    Notes: Abstract A method for rapidly determining the vertical and horizontal distribution of insect eggs in fragile soil is described. Liquefied agar is allowed to permeate intact soil samples from below; after cooling, the resulting solid is cut into thin sections, from which eggs can be recovered by washes with hot water. This technique revealed that in organic (muck) soil in the laboratory, undisturbed onion flies, Delia antiqua (Meigen) (Diptera: Anthomyiidae), laid 95% of their eggs within a 10 mm diameter zone around the base of a surrogate onion ‘stem’ arising vertically from the soil. Ninety % of all eggs were found in the top 12 mm of soil, with an apparent maximum at depth of 2–4 mm. Increasing fly density from 30 to 200 flies per 30×30×42 cm cage flattened the horizontal distribution of eggs and extended the ovipositional range from c. 15 mm to beyond 60 mm, suggesting there was competition for the preferred ovipositional sites. Surface treatment of muck soil in the field by granular formulations of the insecticides Lorsban™ 15 G (active ingredient chlorpyrifos) and Dyfonate™ 15 G (fonofos) three weeks prior to bioassaying reduced egg-laying at depths greater than 8 mm. The relation between the measured egg distribution and mortality factors in soil (low moisture and high temperature) is discussed.

    Notes: Abstract Behavioral responses of female onion flies, Delia antiqua (Meigen), to hostplant cues were quantified during encounters of individual flies with onion plants and onion foliar surrogates. The behavioral repertoire of such females included sitting, grooming, running up and down foliar surfaces, extension of the proboscis such that the labellum contacted foliar and soil surfaces, movements of the tip of the abdomen over surfaces (surface probing), subsurface probing of soil crevices with the ovipositor, and oviposition. Sequences of behaviors preceding oviposition were probabilistic rather than highly stereotyped but generally followed the order given above. Foliar surrogates were used to determine the effects of n-dipropyl disulfide (Pr2S2) on the sequence of behaviors leading up to oviposition. The addition of a Pr2S2-treated surrogate to a cage increased the frequency of alighting on that surrogate but also increased alighting on a nearby foliar surrogate without Pr2S2. After alighting, females encountering surrogates treated with Pr2S2 had shorter latencies to proboscis extension and surface probing, spent less time sitting and grooming, and had runs of shorter duration. These females were also more likely to make the transition from probing of surfaces of foliage and soil to subsurface probing of soil crevices and oviposition. Thus, rather than mediating a particular step in the behavioral sequence, Pr2S2 played a role throughout the sequence leading up to oviposition. Collectively, these data and past studies on the onion fly support the hypothesis that egglaying is triggered by a temporal summation of inputs to the central nervous system from various sensory modalities rather than strict behavioral chaining, with each transition effected by some unique cue.

    Description / Table of Contents: Résumé A la lumière d'opinions récentes concernant la découverte des plantes-hôtes par D. antiqua, nous avons examiné les conditions de la ‘résistance’ chez plusieurs lignées de A. cepa les moins attaquées lors d'essais en plein champ aux Pays-Bas, et dont on considérait que la ‘résistance’ était due à une antixénose. Des femelles gravides en présence, au laboratoire, d'un choix de plants de 6 semaines, pondent en moyenne 1,6 à 34,8 oeufs par plant. Les différences entre les pontes correspondent aux tailles des plants. Si l'on tient compte de ces dernières, l'analyse de covariance ne montre pas de différence significative entre les pontes sur les différentes lignées. Des substituts de feuilles ont été créés de façon à faire varier les paramètres de taille tout en maintenant constants les autres stimuli de la plante: pour des plantes, dont le diamètre à la base variait de 1 à 4 mm et la hauteur de 100 à 350 mm, seul le diamètre a significativement influé sur la ponte. Les pontes sur les plantes hors de ces dimensions n'ont pu être expliquées par les seules différences de diamètre. Comme les paysans des principales régions productrices d'oignons d'Europe et des U.S.A. font des bénéfices importants avec la culture d'oignons précoces, ces lignées, sélectionnées par mégarde pour leur développement lent, ne sont probablement pas une bonne source de ‘résistance’ à la mouche de l'oignon. Cependant, ces résultats soulignent la nécessité de tenir autant compte de la taille de la plante que de son chimisme ou de sa texture lors de l'évaluation et de la création de cultivars résistants.
    Notes: Abstract When gravid onion fly females, Delia antiqua (Meigen) (Diptera: Anthomyiidae) were presented in laboratory choice test with 6-week-old plants of a susceptible cultivar and of onion breeding lines selected for resistance, mean numbers of eggs laid ranged from 34.8 to 1.6 eggs per plant. Differences in ovipositional responses were mirrored by differences in plant size. Analysis of covariance revealed no significant differences in ovipositional responses to breeding lines when differences in size were taken into account. Foliar surrogates were developed so that single size parameters could be varied while holding all other plant stimuli constant. Tests using these surrogates revealed that among plants with basal diameters of 1 to 4 mm and heights of 100 to 350 mm, diameter alone significantly influenced responses of ovipositing females. Ovipositional responses to plants beyond this size range could not be explained strictly by diameter differences. These results underscore the necessity of considering plant size as well as plant chemistry and texture in efforts to evalute and design resistant cultivars.

