    Notes: Abstract Equations for the time-dependent concentrations of all species involved in the general mechanism of human plasminogen activation proposed by Wohlet al. (J. biol. Chem. 255, 2005–2013, 1980) have been derived. These equations are valid for the whole course of the reaction: for both the transient phase and the steady state. In addition, we compare our results with the ones obtained by the above-mentioned authors for the steady state assuming rapid equilibrium conditions. Finally, we propose a method for the determination of all velocity constants.

    Description / Table of Contents: Summary Forty five soil samples, most of them from coffee plantations at more than 500 m altitude, from several parts from Venezuela were inoculated intravenously into mice. Paracoccidioides brasilicnsis was not isolated. The authors discuss the possible reasons of the negative results and propose other techniques to apply in the future in order to isolateParacoccidioides brasilicnsis from nature.
    Abstract: Résumé Quarante cinq échantillons de terre furent inoculés par voie intravéneuse — des souris. Les échantillons provenaient principalement de plantations de café — plus de 500 m d'altitude de plusieurs régions du Venezuela. LeParacoccidioides brasiliensis n'a pu etre isolé. Les auteurs discutent les causes éventuelles de cet échec et proposent des méthodes différentes pour reprendre la recherche duParacoccidioides brasiliensis dans la nature.
    Notes: Resumen Cuarenta y cinco muestras de tierra, preferentemente de cafetales a más de 500 m de altitud sobre el nivel del mar, de varias regiones de Venezuela fueron inoculadas por vía endovenosa en ratones. No pudo aislarse elParacoccidioides brasiliensis. Se discuten brevemente las posibles causas de los resultados negativos y se proponen otras téchnicas para seguir intentando la determinación del habitat deParacoccidioides brasiliensis.

    Notes: Abstract The paper presents an analysis of the losses due to irreversible Barkhausen jump of the 180° Bloch wall in the single-domain wall model. The field-intensity and current-density vectors at unit input were determined for a single instantaneous jump. The losses were calculated for the case of the wall moving sinusoidally. The Parseval formula was used at determining the losses, which made it possible to perform calculations. A similar problem was considered by Allia and Vinai who adopted a simplified assumption of the domain of an infinitely long cylindrical shape.

    Notes: Abstract A newly isolated aerobic hydrogen-oxidizing bacterium, Alcaligenes denitrificans strain 4a-2, differs from related autotrophic bacteria by containing only a single cytoplasmic, NAD-reducing hydrogenase, and by its high resistance to nickel ions, i.e. tolerance to 20 mM NiCl2. Strain 4a-2 harbors a single plasmid of about 250 kb. On helper-assisted mating of 4a-2 with Alcaligenes eutrophus strains H16,G29, and M85 nickelresistant transconjugants were selected; these did not contain the donor plasmid, however. All three transconjugants tolerated 3 to 10 mM NiCl2. The resistance was constitutively expressed. DNA/DNA hybridization showed homology with EcoRI-digested DNA of the wild type 4a-2 and transconjugants using a DNA probe containing nickel resistance genes of pMOL28. This indicated that the 4a-2 nickel resistance genes are located on the chromosome.

    Notes: Abstract Cytochrome cd 1-nitrite reductase and nitrous oxide reductase of Thiobacillus denitrificans were purified and characterized by biochemical and immunochemical methods. In contrast to the generally soluble nature of the denitrification enzymes, these two enzymes were isolated from the membrane fraction of T. denitrificans and remained active after solubilization with Triton X-100. The properties of the membrane-derived enzymes were similar to those of their soluble counterparts from the same organism. Nitrous oxide reductase activity was inhibited by acetylene. Nitrite reductase and nitrous oxide reductase cross-reacted with antisera raised against the soluble enzymes from Pseudomonas stutzeri. The nirS, norBC, and nosZ genes encoding the cytochrome cd 1-nitrite reductase, nitric oxide reductase, and nitrous oxide reductase, respectively, from P. stutzeri hybridized with genomic DNA from T. denitrificans. Cross-reactivity and similar N-terminal amino acid and gene sequences suggest that the primary structures of the Thiobacillus enzymes are homologous to the soluble proteins from P. stutzeri.

