    Notes: Abstract Starting from a complex operator of derivation, we give expressions for derivatives of arbitrary order of the gravity potential with respect to rectangular coordinates. These expressions have a form similar to the original potential expanded in spherical harmonics and are free of singularity at the poles. Computing sets of numerical coefficients once for all, we can compute the derivatives with a very limited work: the same functions are used to compute all derivatives by means of a unique parametrized formula. This is very comfortable for further algebraic manipulations. Numerical tests prove the accuracy and the efficiency of the algorithm derived from our formula to compute the gravity acceleration vector and the gravity gradient tensor.

    Notes: Summary Anti-derivatives of wavelets are used for the numerical solution of differential equations. Optimal error estimates are obtained in the applications to two-point boundary value problems of second order. The orthogonal property of the wavelets is used to construct efficient iterative methods for the solution of the resultant linear algebraic systems. Numerical examples are given.

    Description / Table of Contents: Zusammenfassung Eine Einwaage von 1–10 mg Titandioxid wird mit 35μl 28M Flußsäure in einem geschlossenen Teflongefäß aufgeschlossen. Im selben Gefäß werden mehr als 95% der anwesenden Bleimenge aus 250μl Elektrolyt bei pH 4,3 an einer glasförmigen Kohle-Mikroelektrode abgeschieden und der Differential-Puls-Anodic-Stripping-Voltammetrie zugeführt. Geringe Bleimengen lassen sich mit Hilfe dieses Verfahrens mit einem Fehler von weniger als 5% innerhalb einer Stunde bestimmen.
    Notes: Summary A 1- to 10-mg sample of titanium dioxide powder is decomposed with 35μl of 28M hydrofluoric acid in a closed Teflon microvessel. In the same vessel, more than 95% of lead is electrodeposited on a glassy carbon microelectrode from 250μl of electrolyte at pH 4.3 for differential pulse anodic stripping voltammetry. Low ppm of lead is determined by the proposed method with an error of less than 5% within 1 h.

    Notes: Abstract Using the self-template technique, c-axis-oriented epitaxial YBa2Cu3O7 − δ thin films have been prepared in situ on LaAlO3 substrate by the d.c. magnetron sputtering method. The properties of thin film dependence on the deposition conditions of the two-step self-template method have been systematically investigated. By optimizing the parameters, high-quality YBCO thin films with T c0 ≥ 90 K, ΔT c ≤ 1 K, R s (77 K, 10 GHz)∼500 μΩ were reproducibly obtained. The best sample grown under optimal conditions gave a low R s of 330 μΩ at 77 K, 10 GHz, which can be used in a microwave field.

    Notes: Abstract Our objective was to determine the role that bone marrow-derived stromal cells have on human hematopoiesis in HIV infection. In particular, we dissected the heterogeneous bone marrow microenvironment to study the effect HIV expression might have on the cell population capable of producing the cytokines which will support human CD34+ cell differentiation. A stromal cell line, Lof(11-10), was established from human bone marrow by transfecting a plasmid containing the SV40 large T-antigen and isolating foci exhibiting a transformed phenotype. The Lof(11-10) cell line was characterized to determine its susceptibility to HIV infection, to identify its cytokine production profile, and to test the ability of conditioned media from this line to support CD34+ cell differentiation in the presence and absence of HIV expression. Nine cytokines were detected by RT-PCR and ELISA analysis. Conditioned media obtained from the Lof(11-10) cell line was able to support CD34+ cell differentiation. However, because the Lof(11-10) cells are not infectible by HIV, molecular clones of HIV were introduced into these cells by transfection. There was no qualitative difference in the levels of cytokine production between HIV-expressing and control Lof(11-10) cells. Furthermore, conditioned media derived from HIV-expressing and control Lof(11-10) cells added to bone marrow-derived CD34+ progenitor cells yielded similar colony formation in methylcellulose assays. Our data suggest that HIV infection of the cytokine-producing cells within the bone marrow microenvironment, as represented by the Lof(11-10) cell line, results in both normal cytokine production and hematopoiesis in spite of HIV expression. This report adds to the evidence against stromal cells being a significant target of HIV and establishes a system for comparison with more relevant models.

    Notes: Abstract A novel fluorometric method has been developed for the determination of total protein in human serum with a new near-IR reagent as a fluorescence probe, based on the fluorescence recovery of the cyanine-CTAB system in the presence of protein. Maximum fluorescence is produced with maximum excitation and emission wavelengths at 765 and 812 nm, respectively. Under optimal conditions, the calibration graphs are linear over the ¶range 0.4–12.0 μg/mL for protein. The detection limit is 70 ng/mL, and the relative standard deviation of six replicate measurements is 1.14% for 6.0 μg/mL protein. The results are satisfactory.

    Notes: Abstract A new method with a cationic near-IR cyanine as fluorescent probe was developed for the determination of nucleic acids. The near-IR cyanine shows maximum excitation and emission wavelengths at 765 and 790 nm, respectively, in aqueous solution. The method is based on the fluorescence decrease of near-IR cyanine in the presence of nucleic acids. Under optimal conditions, the ratio of fluorescence intensity in the absence and presence of nucleic acids was proportional to the concentration of ¶nucleic acids over the range 0.10–1.2 μg/mL for CT (calf thymus) DNA or SM (salmon sperm) DNA, and 0.10–¶1.6 μg/mL for yeast RNA. The detection limits were ¶30 ng/mL for CT DNA, 25 ng/mL for SM DNA and ¶70 ng/mL for yeast RNA. The relative standard deviation (n = 6) was 2.1% for 500 ng/mL CT DNA, 2.4% for ¶500 ng/mL SM DNA and 2.7% for 500 ng/mL yeast RNA, respectively.

    Notes: Abstract  A sensitive method using fluorescence quenching for the determination of nucleotides (ATP, ADP, AMP, CTP, UTP) and polynucleotides[poly(A), poly(I), poly(U)] is proposed. It is based on the ability of nucleotides and polynucleotides to inhibit the formation of a strongly fluorescent complex of Tb3+ ion with Tiron. The possibilities of spectrofluorimetric measurements of these systems were studied under optimal conditions (pH 6.9 in hexamethylene tetramine-HCl buffer, 1.2×10-6 mol/L of Tb3+, 4.0×10-6 mol/L of Tiron, λex=317 nm, λem=546 nm). The results showed that the Tb3+-Tiron complex could be used as a fluorescence test for the phosphate moieties of nucleotides and polynucleotides. The detection limits are 0.3, 1.2, 3.7, 0.2, 0.3, 1.1, 0.6 and 0.9 ng/mL for ATP, ADP, AMP, CTP, UTP, poly(A), poly(I), and poly(U), respectively. The relative standard deviations (6 replicates) are within 4.0% in the middle of the linear range. The fluorescence quenching mechanism of these systems is also discussed.

    Notes: Abstract Theoretical relationships suitable for both constant-wavelength and variable-angle synchronous fluorescence in the wavelength domain were obtained for the calculation of peak location, intensity, and half-width of synchronous fluorescence spectra. The calculated values were compared with experimental data and the data derived from the literature equations, indicating that the proposed methods are feasible, less formidable, and more straightforward in the wavelength domain than previous ones. The proposed approaches provide a theoretical guide for the experimental design of synchronous fluorescence spectrometric methods in the wavelength domain. On the basis of the theoretical derivation, special characteristics of the peak half-bandwidths of the spectra were revealed. In certain cases, the bandwidth of a variable-angle synchronous spectrum is of less practical meaning.
