    Notes: Abstract This paper reports on a joint meeting of the Royal Astronomical Society's Joint Association for Geophysics and VERTIC (the Verification Technology Information Centre) held in London in 1992. The topics presented focused on the detection and recognition of underground nuclear explosions. The objective of the meeting was to emphasize the multi-methodological approach that is important in verifying compliance with test-ban treaties. An overview of seismological monitoring was followed by a discussion of the technical and scientific aspects of a global seismic monitoring network, and in particular of the 1991 experiment to test the large-scale international exchange of seismic data between recording stations and data centres world-wide. The current capabilities of satellite remote-sensing were presented, and their use explained in terms of both the provision of information for monitoring the development of foreign nuclear testing programmes and also for providing sufficient information for the evaluation of treaty compliance. A review of radio-isotope sampling showed how the isotopic signature of both air and ground based sampling programmes can be diagnostic of the nuclear source. Finally, previously classified research on the ionospheric effects of underground nuclear explosions was presented, the generated acoustic waves disturbing the ionosphere and producing detectable changes in the reflection of radio and radar signals which have potential as a monitoring technique.

    Notes: Summary Alcian blue combines with purified sulphatide in 1.OM magnesium chloride. In tissue sections from patients with metachromatic leukodystrophy, sulphatide is stained by Alcian blue in 0.8 M magnesium chloride, and the staining can be abolished by prior treatment with chloroform and methanol. The simplicity of the technique, its specificity and ease of interpretation recommend Alcian blue staining at high salt concentrations as a routine method in the diagnosis of metachromatic leukodystrophy.

    Notes: Summary A new buffer system is described for use in histochemical staining with basic dyes. The buffer is made up by mixing solutions of formic acid and sodium acetate. Tables giving the proportions for closely-spaced pH values in the range 3.0–5.6 are presented. A table for an acid phosphate series down to a pH of 2.0 is also included. The value of these buffer mixtures in histological as well as histochemical staining with basic dyes is stressed.

    Description / Table of Contents: Zusammenfassung Es wird eine einfache und handliche Methode zur Darstellung von Kohlenhydratderivaten in Gewebsschnitten beschrieben. Entparaffinierte Schnitte werden 10–15 min in einem 1:1 Essigsäure-Schwefelsäure-Gemisch bzw. durch Behandlung mit Sulfurylchloriddampf während 15–20 min sulfatiert und anschließend mit einer 0,5%igen Methylenblaulösung (gepuffert aufph 2,4) gefärbt. Man erhält eine stark selektive Färbung der Gewebskohlenhydrate. Das Essigsäure-Schwefelsäure-Gemisch hydrolysiert und entfernt das Glykogen sehr rasch. Demgegenüber ist nach Sulfurylchloridbehandlung eine starke Anfärbung von Glykogen möglich. Die einzelnen Schritte der Technik werden diskutiert, besonders die Gründe, die uns bei der endgültigen Wahl der Methodik geleitet haben. Die Methode bietet verschiedene Vorteile gegenüber der PAS-Reaktion: Das Übersichtsbild ist klarer, die Schärfe ist bei starken Vergrößerungen oft besser; die Plastizität der Methode ermöglicht es, Sulfatierung und Färbung den Bedürfnissen verschiedener Probleme anzupassen.
    Notes: Summary A simple and convenient procedure has been devised for the staining of carbohydrate derivatives in tissue sections. Dewaxed sections are sulphated either by immersion in a 1:1 acetic-sulphuric acid mixture for 10–15 minutes or by exposure to sulphuryl chloride vapour for 15–20 minutes. The slides are then stained in a 0.05% solution of methylene blue buffered to aph of 2.4. Highly selective staining of tissue carbohydrates is obtained. The acetic-sulphuric mixture rapidly hydrolyses glycogen which is therefore lost, but the sulphuryl chloride technique produces intense staining of glycogen. The various steps in the technique are discussed, with particular emphasis on the reasons for the final choice of conditions. These sulphation techniques have several advantages over the PAS procedure: the overall picture obtained is clearer; definition at high magnifications is often better; and the versatility of the technique can be widened by varying the precise conditions of sulphation and staining to suit the particular problem being studied.

    Notes: Summary Indoramin, a new antihypertensive drug shown to have cardioinhibitory and alpha adrenoceptor blocking activity in animals, was administered to patients with benign essential hypertension. Single oral doses between 0.1 and 1.0 mg/kg produced dose dependent falls in diastolic arterial pressure without increasing the heart rate. The arterial pressure response to intravenous noradrenaline was diminished in patients taking indoramin, but the response to intravenous angiotensin II and heart rate responses to intravenous isoprenaline were unaltered. Double blind cross-over comparison of indoramin with placebo in 8 patients on continuous oral therapy at maximally tolerated doses, 60–150 mg daily, showed that indoramin significantly lowered diastolic arterial pressure, but that the falls were modest, 7.6 mm Hg lying, 7.1 mm Hg standing and 9.3 mm Hg after exercise. The dose limiting side effect was sedation.

    Notes: Abstract The yellow-cheeked vole (Microtus xanthognathus) shows two electrophoretic hemoglobin components. Electrophoresis of the polypeptide chains from the separated hemoglobin components shows identical β-chains but two α-chains of different mobility, α f and α s . The composition of soluble tryptic peptides was determined for each α-chain. Amino acid differences were found in peptides αT1 and αT9; the compositions of the remainder of the homologous peptides were identical. Differences in αT1, found at α4 (α s -Gly-α f -Val) and α5 (α s -Thr-α f -Asp), were confirmed after a run to residue 20 of the fast component in an automatic sequencer. The differences in charge between αT1 peptides can account for the electrophoretic pattern of two hemoglobins. This is the first time that it has been possible to identity the residues which can account for the charge difference between the two hemoglobins observed in a Microtus species.

    Notes: Abstract Advanced-generation domestication programs for forest-tree species has raised some concerns about the maintenance of genetic diversity in forest-tree breeding programs. Genetic diversity in natural stands was compared with two genetic conservation options for a third-generation elite Pinus taeda breeding population. The breeding population was subdivided either on the basis of geographic origin and selection goals (multiple-population or MPBS option) or stratified according to genetic value (hierarchical or HOPE option). Most allelic diversity in the natural stands of loblolly pine is present in the domesticated breeding populations. This was true at the aggregate level for both multiple-population (MPBS) and the hierarchical (HOPE) populations. Individual subpopulations within each option had less genetic diversity but it did not decline as generations of improvement increased. Genetic differentiation within the subdivided breeding populations ranged from 1 to 5%, genetic variability is within each subpopulation rather than among subpopulations for both MPBS (〉95%) and the HOPE approaches (〉98%). Nei's Gst estimates for amongpopulation differentiation were biased upwards relative to estimates of θ from Weir and Cockerham (1984).
