    Description: Filifolinone is an aromatic geranyl derivative, a natural compound isolated from Heliotropum sclerocarpum, which has immunomodulatory effects on Atlantic salmon, upregulating cytokines involved in Th1-type responses through a mechanism that remains unknown. In this work, we determined whether the immunomodulatory effects of filifolinone depend on the host microbiotic composition. We evaluated the effect of filifolinone on immune genes and intestinal microbiotic composition of normal fish and fish previously treated with bacitracin/neomycin. Filifolinone induced the early expression of IFN-α1 and TGF-β, followed by the induction of TNF-α, IL-1β, and IFN-γ. A pre-treatment with antibiotics modified this effect, mainly changing the expression of IL-1β and IFN-γ. The evaluation of microbial diversity shows that filifolinone modifies the composition of intestinal microbiota, increasing the abundance of immunostimulating organisms like yeast and firmicutes. We identified 69 operational taxonomic units (OTUs) associated with filifolinone-induced IFN-γ. Our results indicate that filifolinone stimulates the immune system in two ways, one dependent on fish microbiota and the other not. To our knowledge, this is the first report of microbiota-dependent immunostimulation in Salmonids.

    Description: The proper estimation of above-ground biomass (AGB) stocks of managed forests is a prerequisite to quantifying their role in climate change mitigation. The aim of this study was to analyze the spatial variability of AGB and its uncertainty between actively managed pine and unmanaged pine-oak reference forests in central Mexico. To investigate the determinants of AGB, we analyzed variables related to forest management, stand structure, topography, and climate. We developed linear (LM), generalized additive (GAM), and Random Forest (RF) empirical models to derive spatially explicit estimates and their uncertainty, and compared them. AGB was strongly influenced by forest management, as LiDAR-derived stand structure and stand age explained 80.9% to 89.8% of its spatial variability. The spatial heterogeneity of AGB varied positively with stand structural complexity and age in the managed forests. The type of predictive model had an impact on estimates of total AGB in our study site, which varied by as much as 19%. AGB densities varied from 0 to 492 ± 17 Mg ha−1 and the highest values were predicted by GAM. Uncertainty was not spatially homogeneously distributed and was higher with higher AGB values. Spatially explicit AGB estimates and their association with management and other variables in the study site can assist forest managers in planning thinning and harvesting schedules that would maximize carbon stocks on the landscape while continuing to provide timber and other ecosystem services. Our study represents an advancement toward the development of efficient strategies to spatially estimate AGB stocks and their uncertainty, as the GAM approach was used for the first time with improved results for such a purpose.

    Description: The metabolic activity of water-limited ecosystems is strongly linked to the timing and magnitude of precipitation pulses that can trigger disproportionately high (i.e., hot-moments) ecosystem CO2 fluxes. We analyzed over 2-years of continuous measurements of soil CO2 efflux (Fs) under vegetation (Fsveg) and at bare soil (Fsbare) in a water-limited grassland. The continuous wavelet transform was used to: (a) describe the temporal variability of Fs; (b) test the performance of empirical models ranging in complexity; and (c) identify hot-moments of Fs. We used partial wavelet coherence (PWC) analysis to test the temporal correlation between Fs with temperature and soil moisture. The PWC analysis provided evidence that soil moisture overshadows the influence of soil temperature for Fs in this water limited ecosystem. Precipitation pulses triggered hot-moments that increased Fsveg (up to 9000%) and Fsbare (up to 17,000%) with respect to pre-pulse rates. Highly parameterized empirical models (using support vector machine (SVM) or an 8-day moving window) are good approaches for representing the daily temporal variability of Fs, but SVM is a promising approach to represent high temporal variability of Fs (i.e., hourly estimates). Our results have implications for the representation of hot-moments of ecosystem CO2 fluxes in these globally distributed ecosystems.

    Description: Soil moisture plays a key role in the Earth’s water and carbon cycles, but acquisition of continuous (i.e., gap-free) soil moisture measurements across large regions is a challenging task due to limitations of currently available point measurements. Satellites offer critical information for soil moisture over large areas on a regular basis (e.g., European Space Agency Climate Change Initiative (ESA CCI), National Aeronautics and Space Administration Soil Moisture Active Passive (NASA SMAP)); however, there are regions where satellite-derived soil moisture cannot be estimated because of certain conditions such as high canopy density, frozen soil, or extremely dry soil. We compared and tested three approaches, ordinary kriging (OK), regression kriging (RK), and generalized linear models (GLMs), to model soil moisture and fill spatial data gaps from the ESA CCI product version 4.5 from January 2000 to September 2012, over a region of 465,777 km2 across the Midwest of the USA. We tested our proposed methods to fill gaps in the original ESA CCI product and two data subsets, removing 25% and 50% of the initially available valid pixels. We found a significant correlation (r = 0.558, RMSE = 0.069 m3m−3) between the original satellite-derived soil moisture product with ground-truth data from the North American Soil Moisture Database (NASMD). Predicted soil moisture using OK also had significant correlation with NASMD data when using 100% (r = 0.579, RMSE = 0.067 m3m−3), 75% (r = 0.575, RMSE = 0.067 m3m−3), and 50% (r = 0.569, RMSE = 0.067 m3m−3) of available valid pixels for each month of the study period. RK showed comparable values to OK when using different percentages of available valid pixels, 100% (r = 0.582, RMSE = 0.067 m3m−3), 75% (r = 0.582, RMSE = 0.067 m3m−3), and 50% (r = 0.571, RMSE = 0.067 m3m−3). GLM had slightly lower correlation with NASMD data (average r = 0.475, RMSE = 0.070 m3m−3) when using the same subsets of available data (i.e., 100%, 75%, 50%). Our results provide support for using geostatistical approaches (OK and RK) as alternative techniques to gap-fill missing spatial values of satellite-derived soil moisture.
