    Notes: Summary To investigate molecular evolution in a mammalian order with a comprehensive fossil record, we have constructedα-globin-like gene cluster maps for members of the order Perissodactyla. Although the arrangement of genes is the same in the five Equidae examined, the tapir and rhinoceros differ from each other and the horse in the position and number of their ζ genes, but not in the arrangement of theirα and θ genes. In contrast to morphological work, a dendrogram derived from restriction site maps associates the tapir with the horse rather than with the rhinoceros; however, this phylogeny is not statistically significant. Among the Equidae,Equus caballus emerges as an outgroup, in agreement with data from other disciplines.

    Notes: Abstract Cytogenetically undetectable deletions are suspected to be an important cause of mental retardation and developmental delay, as suggested by the observation that about 7% of children with undiagnosed mental retardation have rearrangements affecting the chromosome ends. Screening the whole genome for regions of aneuploidy smaller than 5 Mb is not feasible, but the availability of a high resolution map of the X chromosome means that it is possible to look for deletions in males by PCR. We have screened 96 affected males and their 96 unaffected fathers with 110 markers distributed across the X chromosome. No deletions were found in either group. Our results show that the prevalence of deletions greater than 1 Mb in children with mental retardation is less than 3.9% (95% confidence interval). We conclude that X chromosome deletions in the size range 1–5 Mb are a rare cause of mental retardation in males.

    Notes: Abstract. We have used a new method for binning minisatellite alleles (semi-automated allele aggregation) and report the extent of population diversity detectable by eleven minisatellite loci in 2689 individuals from 19 human populations distributed widely throughout the world. Whereas population relationships are consistent with those found in other studies, our estimate of genetic differentiation (Fst) between populations is less than 8%, which is lower than comparative estimates of between 10%–15% obtained by using other sources of polymorphism data. We infer that mutational processes are involved in reducing Fst estimates from minisatellite data because, first, the lowest Fst estimates are found at loci showing autocorrelated frequencies among alleles of similar size and, second, Fst declines with heterozygosity but by more than predicted assuming simple models of mutation. These conclusions are consistent with the view that minisatellites are subject to selective or mutational constraints in addition to those expected under simple stepwise mutation models.

    Notes: Abstract Various processes (selection, mutation, migration and genetic dirft) are known to determine the frequency of genetic disease in human populations, but so far it has proved almost impossible to decide to what extent each is responsible for the presence of a particular genetic disease. The techniques of gene and haplotype analysis offer new hope in addressing this issue, and we review relevant studies of three haemoglobinopathies: sickle cell anaemia, and α and β thalassaemia. We show how for each disease it is possible to recognize a pattern of regionally specific mutations, found in association with one or a few haplotypes, that is best explained as the result of selection; other patterns are due to population migration and genetic drift. However, we caution that such conclusions can be drawn in special circumstances only. In the case of the haemoglobinopathies it is possible because a selective agent (malaria) was already suspected, and the investigations could be carried out in relatively genetically homogenous populations whose migratory histories are known. Moreover, some data reviewed here suggest that gene conversion and the haplotype composition of a population may affect the frequency of a mutation, making interpretation of gene frequencies difficult on the basis of standard population genetics theory. Hence attempts to use the same approaches with other genetic diseases are likely to be frustrated by a lack of suitably untrammelled populations and by difficulties accounting for poorly understood genetic processes. We conclude that although this combination of molecular and population genetics is successful when applied to the study of haemoglobinopathies, it may not be so easy to apply it to the study of other genetic diseases.

    Notes: Summary Tandem-repetitive highly variable loci in the human genome (minisatellites) have been used in gene mapping and as DNA “fingerprints”, but they have not yet found much application in population genetics. We have investigated the capacity of six minisatellites to discriminate between four populations in Oceania. We find that in comparison to Melanesians, Polynesians have a significant loss of heterozygosity (or gene diversity), not noted using more traditional markers. We show also that the number of alleles, the allele distribution and the mutation rates at the Polynesian minisatellite loci do not deviate from those predicted by the neutral mutation/infinite allele model. The low gene diversity is therefore likely to be a result of the maintenance of small population sizes and bottleneck effects during the colonization of the Pacific.

    Notes: Abstract Key results of zero-field (ZF) and transverse-field (TF) muon-spin-relaxation (μSR) experiments on superconducting and insulating RBa2Cu3O y (R123 y , with R=Eu, Gd, Pr and Pr/Y:y=6, 7) are examined. The chemical behavior of the positive muon probe is addressed, and muon-oxygen bonding is shown to occur in all these cuprates. To explain magnetic fields at muon-probe sites in Pr x Y1−x Ba2Cu3O y (0〈=x〈0.5,y=7 andx=0,y=6) samples, improvements on the reported magnetic structures from neutron diffraction are necessary. Cu magnetism in Pr123y (y=6,7) is observed belowT N1, which is near RT. The magnetism seen belowT N2 can be interpreted assuming an additional ordering in the Cutt-O chain layers. Alternatively, Pr ordering is also considered as the cause of the second phase transition. Considering the specific muon-probe location, a more detailed interpretation can be provided for the μSR parameters, measured in the normal and mixed states of these unconventional superconductors.

    Notes: Abstract Transverse-field (TF) muon-spin-relaxation (μSR) and Mössbauer experiments on the uniaxial insulating spin glass Fe2−xTi1+xO5 (x=0.25) have been performed near and below the spin-glass temperature (T g). The effect of a transverse field on the spin-freezing process and spin-glass state has been investigated by measuring the field-and temperature-dependencies of the μSR parameters. Spin-cluster effects signaled by anomalous μ-spin relaxation have been observed in a temperature region just aboveT g. An interpretation supported by recently developed theoretical models addressing non-linear relaxation in an intermediate Griffiths phase is offered.
