    Notes: Abstract The response of the colon to eating, the gastrocolonic response (GCR), may have important implications for the design of drug dosage forms for selective delivery to the colon. Therefore, the effect of meal composition on the GCR and its relation to the transit of non-disintegrating tablets has been investigated. Eight healthy male volunteers each received 5 × 6-mm radiolabeled nondisintegrating tablets, and the transit was followed using a gamma camera. When the tablets reached the ileocolonic region, each volunteer received a test meal (1000 kcal) containing 70% carbohydrate, 15% fat, and 15% protein. The subsequent movement of the tablets was then monitored. The study was repeated using a 70% fat meal and a 70% protein meal, so that the effects of a high-carbohydrate, a high-fat, and a high-protein meal on the GCR could be compared. The incidence of GCRs was similar after all meals. Thus, there appeared to be no effect of meal composition on the movement of the tablets into the colon. This implies that the ingestion of food may not necessarily stimulate the passage of material across the ileocecal junction and that other factors may also be involved.

    Notes: Abstract The aim of the present study was to investigate the effect that different concentrations of mannitol have on small intestinal transit, and whether any observed effect was concentration dependent. Eight, healthy male subjects each received 200ml of radiolabelled purified water, or a 200ml solution of mannitol at three different concentrations; 0.755g/200ml, 1.509g/200ml and 2.264g/200ml, in a randomised, four way cross-over study. Transit of the radiolabelled solutions was followed by gamma scintigraphy. The study demonstrated no significant differences between the gastric emptying times of the four solutions. Rapid gastric emptying was observed in most cases. The mean small intestinal transit times for the 0.755g/200ml, 1.509g/200ml and 2.264g/200ml mannitol solutions was reduced by 11%, 23% and 34% respectively, when compared to the control solution. The intestinal transit data of the four solutions demonstrate that mannitol has a concentration dependent effect on small intestinal transit. Small concentrations of mannitol included in a pharmaceutical formulation could therefore lead to reduced uptake with any drug exclusively absorbed from the small intestine.

    Notes: Abstract The gastric emptying of pellets and single units of different densities has been followed in healthy subjects using the technique of gamma scintigraphy. The gastric emptying of the light pellets was affected by their buoyancy in the upper part of the stomach. However, the mean gastric emptying rates of pellets and single units were not significantly affected by density. Floating or buoyant delivery systems may have little advantage over conventional systems. The presence of food in the stomach was found to be the major factor in determining the gastric emptying of single units.

The pheromone (cruentol) was identified by coupled gas chromatographic-electroantennographic (GC-EAD) analysis of male-produced volatiles, coupled GC-mass spectrometry (MS) in electron impact and chemical ionization mode, and coupled GC-high resolution MS. In laboratory and field assays, a diastereomeric mixture of synthetic cruentol greatly enhanced attraction of weevils to cabbage palmetto,Sabal palmetto (Walter), stem tissue, indicating that cruentol and host volatiles are synergistically attractive. An attractive lure in combination with efficient traps should facilitate development of semiochemical-based management forR. cruentatus.

    Notes: Abstract Laboratory and field assays were conducted to determine if palmetto weevil,Rhynchophorus cruentatus (F.), adults produce an aggregation pheromone. Attraction of females in a Y-tube olfactometer to conspecific males was greater than to clean air. Male and female attraction to conspecific male volatiles combined with host-palm,Sabal palmetto (Walter), volatiles was greater than to host-palm volatiles alone. Similarly, more weevils were caught in the field in traps baited with conspecific males plus host-palm tissue than in similar traps baited with only males, or palm tissue, or females, or females plus palm tissue. These results suggest thatR. cruentatus males produce an aggregation pheromone(s) that is highly attractive to conspecific adults of both sexes when combined with host-palm volatiles. This study is an important step towards understanding the chemical ecology ofR. cruentatus.

    Notes: Abstract The aggregation pheromones were studied from two geographical isolates (Hakalau, Hawaii, and Silkwood, Queensland, Australia) of the New Guinea sugarcane weevil, Rhabdoscelus obscurus. Coupled gas chromatographic–electroantennographic detection (GC-EAD) and GC–mass spectrometric (MS) analyses of Porapak Q volatile extract from male and from female Hawaiian R. obscurus revealed a single EAD-active, male-specific candidate pheromone, which was identified as 2-methyl-4-octanol (1). Corresponding volatile analyses from male and from female Australian R. obscurus consistently revealed three EAD-active, male-specific candidate pheromone components that were identified as 1, (E2)-6-methyl-2-hepten-4-ol (rhynchophorol) (2), and 2-methyl-4-heptanol (3). In field experiment 1 in Hakalau, Hawaii, traps baited with a stereoisomeric mixture of synthetic 1 (3 mg/day) plus sugarcane captured more weevils than did traps baited with 1 or sugarcane alone or no bait, indicating that 1 is the pheromone of the Hawaiian R. obscurus population. In field experiment 2, conducted in Silkwood, Australia, traps baited with stereoisomeric mixtures of synthetic 1, 2, and 3 (3 mg/day each) plus sugarcane caught more weevils than did unbaited traps or traps baited with 1, 2, and 3 or sugarcane. Testing candidate pheromone components 1, 2, and 3 in experiments 2–5 in all possible binary, ternary, and quaternary combinations with sugarcane, indicated that 1 and 2 in combination, but not singly, are pheromone components of the Australian R. obscurus population. Weevils from several locations in Australia and Hawaii could not be differentiated using traditional morphological characters or ultrastructural comparisons with scanning electron microscopy (SEM). However, comparisons of mtDNA sequences (cytochrome oxidase I; regions I1 to M4; 201 base pairs) revealed 5.5% variation between the Hawaiian (N = 2) and the Australian (N = 4) samples. There was no intrapopulation variation in sequence data from the weevils from Hawaii versus Australia, suggesting that they are sibling species.

