    Description: The present experiment was carried out to assess the effects of reconstituted milk (RM), acidified reconstituted milk (ARM), and acidified fresh milk (AFM) on growth performance, diarrhea rate, and hematological parameters of preweaning dairy calves. For this purpose, a total of 27 Holstein female calves (one month of age) with initial body weight of (67.46 ± 4.08) kg were divided into three groups in such a way that each group contained nine calves. Calves were housed individually, and starter was offered ad libitum to each calf. The dietary treatments were RM, ARM, and AFM. The highest milk intake was observed in calves receiving AFM as compared to other treatments (p 〈 0.01). Calves fed AFM had more feed intake than those fed ARM and RM (p 〈 0.01). Feed efficiency was significantly lower for calves offered ARM than those offered RM and AFM (p 〈 0.01). A lower withers height growth was found for calves fed RM than those fed ARM and AFM (p

    Description: The profile distributions of soil organic carbon (SOC), soil organic nitrogen (SON), soil pH and soil texture were rarely investigated in the Lancangjiang River Basin. This study aims to present the vertical distributions of these soil properties and provide some insights about how they interact with each other in the two typical soil profiles. A total of 56 soil samples were collected from two soil profiles (LCJ S-1, LCJ S-2) in the Lancangjiang River Basin to analyze the profile distributions of SOC and SON and to determine the effects of soil pH and soil texture. Generally, the contents of SOC and SON decreased with increasing soil depth and SOC contents were higher than SON contents (average SOC vs. SON content: 3.87 g kg−1 vs. 1.92 g kg−1 in LCJ S-1 and 5.19 g kg−1 vs. 0.96 g kg−1 in LCJ S-2). Soil pH ranged from 4.50 to 5.74 in the two soil profiles and generally increased with increasing soil depth. According to the percentages of clay, silt, and sand, most soil samples can be categorized as silty loam. Soil pH values were negatively correlated with C/N ratios (r = −0.66, p 〈 0.01) and SOC contents (r = −0.52, p 〈 0.01). Clay contents were positively correlated with C/N ratios (r = 0.43, p 〈 0.05) and SOC contents (r = 0.42, p 〈 0.01). The results indicate that soil pH and clay are essential factors influencing the SOC spatial distributions in the two soil profiles.

    Description: : The frequency and intensity of heat waves (HWs) has increased in subtropical regions in recent years. The mechanism underlying the HW response of subtropical trees remains unclear. In this study, we conducted an experiment with broad-leaved Schima superba (S. superba) and coniferous Cunninghamia lanceolata (C. lanceolata) seedlings to examine HW (5-day long) effects on stem water transport, leaf water use efficiency (WUE), morphology and growth, and to elucidate differences in the responses of both species. Our results indicated that HWs can significantly reduce hydraulic conductivity in both species. C. lanceolata experienced significant xylem embolism, with the percentage loss of conductivity (PLC) increasing by 40%, while S. superba showed a non-significant increase in PLC (+25%). Furthermore, HW also caused a reduction in photosynthesis rates (An), but transpiration rates (Tr) increased on the 5th day of the HW, together leading to a significant decrease in leaf WUE. From diurnal dynamics, we observed that the HW caused significant decrease of S. superba An only in the morning, but nearly the all day for C. lanceolata. During the morning, with a high vapor pressure deficit (VPD) environment, the HW increased Tr, which contributed a lot to latently cooling the foliage. In comparing the two tree species, we found that HW effects on S. superba were mostly short-term, with leaf senescence but limited or no xylem embolism. The surviving S. superba recovered rapidly, forming new branches and leaves, aided by their extensive root systems. For C. lanceolata, continued seedling growth initially but with subsequent xylem embolism and withering of shoots, led to stunted recovery and regrowth. In conclusion, apart from the direct thermal impacts caused by HW, drought stress was the main cause of significant negative effects on plant water transport and the photosynthetic system. Furthermore, S. superba and C. lanceolata showed clearly different responses to HW, which implies that the response mechanisms of broad-leaved and coniferous tree species to climate change can differ.