    Notes: Abstract The insertion mutant HF231 of Alcaligenes eutrophus H16 failed to grow anaerobically on nitrate and nitrite. When grown under oxygen limitation, mutant HF231 specifically excreted coproporphyrin III, an intermediate of heme biosynthesis. With the help of a Tn5-labeled fragment, we identified and cloned the corresponding wild-type fragment. Sequence analysis of the mutant locus revealed an open reading frame consisting of 1,473 bp, predicting a protein of 491 amino acids that corresponds to a size of 54.2 kDa. In the non-coding upstream region, consensus elements that are indicative for binding sites of the anaerobic transcriptional regulator Fnr were identified. The deduced polypeptide showed extensive sequence similarity with various bacterial oxygen-independent coproporphyrinogen III oxidases designated HemN. HemN catalyzes the oxidative decarboxylation of coproporphyrinogen III to yield protoporphyrinogen IX. Anaerobic growth on nitrate and nitrite of mutant HF231 was restored by introducing the hemN gene of A. eutrophus or of Pseudomonas aeruginosa on a broad-host-range vector. Likewise, the A. eutrophus hemN complemented heme biosynthesis of a Salmonella typhimurium hemF/hemN double mutant during anaerobic and aerobic growth. Analysis of a transcriptional lacZ gene fusion showed that expression of hemN in A. eutrophus is nitrate-independent and repressed by oxygen.

    Notes: Summary The AQUA and PROCHECK-NMR programs provide a means of validating the geometry and restraint violations of an ensemble of protein structures solved by solution NMR. The outputs include a detailed breakdown of the restraint violations, a number of plots in PostScript format and summary statistics. These various analyses indicate both the degree of agreement of the model structures with the experimental data, and the quality of their geometrical properties. They are intended to be of use both to support ongoing NMR structure determination and in the validation of the final results.

    Notes: Abstract Dihydrofolate reductase has successfully been used as a drug target in the area of anti-cancer, anti-bacterial and anti-malarial chemotherapy. Little has been done to evaluate it as a drug target for treatment of the trypanosomiases and leishmaniasis. A crystal structure of Leishmania major dihydrofolate reductase has been published. In this paper, we describe the modelling of Trypanosoma cruzi and Trypanosoma brucei dihydrofolate reductases based on this crystal structure. These structures and models have been used in the comparison of protozoan, bacterial and human enzymes in order to highlight the different features that can be used in the design of selective anti-protozoan agents. Comparison has been made between residues present in the active site, the accessibility of these residues, charge distribution in the active site, and the shape and size of the active sites. Whilst there is a high degree of similarity between protozoan, human and bacterial dihydrofolate reductase active sites, there are differences that provide potential for selective drug design. In particular, we have identified a set of residues which may be important for selective drug design and identified a larger binding pocket in the protozoan than the human and bacterial enzymes.

    Notes: Abstract Euglena gracilis and Chlamydomonas reinhardtii were used as model organisms to establish the best conditions for studying photosynthetic efficiency using the Light Pipette – experimental system, which enables sensitive detection of changes in oxygen evolution/consumption and an easy collection and digitalisation of data. Chlorophyll concentrations of 0.005, 0.025, 0.050 and 0.075 mgmL-1 were investigated using different light regimes. Cultures of E. gracilis at the same chlorophyll concentration absorbed more light(measured at 580 μmol m-2 s-1) than those of C. reinhardtii. Cell density had a considerable effect on the reliability of measurements. Chlorophyll concentrations between 0.025 mgChl ml-1 and 0.050 mgChlml-1 can be recommended when applying the Light Pipette system in bioassays using microalgae.
