    Description: Spent nuclear fuel and high-level radioactive waste can be disposed in deep horizontal drillholes in sedimentary, metamorphic or igneous rocks. Horizontal drillhole disposal has safety, operational and economic benefits: the repository is deep in the brine-saturated zone far below aquifers in a reducing environment of formations that can be shown to have been isolated from the surface for exceedingly long times; its depth provides safety against inadvertent intrusion, earthquakes and near-surface perturbations; it can be placed close to the reactors and interim storage facilities, minimizing transportation; disposal costs per ton of waste can be kept substantially lower than for mined repositories by its smaller size, reduced infrastructure needs and staged implementation; and, if desired, the waste could be retrieved using “fishing” technology. In the proposed disposal concept, corrosion-resistant canisters containing unmodified fuel assemblies from commercial reactors would be placed end-to-end in up to 50 cm diameter horizontal drillholes, a configuration that reduces mechanical stresses and keeps the temperatures below the boiling point of the brine. Other high-level wastes, such as capsules containing 137Cs and 90Sr, can be disposed in small-diameter horizontal drillholes. We provide an overview of this novel disposal concept and its technology, discuss some of its safety aspects and compare it to mined repositories and the deep vertical borehole disposal concept.

    Description: Hepatitis C (HCV) is a global pandemic. The World Health Organization has developed a strategic plan for HCV elimination that focuses on low- and middle-income countries (LMICs) and high-risk populations, including people who inject drugs (PWID). While direct-acting antiviral (DAA) therapies are highly effective at eliminating HCV infections and have few side effects, medical professionals and policymakers remain concerned about the risk of reinfection among PWID. This study is a systematic review of research measuring the rate of HCV reinfection among PWID in LMICs and identifies additional areas for further research. A systematic search strategy was used to identify studies documenting HCV reinfection after sustained virologic response in PWID in LMICs. We refined results to include studies where at least 50% of participants had DAA treatment for primary HCV infection. Pooled reinfection rate was calculated across all studies. Seven studies met eligibility criteria. Most studies were conducted in six upper middle-income countries (Mexico, Romania, Russia, Taiwan, Georgi, and Brazil) and one lower middle-income country (Bangladesh) with a total of 7665 participants. No study included information from PWID in low-income countries. Sample sizes ranged from 200 to 3004 individuals, with demographic data missing for most participants. Four studies used deep gene sequencing, and reflex genotyping procedures to differentiate reinfection (infection by a different HCV genotype/subtype) from virologic relapse (infection by the same strain). The follow-up time of people cured from primary chronic HCV infection ranged from 12 weeks to 6.6 years. The pooled reinfection rate of all seven studies was 2.8 (range: 0.02 to 10.5) cases per 100 person-years (PY). In the five studies that differentiated relapse from reinfection, the incidence of reinfection was 1.0 per 100 PY. To date, research on reinfection rates among PWID in LMICs remains limited. Research focused on PWID in low-income countries is particularly needed to inform clinical decision making and evidence-based programs. While rates of reinfection among PWID who complete DAA treatment in upper and lower middle-income countries were similar or lower than rates observed in PWID in high-income countries, the rates were highly variable and factors may influence the accuracy of these measurements. This systematic review identifies several areas for continued research. Policies concerning access to HCV testing and treatment should be comprehensive and not place restrictions on PWID in these settings.