    Description: The Chinese project, better known as the Grain for Green Project (GGP), has changed the land-use type in the karst area of Puding county, Guizhou province, southwest China, and this study is aimed at evaluating the Hg distribution and determining factors in soils after the land-use change. A total of ten soil profiles were selected in the typical karst region, and the land-use types were divided into native vegetation land (NVL), farmland (FL), and abandoned farmland (AFL). Total Hg concentration under different land-use types increased in the order: NVL (average 63.26 μg∙kg−1) 〈 FL (average 71.48 μg∙kg−1) 〈 AFL (average 98.22 μg∙kg−1). After agricultural abandonment for four to five years with a cover of native vegetation in the AFL, a higher concentration of Hg compared to the other two land-use types indicate that the Hg accumulation in soil results from vegetation restoration of AFL due to land-use change. Soil organic carbon (SOC) and macro-aggregates were highly correlated to Hg concentration in this study. Macro-aggregates can provide a stable condition for Hg due to the thin regolith and high porosity in the karst region. A high proportion of macro-aggregates can reduce the mobility of Hg in the karst area. Intense tillage can significantly reduce the formation of macro-aggregates in FL, but the macro-aggregates in AFL were recovered as well as those in NVL, resulting in the accumulation of Hg.

    Description: The effects of flavonoids and copper (Cu) on metabolic syndrome (MetS) have been investigated separately, but no information exists about the joint associations between flavonoids and Cu on the risk of MetS in population studies. In this cross-sectional study, a total of 9108 people aged 20–75 years from the Harbin Cohort Study on Diet, Nutrition, and Chronic Non-Communicable Diseases (HDNNCDS) were included. Flavonoid intakes were calculated based on the flavonoid database created in our laboratory. Cu and other nutrient intakes were estimated using the Chinese Food Composition Table. Among all study subjects, a total of 2635 subjects (28.9%) met the diagnostic criteria for inclusion in the MetS group. Total flavonoids (fourth vs. first quartile, odds ratio (OR): 0.77, 95% confidence interval (CI) 0.66–0.90, Ptrend = 0.002) and Cu (OR 0.81, 90% CI: 0.70–0.94, Ptrend = 0.020) were inversely associated with the risk of MetS after adjusting for potential confounders. Higher flavonoid intake was more strongly associated with a lower risk of MetS with high levels of Cu intake (Pinteraction = 0.008). Dose–response effects showed an L-shaped curve between the total intake of five flavonoids and the risk of MetS. These results suggest that higher flavonoid intake is associated with a lower risk of MetS, especially under high levels of Cu intake.

    Description: Oral administration of pathogen-specific recombinant antibodies may help to prevent infant gastrointestinal (GI) pathogen infection; however, to neutralize an infectious agent, these antibodies must resist degradation in the GI tract. Palivizumab, a recombinant antibody specific for the respiratory syncytial virus (RSV), was used as a model for pathogen-specific IgG in human milk. The aim was to compare the remaining binding capacity of palivizumab in milk between three mothers after exposure to an in vitro model of infant gastrointestinal digestion (gastric and duodenal fluids) using ELISA. The neutralizing capacity of palivizumab in pooled human milk, gastric contents, and stools from preterm infants was also evaluated for blocking RSV with green fluorescent protein (RSV-GFP) infection in Hep-2 cells using confocal and inverted microscopy and flow cytometry. The reduction of palivizumab binding capacity in human milk and digested samples was slightly different between mothers. Overall, palivizumab decreased 50% after simulated gastric digestion with pepsin and 62% after simulated intestinal digestion with pancreatin. Palivizumab (2–8 μg/mL) in human milk or stool samples blocked RSV (3.4 × 104 FFU/mL) infection (no syncytia formation on Hep-2 cells) by microscopy. Syncytia formation was detected on Hep-2 cells when RSV was incubated in gastric contents or virus medium with 2–4 μg/mL of palivizumab, but no infection was observed at 8 μg/mL. No fluorescence (absence of infected cells) was detected when palivizumab (100 μg/mL) was incubated in human milk or medium with RSV-GFP (1.1 × 105 FFU/mL), whereas fluorescence increased with the reduced concentration of palivizumab using flow cytometry. These results suggest that undigested and digested matrices could change the binding and neutralizing capacity of viral pathogen-specific antibodies.

