Notes: Abstract An experimental investigation on electrophoresis of calcium oxalate monohydrate in several liquid mixtures (methanol-, ethanol-, 2-propanol-, and methanol-acetonewater) is described. The electrophoretic transport is considered from the view-point of the Thermodynamics of Irreversible Processes. Linear relations have always been found between the mass flux and the applied electric field. The ζ-potential of the interface C.O.M./liquid mixture has been estimated by using the classial theory of electrophoretic mobility of colloidal particles. The influence of the composition of the mixture on the electrophoretic coefficient has been discussed on the basis of the variation of ζ-potential, viscosity and dielectric constant with the molar fraction of alcohol for binary mixtures and acetone for ternary mixtures. The concentration dependence of the electrophoretic coefficient is found to be linear only in the latter case.

Notes: Abstract The Doppler-free line shape of the 6s 2 1 S 0 − 6s 16p 1 1 P 1 transition at 553.5 nm in natural Ba in the presence of an argon or helium buffer gas has been measured at 744 K for gas pressures from 0.2 to 100 Torr. Using the measured data for pressures above 5 Torr, the broadening rate coefficients for the half width at half maximum (HWHM) are determined to be (4.9±0.5)⊗10−9 and (5.0±0.5)⊗10−9 s−1 cm−3 for helium and argon respectively.

Notes: Abstract Hydrolyzed olive pit residues from the production of furfural were ammoniated to obtain a nitrogenenriched humic fertilizer. Samples had been previously oxidized with nitric acid. The oxidized samples were further ammoniated in a batch reactor in the following range of experimental conditions: T = 80−210°C, $$C_{HNO_3 } = 0.5 - 5.0 wt\%$$ , solid/liquid ratio, S/L = 0.5−2.8 w/w and reaction time, t = 1−4 h. Planning of experiments was made by statistical design and results were fitted by multiple non-lineal regression polynomials. The product consists of two fractions: a water soluble fraction which contains most of the nitrogen (10−13 wt%) and a solid fraction with a very low nitrogen content (〈4.0 wt%), which is likely to be fixed during the oxidation step. The soluble fraction can be used as a humic fertilizer and the solid product as a soil conditioner.

Notes: Abstract Tadpoles of Rana perezi were kept for 14 weeks in water containing two sublethal levels of the carbamate insecticide ZZ-Aphox® or the organophosphate Folidol®. Approximate concentrations of their active ingredients were 0.25 and 1 mg/L. Resulting malformations were studied by skeletal analysis and histological and histochemical investigation of the rear limbs of the tadpoles. The pesticides caused the animals to have malformations of the spinal column (scoliosis) and/or limbs (short and thick long bones with the epiphyses grossly twisted). Histochemical study showed differences in the composition of the connective matrix, and microscopic examination of the long bones indicated alterations in the thickness of the uncalcified bone matrix (osteoid) and the presence of abundant vascularised connective tissue in the region of the periosteum. The results confirmed changes in the composition of the connective tissue matrix as the cause of the defects observed in bone formation which are also discussed in relation to vitamin D absorption and calcium homeostasis.

Notes: The aim of the present work is to prepare and characterize a functionalized latex with acetal groups on the surface and to obtain the covalent coupling of an a-CRP IgG protein. The acetal latex was synthesized by means of a core-shell emulsion polymerization in a batch reactor. The core was a seed of polystyrene and the shell was obtained by terpolymerization of styrene, methacrylic acid and methacryloylacetaldehyde di(n.methyl)acetal. The latex was characterized by TEM and conductimetric and potentiometric titration, in order to obtain the particle size distribution and the amount of carboxyl and acetal groups on the surface, respectively. Several latex-protein particles with the IgG physically or chemically bound to the surface were obtained by modifying the incubation conditions. In the covalent coupling experiments of the IgG, the protein physically adsorbed was removed by redispersion of the complexes in the presence of a non-ionic surfactant (Tween 20). The latex-protein complexes were characterized from the electrokinetic point of view with the aim to determine the isoelectric point of the complexes and to detect any difference in the electric state of the protein when these molecules are physically or chemically coupled to the surface. The final part of this work was to study the immunoreactivity of several latex-IgG complexes at several experimental conditions. By measuring the change in the turbidity after the addition of CRP antigen into the dispersion, it was possible to compare the immunoreactivity results when the protein is physically or chemically bound to the surface, and to study the effect of the presence of a surfactant in the reaction medium.

