the platyfish. The results of the present study are discussed in comparison to those from other studies on teleost esterases.

    Notes: Summary Nerve growth factor (NGF) was localized in the submandibular gland of adult male mice by a direct immunocytochemical method using highly purified antibodies against NGF coupled to horseradish peroxidase. In light microscopic sections the reaction product was entirely confined to the cells of the secretory tubules. The acinar part of the gland was free of reaction product. This finding was confirmed by electron microscopy. Within the cells NGF was localized exclusively in the apical secretory granules. No reaction was observed in the rough endoplasmic reticulum, the Golgi region or in the granules of the basal part of the cells. This observation favours the assumption that NGF is derived from a precursor molecule and that the precursor is transformed into immunologically active NGF within the secretory granules during their transport from the basal to the apical part of the tubular cells. Stimulation of the submandibular gland with carbachol (2 mg/kg) led to a massive release of the content of the secretory granules, including NGF, into the salivary duct.

    Notes: Summary The particular suitability of the Xiphophorine fish system for achieving genetic transformation is presented, and it was analyzed whether information carrying donor DNA might be available to the propigment cells of embryos ofXiphophorus helleri, which are the target cells for the transformation. Heterologous2H3H-labelled donor DNA fromE. coli, which was taken for technical reasons instead of homologous fish DNA, undergoes degradation both after injection into the neural crest region and after injection into the yolk sac (molecular weight at 0 h: 50×106; at 2 h: 1×106; at 5 h: 〈3×105; at 10 h: 〈1×104). It is concluded therefore, that informative donor DNA is present for about 2 to 3 hours after injection. The DNA of the recipient embryo is labelled radioactively during that time at which informative DNA is present only, if the donor DNA is injected into the neural crest region. The probability that a foreign gene might become available to the propigment cells and might induce transformation is discussed.

    Notes: Abstract  The structure of a series of three molecular weights of diblock copolymers polystyrene/b-isoprene with 11% volume fraction of polystyrene and low polydispersity has been investigated using small angle X-ray scattering (SAXS). In between the disordered and the BCC ordered state a micellar state with liquid-like order was found. The transition between these states was investigated in a temperature-driven experiment. Whereas the micelles appear gradually with lowering temperature the formation of the BCC ordered state occurs discontinuously at a well-defined temperature. Detailed analysis of the scattering profiles provides access to the micellar size and distance in the liquid-like as well as in the BCC state. The kinetics of the ordering transition was studied using time-resolved SAXS after temperature jumps from the liquid-like to the BCC state. The growth of the micelles and their ordering on the periodic lattice were found to occur on clearly separated time scales.