    Notes: Summary To test the importance of 24-hydroxylation of vitamin D3 on bone mineralization, rat pups born to vitamin D-deficient females were given either 25-hydroxyvitamin D3 or 24,24-difluoro-25-hydroxyvitamin D3 for 16 days beginning at the time of weaning. Following such treatment analysis of blood samples revealed no detectable 24R,25-(OH)2D3 and 1,25-(OH)2D3 in the rats given the difluoro compound while revealing the expected 24,24-difluoro-25-hydroxyvitamin D3 and 24,24-difluoro-1,25-dihydroxyvitamin D3. The rats given 25-hydroxyvitamin D3 had the expected levels of 25-hydroxyvitamin D3, 24,25-dihydroxyvitamin D3, and 1,25-dihydroxyvitamin D3. Following sacrifice at day 17, postweaning bone mineralization and modeling were studied in long bones using histological methods. Bones taken from vitamin D-deficient rats at the beginning and end of the experimental period had lesions typical of rickets. These included wide growth plates, excessive amounts of osteoid, and metaphyseal fibrosis. Following treatment with either 25-hydroxyvitamin D3 or 24,24-difluoro-25-hydroxyvitamin D3, bone mineralization returned to normal. Growth plate widths and the amount of osteoid on bone surfaces were both substantially reduced and to a similar degree in both treatment groups. Normal cartilage core formation and trabecularization of the metaphyseal primary spongiosa were also restored to a similar degree in both groups. In effect, no difference was observed in any bone parameter studied between the 25-hydroxyvitamin D3- and the 24,24-difluoro-25-hydroxyvitamin D3-treated animals. These results provide strong evidence that 24-hydroxylation of the vitamin D molecule plays little or no role in the modeling and mineralization of bone.

    Notes: Summary Male rats weighing 100 g were assigned to groups and injected daily for 10 days with vehicle (control), 0.4, 2.0, 4.0, 10.0, or 20.0 mg/kg/day of ethane-1-hydroxy-1,1-diphosphonate (EHDP) or dichloromethylene diphosphonate (Cl2MDP). The proximal tibial metaphysis and epiphysis were assayed for changes in percentage of hard tissue and bone formation parameters. From the data, information about hard tissue resorption was deduced. All doses of Cl2MDP and doses of 2.0 mg EHDP/kg/day and greater caused significant increases in percentage of hard tissues with Cl2MDP being more effective than similar doses of EHDP in decreasing bone resorption. Osteoclast population parameters were increased with all doses of both Cl2MDP and EHDP with Cl2MDP having a greater effect than similar doses of EHDP. Decreases in the proliferation of the osteoprogenitor pool parallel the decreases in osteoblasts and bone formation parameters. These decreases in osteoprogenitor pool proliferation do not account for the increases with diphosphonates in osteoclast population parameters.

    Notes: Summary The role of vitamin D in early skeletal development was studied by measuring serum calcium and phosphorus, osseous tissue quantity and mineralization, and endochondral bone elongation in rat fetuses and pups from vitamin D-replete and vitamin D-deficient mothers. At the 20th day of pregnancy there was a slight, yet significant, increase in the amount of osteoid on trabecular bone surfaces in fetuses from vitamin D-deficient mothers. The fetal bones otherwise appeared normal in spite of severe skeletal changes in the vitamin D-deficient mothers. After parturition, the importance of vitamin D in skeletal development becomes progressively more obvious. Serum calcium levels were slightly, yet significantly, lower in vitamin D-deficient than in vitamin D-replete pups and these levels continued to fall in the vitamin D-deficient pups through lactation and after weaning. At 3 days postpartum, there was a small, yet significant, increase in the amount of osteoid on bone surfaces of the vitamin D-deficient pups. The relative amounts of osteoid in the vitamin D-deficient pups continued to increase through lactation and after weaning when compared with vitamin D-replete pups. By the 14th day of lactation and at later periods, there were significant reductions in metaphyseal mineralized tissues in the vitamin D-deficient pups when compared with the vitamin D-replete pups. At weaning and after weaning, there were substantial increases in growth plate thickness and decreases in longitudinal bone growth in the vitamin D-deficient pups when compared with the vitamin D-replete pups. The results from this study indicate that vitamin D does not appear to play a major role in fetal skeletal development. However, after birth, vitamin D becomes progressively more important with age for normal bone development, mineralization, and endochondral growth.