    Description: Based on the previous GWAS research related to bovine heat tolerance trait, this study aimed to explore the effect of myosin-1a (MYO1A) gene on bovine heat tolerance trait, and find the molecular markers related to the heat tolerance of Chinese cattle. In our study, four novel candidate SNPs highly conserved in B. indicus breeds but barely existed in B. taurus were identified in MYO1A gene according to Bovine Genome Variation Database and Selective Signatures (BGVD). PCR and DNA sequencing were used to genotype 1072 individuals including 34 Chinese indigenous cattle breeds as well as Angus and Indian zebu. Two synonymous mutations (rs208210464 and rs110123931), one missense mutation (rs209999142; Phe172Ser), and one intron mutation (rs135771836) were detected. The frequencies of mutant alleles of the four SNPs gradually increased from northern groups to southern groups of Chinese cattle, which was consistent with the distribution of various climatic conditions of China. Additionally, four SNPs were significantly associated with four climatic conditions including annual mean temperature (T), relative humidity (H), temperature-humidity index (THI), and average annual sunshine hours (100-cloudiness) (SR). Among these, rs209999142 and Hap 1/1 had better performance than others. Our results suggested that rs209999142 was associated with heat-tolerance trait and rs208210464, rs110123931, and rs135771836 showed high phenotypic effect on heat-tolerance trait because of the strong linkage with rs209999142. These SNPs could be used as candidates for marker-assisted selection (MAS) in cattle breeding.

    Description: The study investigated the effects of dietary zeolite supplementation as an antibiotic alternative on growth performance, intestinal integrity, and cecal antibiotic resistance genes abundances of broilers. One-day-old chicks were assigned into three groups and fed a basal diet or a basal diet supplemented with antibiotics (50 mg/kg) or zeolite (10 g/kg). Antibiotic or zeolite increased (p 〈 0.05) average daily gain (ADG) from 1 to 42 days and duodenal villus height to crypt depth ratio (VH:CD) at 21 days. Zeolite increased (p 〈 0.05) ADG and average daily feed intake from 1 to 21 days, jejunal VH:CD at 21 and 42 days, ileal VH and VH:CD at 42 days, zonula occludens-1 mRNA abundance at 21 days, and duodenal occludin mRNA abundance at 42 days, whereas reduced (p 〈 0.05) jejunal CD and malondialdehyde levels in ileum at 21 days and duodenum at 42 days, serum D-lactic acid and diamine oxidase levels at 42 days, and plasma lipopolysaccharide content at 21 and 42 days. Antibiotics reduced (p 〈 0.05) duodenal claudin-2 mRNA abundance at 21 days, whereas increased (p 〈 0.05) cecal tetB abundance at 42 days. These findings suggested that the beneficial effects of zeolite in broilers were more pronounced than that of antibiotics.

    Description: EGF acts as a ligand of the EGF receptor (EGFR) to activate the EGFR-mediated signaling pathways and is involved in the regulation of cell physiology. However, the roles of EGFR mediated signaling pathways in the regulation of lipid metabolism in goat mammary epithelial cells (GMECs) are poorly understood. To evaluate the impact of EGF on GMECs, the triglyceride (TG) content and lipid droplet were detected, using TG assay and immunofluorescence. Further, expression of lipogenic genes, the protein kinase B (Akt), phospholipase C-γ1 (PLC-γ1) and extracellular signal-regulated kinases (ERK)1/2 signaling pathways were measured by real-time polymerase chain reaction and Western blot, respectively. The results showed that the mRNA expression of EGFR gene was significantly upregulated in lactating goat mammary gland tissues compared to non-lactation period (p 〈 0.05). TG contents in EGF-treated GMECs were significantly increased (p 〈 0.05), and an increase of lipid droplets was also detected. In vitro studies demonstrated that the mRNA levels of lipogenesis-related FASN, ACC, SCD1, LXRa, LXRb and SP1 genes were positively correlated to the mRNA level of EGFR gene shown by gene overexpression and silencing (p 〈 0.05). The phosphorylations of Akt, ERK1/2 and PLC-γ1 in GMECs were greatly upregulated in the presence of EGF, and specific inhibitors were capable of blocking the phosphorylation of Akt, ERK1/2 and PLC-γ1. Compared with EGF-treated GMECs, the mRNA levels of FASN, ACC and SCD1 were significantly decreased in GMECs co-treated with PLC-γ1 and Akt inhibitor and EGF (p 〈 0.05), and TG content was also dropped significantly. These observations implied that EGFR plays an important role in regulating de novo fatty acid synthesis in GMECs, mainly mediated by Akt and PLC-γ1 signaling pathways.
