    Beschreibung: Silicon nanotubes (SiNTs) with unique well-defined structural morphologies have been successfully fabricated and recognized as a novel architecture in the nanoscale Si family. While the typical dendritic microstructure of mesoporous silicon prepared anodically has been exploited previously for therapeutics and biosensing, our status of utilizing SiNTs in this regard is still in its infancy. In this review, we focus on the fundamental properties of such nanotubes relevant to therapeutic applications, beginning with a description of our ability to sensitively tune the structure of a given SiNT through synthetic control and the associated detailed in vitro dissolution behavior (reflecting biodegradability). Emphasis is also placed here on the range of functional moieties available to attach to the surface of SiNTs through a summary of current studies involving surface functionalization and strategies that facilitate conjugation with molecules of interest for multiple purposes, including cell labeling, nucleotide attachment, and scaffolding of therapeutic metallic nanoparticles. Experiments addressing our ability to load the interior of a given nanotube with species capable of providing magnetic field-assisted drug delivery are also briefly described. Given the range of diverse properties demonstrated to date, we believe the future to be quite promising for employing SiNTs as therapeutic platforms.

    Beschreibung: Invasive predatory lake trout Salvelinus namaycush were discovered in Yellowstone Lake in 1994 and caused a precipitous decrease in abundance of native Yellowstone cutthroat trout Oncorhynchus clarkii bouvieri. Suppression efforts (primarily gillnetting) initiated in 1995 did not curtail lake trout population growth or lakewide expansion. An adaptive management strategy was developed in 2010 that specified desired conditions indicative of ecosystem recovery. Population modeling was used to estimate effects of suppression efforts on the lake trout and establish effort benchmarks to achieve negative population growth (λ 〈 1). Partnerships enhanced funding support, and a scientific review panel provided guidance to increase suppression gillnetting effort to 〉46,800 100-m net nights; this effort level was achieved in 2012 and led to a reduction in lake trout biomass. Total lake trout biomass declined from 432,017 kg in 2012 to 196,675 kg in 2019, primarily because of a 79% reduction in adults. Total abundance declined from 925,208 in 2012 to 673,983 in 2019 but was highly variable because of recruitment of age-2 fish. Overall, 3.35 million lake trout were killed by suppression efforts from 1995 to 2019. Cutthroat trout abundance remained below target levels, but relative condition increased, large individuals (〉 400 mm) became more abundant, and individual weights doubled, probably because of reduced density. Continued actions to suppress lake trout will facilitate further recovery of the cutthroat trout population and integrity of the Yellowstone Lake ecosystem.

    Beschreibung: Facilitating voluntary conservation on private lands is a crucial element of policies that seek to mitigate forest habitat loss and fragmentation around the world. Previous research emphasizes the role of social factors (e.g., landowner characteristics, economics) in forest management, but environmental outcomes of past management can also affect landowner decisions. Our objective was to evaluate how positive outcomes for wildlife and habitat might reinforce or amplify landowner efforts to manage forest habitats. We applied the lens of coupled human and natural systems to investigate private lands management for early successional forests, which are declining along with associated wildlife in rural areas of the eastern U.S. Efforts to restore early successional forest in this region involve active forest management to create patches of successional forest in native, mature mixed hardwood stands. By integrating field-based monitoring of wildlife with surveys of landowner perceptions, we examined how landowners observed, interpreted, and responded to property-scale ecological outcomes of forest management. We recorded presence of Golden-winged Warbler (Vermivora chrysoptera) and American Woodcock (Scolopax minor) and estimated bird species richness in spring 2015 and/or 2016 on private properties located in the Appalachians (Maryland, New Jersey, Pennsylvania) and Upper Great Lakes (Minnesota, Wisconsin). These properties were enrolled in early successional forest management programs administered through the Natural Resources Conservation Service. Bird surveys were paired with landowner responses to a telephone survey conducted from January to May 2017 (n = 102). Most (71.6–81.6%) landowners’ perceptions of avian presence on their properties matched monitoring results. These perceptions were informed by personal observations and by outreach from agency partners and field technicians. Landowners who already completed their conservation program contracts (n = 85) continued managing early successional forests. Continued management for early successional habitat was positively associated with perceived benefits to birds, forest health, and scenery. Our findings give insight into how private landowners respond to environmental effects of forest management. We conclude that positive environmental outcomes of these conservation programs are related to continued early successional forest conservation by private landowners.

    Beschreibung: Pedagogical tools are needed that link multidisciplinary nanoscience and technology (NST) to multiple state-of-the-art applications, including those requiring new fabrication routes relying on green synthesis. These can both educate and motivate the next generation of entrepreneurial NST scientists to create innovative products whilst protecting the environment and resources. Nanoporous silicon shows promise as such a tool as it can be fabricated from plants and waste materials, but also embodies many key educational concepts and key industrial uses identified for NST. Specific mechanical, thermal, and optical properties become highly tunable through nanoporosity. We also describe exceptional properties for nanostructured silicon like medical biodegradability and efficient light emission that open up new functionality for this semiconductor. Examples of prior lecture courses and potential laboratory projects are provided, based on the author’s experiences in academic chemistry and physics departments in the USA and UK, together with industrial R&D in the medical, food, and consumer-care sectors. Nanoporous silicon-based lessons that engage students in the basics of entrepreneurship can also readily be identified, including idea generation, intellectual property, and clinical translation of nanomaterial products.

