    Notes: Accurate moulding of polymer foams of density 0.02-0.3 g cm-3 has been achieved by crystallising polyethylene and ethylene/methlacrylate copolymers from dry supercritical solutions. Adherent thick films can be sealed to electronic substrates and are proposed as insulating layers for interconnections on multichip modules and other high-speed electronic devices. The dielectric constant achieved for films of several-mil thickness is between 1.05 and 1.3 and can be tailored along with the physical properties of the film by adjusting the density and composition of the copolymer. Planarisation or complex patterning is possible by detailing the mould.

    Description: Much remains to be discovered about the intersection of tissue-specific transcription control and the epigenetics of skeletal muscle (SkM), a very complex and dynamic organ. From four gene families, Leucine-Rich Repeat Containing (LRRC), Oxysterol Binding Protein Like (OSBPL), Ankyrin Repeat and Socs Box (ASB), and Transmembrane Protein (TMEM), we chose 21 genes that are preferentially expressed in human SkM relative to 52 other tissue types and analyzed relationships between their tissue-specific epigenetics and expression. We also compared their genetics, proteomics, and descriptions in the literature. For this study, we identified genes with little or no previous descriptions of SkM functionality (ASB4, ASB8, ASB10, ASB12, ASB16, LRRC14B, LRRC20, LRRC30, TMEM52, TMEM233, OSBPL6/ORP6, and OSBPL11/ORP11) and included genes whose SkM functions had been previously addressed (ASB2, ASB5, ASB11, ASB15, LRRC2, LRRC38, LRRC39, TMEM38A/TRIC-A, and TMEM38B/TRIC-B). Some of these genes have associations with SkM or heart disease, cancer, bone disease, or other diseases. Among the transcription-related SkM epigenetic features that we identified were: super-enhancers, promoter DNA hypomethylation, lengthening of constitutive low-methylated promoter regions, and SkM-related enhancers for one gene embedded in a neighboring gene (e.g., ASB8-PFKM, LRRC39-DBT, and LRRC14B-PLEKHG4B gene-pairs). In addition, highly or lowly co-expressed long non-coding RNA (lncRNA) genes probably regulate several of these genes. Our findings give insights into tissue-specific epigenetic patterns and functionality of related genes in a gene family and can elucidate normal and disease-related regulation of gene expression in SkM.

    Description: KLHL and the related KBTBD genes encode components of the Cullin-E3 ubiquitin ligase complex and typically target tissue-specific proteins for degradation, thereby affecting differentiation, homeostasis, metabolism, cell signaling, and the oxidative stress response. Despite their importance in cell function and disease (especially, KLHL40, KLHL41, KBTBD13, KEAP1, and ENC1), previous studies of epigenetic factors that affect transcription were predominantly limited to promoter DNA methylation. Using diverse tissue and cell culture whole-genome profiles, we examined 17 KLHL or KBTBD genes preferentially expressed in skeletal muscle or brain to identify tissue-specific enhancer and promoter chromatin, open chromatin (DNaseI hypersensitivity), and DNA hypomethylation. Sixteen of the 17 genes displayed muscle- or brain-specific enhancer chromatin in their gene bodies, and most exhibited specific intergenic enhancer chromatin as well. Seven genes were embedded in super-enhancers (particularly strong, tissue-specific clusters of enhancers). The enhancer chromatin regions typically displayed foci of DNA hypomethylation at peaks of open chromatin. In addition, we found evidence for an intragenic enhancer in one gene upregulating expression of its neighboring gene, specifically for KLHL40/HHATL and KLHL38/FBXO32 gene pairs. Many KLHL/KBTBD genes had tissue-specific promoter chromatin at their 5′ ends, but surprisingly, two (KBTBD11 and KLHL31) had constitutively unmethylated promoter chromatin in their 3′ exons that overlaps a retrotransposed KLHL gene. Our findings demonstrate the importance of expanding epigenetic analyses beyond the 5′ ends of genes in studies of normal and abnormal gene regulation.