    Description: A shortage of nutrients during the final period of gestation can decrease secretion of colostrum, which is critical to newborn calf survival. The physiological mechanism modulating the nutritional control of colostrum secretion is poorly understood. Because the decline in progesterone before parturition is required for lactogenesis to occur, the objective was to evaluate the correlation between prepartum progesterone in cows and plasma immunoglobulin-G1 (IgG1) concentration in neonatal calves. From 135 pregnant cows, successful data on both prepartum progesterone and calf IgG1 was collected from 59 cow/calf pairs (animal ethics approval SA2018/05/638). The cow/calf pairs were classified into three categories according to the transfer of passive immunity: low (n = 19), medium (n = 18), or high (n = 22). Plasma IgG1 was 1025, 2395, and 3347 mg/dL for the low, medium, and high groups, respectively. Plasma progesterone 1 day prepartum was 18.3, 14.2, and 12.4 nmol/L for the low, medium and high groups, respectively. This indicates that calves with failure of passive transfer were born from cows with higher prepartum progesterone, compared to calves with high IgG1 (P = 0.05). Non-linear modelling of the progesterone data indicated the moment that progesterone started to decline (change-point). Change-point was 0.8, 2.5, and 2.4 days before parturition for the low, medium, and high groups, respectively. There was a delay in progesterone decline in the Low group compared with the High group (P 〈 0.05). These results corroborate the hypothesis that the delay in progesterone decline before parturition is responsible for the failure of IgG1 transfer from cows to calves.

    Description: Chlorophyll fluorescence (ChlF) information offers a deep insight into the plant physiological status by reason of the close relationship it has with the photosynthetic activity. The unmanned aerial systems (UAS)-based assessment of solar induced ChlF (SIF) using non-imaging spectrometers and radiance-based retrieval methods, has the potential to provide spatio-temporal photosynthetic performance information at field scale. The objective of this manuscript is to report the main advances in the development of UAS-based methods for SIF retrieval with non-imaging spectrometers through the latest scientific contributions, some of which are being developed within the frame of the Training on Remote Sensing for Ecosystem Modelling (TRuStEE) program. Investigations from the Universities of Edinburgh (School of Geosciences) and Tasmania (School of Technology, Environments and Design) are first presented, both sharing the principle of the spectroradiometer optical path bifurcation throughout, the so called ‘Piccolo-Doppio’ and ‘AirSIF’ systems, respectively. Furthermore, JB Hyperspectral Devices’ ongoing investigations towards the closest possible characterization of the atmospheric interference suffered by orbital platforms are outlined. The latest approach focuses on the observation of one single ground point across a multiple-kilometer atmosphere vertical column using the high altitude UAS named as AirFloX, mounted on a specifically designed and manufactured fixed wing platform: ‘FloXPlane’. We present technical details and preliminary results obtained from each instrument, a summary of their main characteristics, and finally the remaining challenges and open research questions are addressed. On the basis of the presented findings, the consensus is that SIF can be retrieved from low altitude spectroscopy. However, the UAS-based methods for SIF retrieval still present uncertainties associated with the current sensor characteristics and the spatio-temporal mismatching between aerial and ground measurements, which complicate robust validations. Complementary studies regarding the standardization of calibration methods and the characterization of spectroradiometers and data processing workflows are also required. Moreover, other open research questions such as those related to the implementation of atmospheric correction, bidirectional reflectance distribution function (BRDF) correction, and accurate surface elevation models remain to be addressed.

    Description: The post-closure performance of a generic horizontal drillhole repository for the disposal of spent nuclear fuel (SNF) is quantitatively evaluated using a physics-based numerical model that accounts for coupled thermal-hydrological flow and radionuclide transport processes. The model incorporates most subcomponents of the repository system, from individual waste canisters to the geological far field. The main performance metric is the maximum annual dose to an individual drinking potentially contaminated water taken from a well located above the center of the repository. Safety is evaluated for a wide range of conditions and alternative system evolutions, using deterministic simulations, sensitivity analyses, and a sampling-based uncertainty propagation analysis. These analyses show that the estimated maximum annual dose is low (on the order of 10−4 mSv yr−1, which is 1000 times smaller than a typical dose standard), and that the conclusions drawn from this dose estimate remain valid even if considerable changes are made to key assumptions and property values. The depth of the repository and the attributes of its configuration provide the main safety function of isolation from the accessible environment. Long-term confinement of radionuclides in the waste matrix and slow, diffusion-dominated transport leading to long migration times allow for radioactive decay to occur within the repository system. These preliminary calculations suggest that SNF can be safely disposed in an appropriately sited and carefully constructed and sealed horizontal drillhole repository.