    Beschreibung: The purpose of this study was (1) to determine the effect of single bouts of volume- and intensity-equated low- (LL) and high-load (HL) full-body resistance exercise (RE) on AR-DNA binding, serum/muscle testosterone and dihydrotestosterone, muscle androgen receptor (AR), and AR-DNA binding; and, (2) to determine the effect of RE on sarcoplasmic and nucleoplasmic β-catenin concentrations in order to determine their impact on mediating AR-DNA binding in the absence/presence of serum/muscle androgen and AR protein. In a cross-over design, 10 resistance-trained males completed volume- and intensity-equated LL and HL full-body RE. Blood and muscle samples were collected at pre-, 3 h-, and 24 h post-exercise. Separate 2 × 3 factorial analyses of variance (ANOVAs) with repeated measures and pairwise comparisons with a Bonferroni adjustment were used to analyze the main effects. No significant differences were observed in muscle AR, testosterone, dihydrotestosterone, or serum total testosterone in either condition (p 〉 0.05). Serum-free testosterone was significantly decreased 3 h post-exercise and remained significantly less than baseline 24 h post-exercise in both conditions (p 〈 0.05). In response to HL, AR-DNA binding significantly increased at 3 h post-exercise (p 〈 0.05), whereas no significant differences were observed at any time in response to LL (p 〉 0.05). Moreover, sarcoplasmic β-catenin was significantly greater in HL (p 〈 0.05) without significant changes in nucleoplasmic β-catenin (p 〉 0.05). In conclusion, increases in AR-DNA binding in response to HL RE indicate AR signaling may be load-dependent. Furthermore, despite the lack of increase in serum and muscle androgens or AR content following HL RE, elevations in AR-DNA binding with elevated sarcoplasmic β-catenin suggests β-catenin may be facilitating this response.

    Beschreibung: Cisplatin is a chemotherapy agent commonly used to treat a wide variety of cancers. Despite the potential for both severe acute and chronic side effects, it remains a preferred therapeutic option for many malignancies due to its potent anti-tumor activity. Common cisplatin-associated side-effects include acute kidney injury (AKI) and chronic kidney disease (CKD). These renal injuries may cause delays and potentially cessation of cisplatin therapy and have long-term effects on renal function reserve. Thus, developing mechanism-based interventional strategies that minimize cisplatin-associated kidney injury without reducing efficacy would be of great benefit. In addition to its action of cross-linking DNA, cisplatin has been shown to affect mitochondrial metabolism, resulting in mitochondrially derived reactive oxygen species (ROS). Increased ROS formation in renal proximal convoluted tubule cells is associated with cisplatin-induced AKI and CKD. We review the mechanisms by which cisplatin may induce AKI and CKD and discuss the potential of mitochondrial superoxide dismutase mimetics to prevent platinum-associated nephrotoxicity.

    Beschreibung: Background: EPA and DHA n-3 FA play crucial roles in both neurological and cardiovascular health and high dietary intakes along with supplementation suggest potential neuroprotection and concussion recovery support. Rugby athletes have a high risk of repetitive sub-concussive head impacts which may lead to long-term neurological deficits, but there is a lack of research looking into n-3 FA status in rugby players. We examined the dietary n-3 FA intake through a FFQ and n-3 FA status by measuring the percentage of n-3 FA and O3I in elite Canadian Rugby 7s players to show distribution across O3I risk zones; high risk, 8%. Methods: n-3 FA profile and dietary intake as per FFQ were collected at the beginning of the 2017–2018 Rugby 7s season in male (n = 19; 24.84 ± 2.32 years; 95.23 ± 6.93 kg) and female (n = 15; 23.45 ± 3.10 years; 71.21 ± 5.79 kg) athletes. Results: O3I averaged 4.54% ± 1.77, with female athlete scores slightly higher, and higher O3I scores in supplemented athletes (4.82% vs. 3.94%, p = 0.183), with a greater proportion of non-supplemented athletes in the high-risk category (45.5% vs. 39.1%). Dietary intake in non-supplemented athletes did not meet daily dietary recommendations for ALA or EPA + DHA compared to supplemented athletes. Conclusions: Overall, despite supplementation, O3I score remained in the high-risk category in a proportion of athletes who met recommended n-3 FA dietary intakes, and non-supplemented athletes had a higher proportion of O3I scores in the high-risk category, suggesting that dietary intake alone may not be enough and athletes may require additional dietary and n-3 FA supplementation to reduce neurological and cardiovascular risk.

    Beschreibung: Public lands alone are insufficient to address the needs of most at-risk wildlife species in the U.S. As a result, a variety of voluntary incentive programs have emerged to recruit private landowners into conservation efforts that restore and manage the habitats needed by specific species. We review the role of one such effort, Working Lands for Wildlife (WLFW), initiated by the Natural Resources Conservation Service in partnership with the U.S. Fish and Wildlife Service. Using two at-risk species in the eastern U.S. (where private lands dominate), we show the substantial potential that WLFW has for restoring and maintaining needed habitats. Monitoring how effective these efforts are on populations of the target species has been challenging, and both monitoring and implementation are being modified in response to new information. Identifying landowner motivations is essential for developing long-term relationships and conservation success. As WLFW projects develop, they are moving toward a more holistic ecosystem approach, within which the conservation goals of at-risk species are embedded.
