    Description: Lower limb asymmetries have been observed in persons with multiple sclerosis (PwMS), and have been associated with mobility impairment. An incremental cycling test was performed on a cycle ergometer to determine peak power output (PPO) and peak oxygen consumption (VO2peak). Then, participants cycled at 50%, 60%, and 70% of their PPO to assess the contribution of each lower limb to power production. Two-way repeated measures ANOVA was used to detect group × intensity differences in power production asymmetry. Eight PwMS and six healthy individuals (Non-MS) completed the study. No statistically significant (p 〉 0.05) group × intensity interactions or main effects were present when examining between-limb differences in power production. The current data do not indicate a statistically significant difference in power production asymmetry between groups and exercise intensities. Previous research has established a 10% difference between contralateral limbs as a threshold for asymmetry. The average asymmetry in power production in PwMS exceeded the 10% threshold at all measured outputs, suggesting the presence of asymmetry in power production.

    Description: Because studies of rare variant effects on gene expression have limited power, we investigated set-based methods to identify rare expression quantitative trait loci (eQTL) related to Alzheimer disease (AD). Gene-level and pathway-level cis rare-eQTL mapping was performed genome-wide using gene expression data derived from blood donated by 713 Alzheimer’s Disease Neuroimaging Initiative participants and from brain tissues donated by 475 Religious Orders Study/Memory and Aging Project participants. The association of gene or pathway expression with a set of all cis potentially regulatory low-frequency and rare variants within 1 Mb of genes was evaluated using SKAT-O. A total of 65 genes expressed in the brain were significant targets for rare expression single nucleotide polymorphisms (eSNPs) among which 17% (11/65) included established AD genes HLA-DRB1 and HLA-DRB5. In the blood, 307 genes were significant targets for rare eSNPs. In the blood and the brain, GNMT, LDHC, RBPMS2, DUS2, and HP were targets for significant eSNPs. Pathway enrichment analysis revealed significant pathways in the brain (n = 9) and blood (n = 16). Pathways for apoptosis signaling, cholecystokinin receptor (CCKR) signaling, and inflammation mediated by chemokine and cytokine signaling were common to both tissues. Significant rare eQTLs in inflammation pathways included five genes in the blood (ALOX5AP, CXCR2, FPR2, GRB2, IFNAR1) that were previously linked to AD. This study identified several significant gene- and pathway-level rare eQTLs, which further confirmed the importance of the immune system and inflammation in AD and highlighted the advantages of using a set-based eQTL approach for evaluating the effect of low-frequency and rare variants on gene expression.

    Description: The purpose of the current investigation was to examine the effects of exercise intensity and a participant’s cycling experience on asymmetry in pedal forces during cycling. Participants were classified as cycling experienced (CE) or non-cycling experienced (NCE) based on self-reported training history. Participants completed an incremental cycling test via a cycle ergometer with inspired and expired gases, capillary blood lactate and pedaling forces collected throughout the test. Group X exercise intensity comparisons were analyzed at workloads corresponding to 2 mmol/L and 4 mmol/L for the blood lactate accumulation and peak power output, respectively. No Group X exercise intensity interactions for any variables (p 〉 0.05) were observed. The main effect on the exercise intensity was observed for absolute (p = 0.000, η2 = 0.836) and relative (p = 0.000, η2 = 0.752) power outputs and pedal force effectiveness (PFE) (p = 0.000, η2 = 0.728). The main effect for the group was observed for absolute (p = 0.007, η2 = 0.326) and relative (p = 0.001, η2 = 0.433) power outputs, the absolute difference between the lower limbs in power production (p = 0.047, η2 = 0.191), the peak crank torque asymmetry index (p = 0.031, η2 = 0.222) and the PFE (p = 0.014, η2 = 0.280). The exercise intensity was observed to have no impact on asymmetry in pedaling forces during cycling.