    Notes: The preparation of L-lysine peptides (Lysn, n = 2-14) from polyL-lysine is described. Fractionation by ion-exchange column chromatography of poly-L-lysine hydrolysates on a preparative scale resulted in 0.2-1.0 g quantities of individual members of the poly-L-lysine series. The peptides isolated proved to be analytically pure and the optical configuration was fully retained, as demonstrated by complete enzymic digestion. Peptides higher than n = 14 were also prepared. They consisted of oligolysine groups of narrow and accurately determined size distribution. Potentiometric titrations were used both to characterize the products and to demonstrate the characteristic dependence of the dissociation constants on size of the peptide.

    Notes: The molecular weights and radii of gyration of Streptococcus salivarius levan fractions were obtained from light-scattering measurements in water. Sedimentation coefficients and partial specific volumes of the fractions were also obtained. Double logarithmic plots of [η] versus M̄w and S0 versus M̄w yielded slopes having values of 0.17 and 0.62, respectively. The data and various calculated parameters show that levan from Streptococcus salivarius is highly branched and behaves hydrodynamically as a compact particle of spherical symmetry.

    Notes: Males of Euphydryas editha (Lepidoptera: Nymphalidae) need their antennae to mate successfully, but females do not. Antennal structure was investigated in the hope of explaining this functional dimorphism, which is opposite to that in other butterflies (e.g., Myers, '68; Grula and Taylor, '80). No external differences between the sexes were observed with electron microscopy. There are four types of antennal sensilla: the spine, which acts mainly as a mechanoreceptor, shallow dish hairs and hidden hairs, which are chemoreceptors, and a whiplike sensillum of uncertain function. The internal morphology of male and female antennae differs in several respects which may relate to functional differences. The mating systems of butterflies are discussed briefly to explain our results and those of others.

    Notes: This study examines the morphology of sporadic congenital microphthalmia in 1-day-old chicks, with particular emphasis on the neural retina. On the basis of the size of the eyeball it is possible to classify microphthalmia into two groups, severe and mild. In severe microphthalmia (less than 5 mm in equatorial diameter), the eyeball is severely malformed, but in most cases it shows evidence of an organized neural retina. Although ganglion cells and an optic nerve head are present in a small proportion of these retinae, we could not trace an optic nerve projection to the brain. These results indicate that some ganglion cells are able to be sustained after the period of naturally occurring cell death, suggesting either that those ganglion cells have established some contact with the central nervous system or that the presence of their axons in a rudimentary optic nerve is adequate for survival. In mild microphthalmia (greater than 5 mm in equatorial diameter), the most consistent abnormality is a defect in the pecten, which together with other abnormalities such as orbital cysts and colobomas indicates that the major abnormality occurs in the region of the choroid fissure. Associated with these defects are abnormalities within the ganglion cell layer. In some cases the number of ganglion cells was reduced, and in others the numbers of both ganglion and displaced amacrine cells were reduced. Unexpectedly, there were localized regions completely devoid of cells in the ganglion cell layer. The timing of the congenital defect may provide some clue as to the presence of a critical period in which displaced amacrine cells are formed or are sensitive to events related to ganglion cell loss.

    Notes: The viscosities of moderately concentrated solutions of low-density polyethylenes in ethane, propane, and ethylene have been measured at low shear rate in the temperature range of 150-250°C and in the pressure range of about 15000-30000 psi. Within the precision of the measurements, the relative viscosity is independent of pressure over the range investigated but increases as the solvent is changed from propane through ethane to ethylene. The activation energy for the relative viscosity in ethane varies from about 0.5 to 2.5 kcal/mole as the concentration changes from 5 to 15 g/dl. Effects of polymer concentration and molecular weight on solution viscosity in ethane at 150°C have been determined, and all of the data can be represented by a single straight-line plot of the logarithm of relative viscosity versus the intrinsic viscosity (in p-xylene at 105°C) times concentration. This simple relation is valid over wide ranges of polymer concentration and molecular weight and over more than two orders of magnitude of relative viscosity. The solution viscosities of the polyethylenes in the three supercritical fluid solvents used appear surprisingly low at first sight. This behavior is partly a result of the low solvent viscosities but also might mean that the polymer has an abnormally low segmental friction factor compared to that in solutions under more familiar conditions.