Notes: Abstract Mature embryos of Amaranthus hypochondriacus (amaranth) were used to develop an in vitro culture system for plant regeneration and genetic transformation. Plants were regenerated from embryo-derived callus cultivated on Murashige and Skoog medium supplemented with 10 µM 2,4-dichlorophenoxyacetic acid or 3,6-dichloro-2-methoxybenzoic acid and 10% coconut liquid endosperm. Transgenic plants were obtained by inoculation of mature embryo explants with a disarmed Agrobacterium strain containing the plasmid pGV2260(pEsc4), which carried the genes encoding neomycin phosphotransferase type II and β-glucuronidase. The presence of transgenes in the genome of transformed amaranth plants and their progeny was demonstrated by Southern blot hybridization. Tissue specific and light-inducible expression directed by a pea chlorophyll a/b-binding protein promoter was observed in transgenic amaranth plants and their progeny.

Notes: Summary The dry mass of reaction products in ultrathin sections was determined using electron micrographs of polystyrene spheres of known weight deposited on Formvar membranes and evaluating the negatives photometrically. This method was applied to the quantification of the final reaction product of the acid phosphatase reaction in a model system in which enzyme was incorporated in gelatin. The enzyme activity was demonstrated by the lead precipitation method and quantified by direct microphotometry at the light microscope level. Models were then embedded and sectioned for electron microscopy. Microphotometric values afforded by the electron negatives were in linear correlation with incubation times and enzyme concentration. Section thickness and its possible variations due to deformation or contamination under the electron beam were also evaluated. Measurements of lysosomal acid phosphatase activity in rat kidney sections served to illustrate the application of the technique.

Notes: Abstract  The latex agglutination immunoassay technique uses polymer colloids as a carrier for the adsorbed proteins to enhance the antigen–antibody reaction. Competitive co-adsorption of IgGaCRP and m-BSA proteins on polystyrene latexes with different functionality (sulfate and sulfonate groups) was carried out looking for the increase in the immunoreactivity and colloid stability of latex–protein complexes. The preferential adsorption of a protein is also studied, comparing both surface types. Regarding the application in the development of a diagnostic test system, it is necessary to study the latex–protein complexes from an electrokinetic and colloid stability point of view. The presence of protein on the surface latex shifts the iso-electric point (i.e.p.) of the latex–protein complexes to pH values near the i.e.p. of the protein which is the majority. Thus by the adsorption of m-BSA we can obtain complexes with the i.e.p. near pH 5 and, therefore, with a significant electrostatic repulsion at neutral pH. Due to the higher surface charge density of the sulfonate latexes there is a higher adsorption of both proteins, which can provide a better colloidal stability (by the adsorption of m-BSA) and a better immunoreactivity (by the adsorption of IgG).

Notes: Abstract The structure factorS(q) was measured for a colloidal dispersion of latex microspheres (40 nm of diameter) by both static and dynamic light scattering (using a Malvern Instruments spectrometer, λ=488 nm). The structure was formed for a sample of particle number density 2×1013 ml−1. The interaction potential was determined from the experimentalS(q) by means of the straightforward use of three different closures (Percus-Yevick, hipernetted chain, mean spherical approximation). Some difficulties found with this procedure (such as the cut-off effect) are discussed. However, these closures lead to very similar results. Furthermore, the results are tested and compared with the simulations based on a Yukawa potential.