    Description: In vitro fertilization (IVF) involves controlled ovarian hyperstimulation using hormones to produce large numbers of oocytes. The success of IVF is tightly linked to the availability of mature oocytes. In most cases, about 70% to 80% of the oocytes are mature at the time of retrieval, however, in rare instances, all of them may be immature, implying that they were not able to reach the metaphase II (MII) stage. The failure to obtain any mature oocytes, despite a well conducted ovarian stimulation in repeated cycles is a very rare cause of primary female infertility, for which the underlying suspected genetic factors are still largely unknown. In this study, we present the whole exome sequencing analysis of a consanguineous Turkish family comprising three sisters with a recurrent oocyte maturation defect. Analysis of the data reveals a homozygous splice site mutation (c.1775-3C〉A) in the zona pellucida glycoprotein 1 (ZP1) gene. Minigene experiments show that the mutation causes the retention of the intron 11 sequence between exon 11 and exon 12, resulting in a frameshift and the likely production of a truncated protein.

    Description: Land surface temperature (LST) is a fundamental parameter within the system of the Earth’s surface and atmosphere, which can be used to describe the inherent physical processes of energy and water exchange. The need for LST has been increasingly recognised in agriculture, as it affects the growth phases of crops and crop yields. However, challenges in overcoming the large discrepancies between the retrieved LST and ground truth data still exist. Precise LST measurement depends mainly on accurately deriving the surface emissivity, which is very dynamic due to changing states of land cover and plant development. In this study, we present an LST retrieval algorithm for the combined use of multispectral optical and thermal UAV images, which has been optimised for operational applications in agriculture to map the heterogeneous and diverse agricultural crop systems of a research campus in Germany (April 2018). We constrain the emissivity using certain NDVI thresholds to distinguish different land surface types. The algorithm includes atmospheric corrections and environmental thermal emissions to minimise the uncertainties. In the analysis, we emphasise that the omission of crucial meteorological parameters and inaccurately determined emissivities can lead to a considerably underestimated LST; however, if the emissivity is underestimated, the LST can be overestimated. The retrieved LST is validated by reference temperatures from nearby ponds and weather stations. The validation of the thermal measurements indicates a mean absolute error of about 0.5 K. The novelty of the dual sensor system is that it simultaneously captures highly spatially resolved optical and thermal images, in order to construct the precise LST ortho-mosaics required to monitor plant diseases and drought stress and validate airborne and satellite data.

    Description: The ADAM (A Surface Reflectance Database for ESA’s Earth Observation Missions) product (a climatological database coupled to its companion calculation toolkit) enables users to simulate realistic hyperspectral and directional global Earth surface reflectances (i.e., top-of-canopy/bottom-of-atmosphere) over the 240–4000 nm spectral range (at 1-nm resolution) and in any illumination/observation geometry, at 0.1° × 0.1° spatial resolution for a typical year. ADAM aims to support the preparation of optical Earth observation missions as well as the design of operational processing chains for the retrieval of atmospheric parameters by characterizing the expected surface reflectance, accounting for its anisotropy. Firstly, we describe (1) the methods used in the development of the gridded monthly ADAM climatologies (over land surfaces: monthly means of normalized reflectances derived from MODIS observations in seven spectral bands for the year 2005; over oceans: monthly means over the 1999–2009 period of chlorophyll content from SeaWiFS and of wind speed from SeaWinds), and (2) the underlying modeling approaches of ADAM toolkit to simulate the spectro-directional variations of the reflectance depending on the assigned surface type. Secondly, we evaluate ADAM simulation performances over land surfaces. A comparison against POLDER multi-spectral/multi-directional measurements for year 2008 shows reliable simulation results with root mean square differences below 0.027 and R2 values above 0.9 for most of the 14 land cover IGBP classes investigated, with no significant bias identified. Only for the “Snow and ice” class is the performance lower pointing to a limitation of climatological data to represent actual snow properties. An evaluation of the modeled reflectance in the specific backscatter direction against CALIPSO data reveals that ADAM tends to overestimate (underestimate) the so-called “hot-spot” by a factor of about 1.5 (1.5 to 2) for barren (vegetated) surfaces.