    Notes: Abstract Histochemical methods were used to detect differences in amino acid reactive groups between bone and osteoid, dentine and predentine. Three major differences were found: Predentine and osteoid contained more reactive sulphydryls than their mineralized counterparts. The greatest reaction for this reactive group was obtained at the mineralizing front. Dentine and bone contained a complex between amino and carboxylic acid groups; predentine and osteoid did not have this complex and carboxylic acid residues were less in these tissues. Dentine and bone contained much greater amounts of histidine or a histidine-like component than did predentine and osteoid. It is proposed that these differences are involved in initiation of mineralization.

    Notes: Summary A fraction (brA), which causes resorption of fetal rat bones in vitro, has been concentrated from bovine serum albumin by anion exchange column chromatography on DEAE Sephadex. This active fraction has also been prepared using DEAE Sephadex A-50 by a batch method with a 0.09M NaCl, 0.1M TRIS buffer, pH 8.35. BrA was 10–30 times more potent than the original albumin. The retained material, which constitutes the bulk of the protein and has less activity than the original albumin, elutes with 0.45M NaCl. Similar treatment of serumα,β or γ globulins does not yield brA. Further enhancement of the bone resorbing activity of brA can be obtained with (NH4)2SO4 fractionation or extraction with CH3OH∶CHCl3. Heating at 55° C for 2 h or at 100° C for 10 min does not affect the activity; overnight incubation with protease destroys the bone resorbing effect. The bone resorbing activity is not removed by dialysis and does not correlate with the protease activity of the fraction. The action of brA is inhibited by 3 mM PO4, 1 μg/ml calcitonin or glucagon, 10−7 M dexamethasone or 0.02 μg/ml actinomycin D. The bone resorbing activity of brA is partially inhibited by 10−7–10−5 M indomethacin. PTH did not elicit bone resorption when added to cultures incubated in chemically defined medium supplemented with 0.1 mg/ml brA. However, brA did not inhibit PTH-induced resorption.

    Notes: Summary The effect of vitamin D on bone changes during the reproductive cycle in female rats has been investigated. One group of female rats was maintained on a vitamin D-deficient diet and another group on a vitamin D-replete diet from weaning. Both groups were mated with normal males and changes in their bones were determined histomorphometrically during pregnancy, lactation, and after weaning. All vitamin D-deficient rats had bone changes typical of rickets. Pregnancy caused significant reductions in mineralized tissue of trabecular and cortical bone in the vitamin D-deficient rats. Lactation caused further significant reductions in mineralized tissues of cortical and trabecular bone in both the vitamin D-deficient and vitamin D-replete animals, with the greatest changes seen at weaning. Some restoration of mineralized tissues occurred following weaning. There was an increase in tetracycline-labeled bone surface in the vitamin D-replete animals during lactation, likely due to an increase in bone formation rates. In the vitamin D-deficient animals during lactation, there was a decrease in tetracyclinelabeled bone surface, likely due to severely depressed bone mineralization. These results indicate that the mobilization of calcium from bone to maintain pregnancy and lactation occurs by a mechanism independent of vitamin D.

    Notes: Abstract Male rats weighing 100 g were injected with vehicle (control group), 0.4 or 4.0 mg/kg/day of ethane-1-hydroxy-1,1-diphosphonate (EHDP). Beginning after 6 days of EHDP treatment all rats received six subcutaneous injections of3H-thymidine at 8 h intervals. The multiple labeling index of autoradiographically defined labeled cells was determined for the osteoprogenitor cells of the primary spongiosa of the proximal tibial metaphysis. The osteoclasts in the proximal tibial metaphysis were assayed for population parameters and incorporation of labeled nuclei at sequential intervals after the administration of3H-thymidine. After 6 days of EHDP pre-treatment the multiple labeling index of the osteoprogenitor cells was reduced with 4.0 mg/kg/day dose of EHDP. At the beginning of the experimental kinetic sampling period, after the 6 days of EHDP pre-treatment, EHDP treatment resulted in significant increases in the numbers of osteoclasts, nuclei per osteoclast profile, and total osteoclast nuclei. During the sampling period, the 4.0 mg/kg/day dose of EHDP caused further increases in the number of total osteoclasts. EHDP treatment caused a marked increase in the rate of incorporation and accumulation of3H-thymidine-labeled nuclei in osteoclasts. Thus, EHDP causes an accelerated rate of osteoclast production from precursor cells in spite of its ability to slow bone resorption. Although the origin of osteoclast nuclei is not clear, the kinetic data from this study suggests that osteoclast nuclei are not arising from osteoprogenitor cells or osteocytes in the EHDP-treated animals.