The pheromone was identified by coupled gas chromatographic–electroantennographic detection (GC-EAD) analysis of male- and female-produced volatiles, and comparative GC–mass spectrometry (MS) of weevil-produced 1. In field experiments in Colombia, traps baited with a stereoisomeric mixture of synthetic 1 (3 mg/day) plus sugarcane captured more weevils than traps baited with 1 or sugarcane alone, suggesting that pheromone and plant volatiles are synergistically attractive. Use of a chiral, stereoisomer-separating Cyclodex-B column in GC-EAD and GC-MS analyses revealed that D. borassi males produce, and antennae of males and females respond to (4S,5S)-1. Previously identified palm weevil (Rhynchophorus spp.) aggregation pheromones 5-methyl-4-octanol (cruentol) and 6-methyl-2-hepten-4-ol (rhynchophorol) also elicited antennal responses by D. borassi. In field experiments, D. borassi females were captured equally well in traps baited with sugarcane plus either 1, cruentol or rhynchophorol. In contrast, D. borassi males were captured most often in traps baited with sugarcane plus 1. Because D. borassi is a potential vector of the red ring nematode that causes the lethal red ring disease of palms, pheromone-based trapping of D. borassi could aid in monitoring or management of red ring disease in commercial palm plantations.

    Notes: Abstract Coupled gas chromatographic–electroantennographic detection (GC-EAD) analyses and coupled GC-mass spectrometry (MS) of volatiles produced by male and female West Indian sugarcane weevils (WISW), Metamasius hemipterus sericeus (Oliv.), revealed eight male specific, EAD-active compounds: 3-pentanol (1), 2-methyl-4-heptanol (2), 2-methyl-4-octanol (3), 4-methyl-5-nonanol (4), and the corresponding ketones. In field experiments in Florida, alcohols 1–4 in combination with sugarcane were most attractive, whereas addition of the ketones or replacement of alcohols with ketones significantly reduced attraction. In Costa Rica field experiments testing alcohols 1–4 singly and in all binary, ternary, and quaternary combinations revealed 4 in combination with 2 was the major aggregation pheromone, equally attracting male and female WISW. Stereoisomeric 4 and (4S,5S)-4, the only isomer produced by WISW, were equally attractive. Addition of 4S-, 4R- or (±)-2 to (4S,5S)-4 significantly enhanced attraction. Sugarcane stalks in combination with 2 plus 4 (ratio of 1:8) were highly synergistic, whereas EAD-active sugarcane volatiles ethyl acetate, ethyl propionate, or ethyl butyrate only moderately increased attractiveness of the pheromone lure.

    Notes: Abstract Purpose. In this paper we report on the first scintigraphic evaluation of vaginal dosage forms in post-menopausal women. To date, almost nothing is known about the in vivo performance of pharmaceutical formulations in the human vagina, which is a major deficiency in the rational design of drug delivery systems for both existing and new indications. Methods. The vaginal spreading and clearance of a radiolabelled pessary formulation and Replens® (polycarbophil) gel, was assessed in six healthy, post-menopausal female volunteers over a six hour period using the technique of gamma scintigraphy. Results. In five out of the six subjects studied, clearance of the two formulations exhibited very little intra-subject variation. However, there was considerable inter-subject variability in clearance; in Subject 5 circa 80% of the products were retained whilst in Subject 2 less than 2% was present at the end of the six hour imaging period. Importantly, there was no evidence to suggest that either of the formulations dispersed material beyond the cervix, into the uterus, in any of the subjects studied. Conclusions. The lack of significant retention of these products in most of the volunteers has obvious implications for the delivery of therapeutic agents. This study shows that gamma scintigraphy is an invaluable technique with which to assess novel formulations aimed at optimising retention in the vagina for topical or systemic drug delivery.

    Notes: Abstract The aim of the present study was to correlate the gastric emptying (GE) of nondisintegrating tablets with changes in gastrointestinal (GI) motility. Eight, healthy, male subjects each received 5 × 7-mm radiolabeled tablets, a radiolabeled meal, and a radiotelemetry capsule (RTC). Transit of the radiolabeled formulations was followed by gamma scintigraphy and the RTC detected contractile activity in the GI tract. The study demonstrated that 7-mm tablets can empty from the fed stomach, prior to the onset of interdigestive activity. Those tablets that were not emptied during fed activity were retained through the period of quiescence associated with the onset of the migrating myoelectric complex (MMC) and left the stomach during contractions associated with phase 2 and 3 activity. The RTC was retained in the stomach and was emptied only by large phase 3 contractions commonly termed the “housekeeper” wave. However, in one subject, the RTC was retained in the stomach for over 12 hr, during which time three distinct phase 3 complexes were monitored.
