Description: Many natural materials possess built-in structural variation, endowing them with superior performance. However, it is challenging to realize programmable structural variation in self-assembled synthetic materials since self-assembly processes usually generate uniform and ordered structures. Here, we report the formation of asymmetric microribbons composed of directionally self-assembled two-dimensional nanoflakes in a polymeric matrix during three-dimensional direct-ink printing. The printed ribbons with embedded structural variations show site-specific variance in their mechanical properties. Remarkably, the ribbons can spontaneously transform into ultrastretchable springs with controllable helical architecture upon stimulation. Such springs also exhibit superior nanoscale transport behavior as nanofluidic ionic conductors under even ultralarge tensile strains (〉1,000%). Furthermore, to show possible real-world uses of such materials, we demonstrate in vivo neural recording and stimulation using such springs in a bullfrog animal model. Thus, such springs can be used as neural electrodes compatible with soft and dynamic biological tissues.

Description: The flow of cargo vesicles along the secretory pathway requires concerted action among various regulators. The COPII complex, assembled by the activated SAR1 GTPases on the surface of the endoplasmic reticulum, orchestrates protein interactions to package cargos and generate transport vesicles en route to the Golgi. The dynamic nature of COPII, however, hinders analysis with conventional biochemical assays. Here we apply proximity-dependent biotinylation labeling to capture the dynamics of COPII transport in cells. When SAR1B was fused with a promiscuous biotin ligase, BirA*, the fusion protein SAR1B-BirA* biotinylates and thus enables the capture of COPII machinery and cargos in a GTP-dependent manner. Biochemical and pulse–chase imaging experiments demonstrate that the COPII coat undergoes a dynamic cycle of engagement–disengagement with the transmembrane cargo receptor LMAN1/ERGIC53. LMAN1 undergoes a process of concentrative sorting by the COPII coat, via a dimeric sorting code generated by oligomerization of the cargo receptor. Similar oligomerization events have been observed with other COPII sorting signals, suggesting that dimeric/multimeric sorting codes may serve as a general mechanism to generate selectivity of cargo sorting.

Description: The cytoplasmic coat protein complex-II (COPII) is evolutionarily conserved machinery that is essential for efficient trafficking of protein and lipid cargos. How the COPII machinery is regulated to meet the metabolic demand in response to alterations of the nutritional state remains largely unexplored, however. Here, we show that dynamic changes of COPII vesicle trafficking parallel the activation of transcription factor X-box binding protein 1 (XBP1s), a critical transcription factor in handling cellular endoplasmic reticulum (ER) stress in both live cells and mouse livers upon physiological fluctuations of nutrient availability. Using live-cell imaging approaches, we demonstrate that XBP1s is sufficient to promote COPII-dependent trafficking, mediating the nutrient stimulatory effects. Chromatin immunoprecipitation (ChIP) coupled with high-throughput DNA sequencing (ChIP-seq) and RNA-sequencing analyses reveal that nutritional signals induce dynamic XBP1s occupancy of promoters of COPII traffic-related genes, thereby driving the COPII-mediated trafficking process. Liver-specific disruption of the inositol-requiring enzyme 1α (IRE1α)–XBP1s signaling branch results in diminished COPII vesicle trafficking. Reactivation of XBP1s in mice lacking hepatic IRE1α restores COPII-mediated lipoprotein secretion and reverses the fatty liver and hypolipidemia phenotypes. Thus, our results demonstrate a previously unappreciated mechanism in the metabolic control of liver protein and lipid trafficking: The IRE1α-XBP1s axis functions as a nutrient-sensing regulatory nexus that integrates nutritional states and the COPII vesicle trafficking.

Description: To assess different impacts of land, population and economy factors on the lexicographic minimax optimal allocation of blue and green water footprints, a comprehensive discriminant rule is constructed in this paper based on the Gini coefficient and Theil entropy index. The proposed rule is employed to estimate the influence of the aforesaid factors (land, population and economy) on the corresponding allocation schemes from a fairness perspective. To demonstrate its applicability, the proposed approach is applied to a water resources allocation study for 11 provinces in the Yangtze River Economic Belt (YREB). The results indicate that: (1) the economy-based lexicographic allocation of water footprints (LAWF) is more equalitarian for the provinces with high water footprint quotas. The land area-based LAWF is more equalitarian for the provinces with low water footprint quotas. The population-based LAWF is more equalitarian for the provinces with medium water footprint quotas. (2) The contribution of intra-regional variation in the population-based LAWF scheme is the largest of the three schemes. The inter-regional variation contributed the largest in the land area-based LAWF scheme. (3) Two synthetic schemes which integrate multiple factors among land area, economy and population are more equalitarian than the three single-factor schemes. Compared with the original situation which is an equalitarian but ineffective allocation, the two synthetic schemes have greater effect on the improvement of the supply-demand balance of water resources carrying capacity. Therefore, the defect of the population, economy and land area factors acting alone should be resolved by designing a weighting system, in order to optimize the allocation of water resources.

Description: Understanding and improving public flood risk perception is conducive to the implementation of effective flood risk management and disaster reduction policies. In the flood-prone city of Jingdezhen, flood disaster is one of the most destructive natural hazards to impact the society and economy. However, few studies have been attempted to focus on public flood risk perception in the small and medium-size city in China, like Jingdezhen. Therefore, the purpose of this study was to investigate the public flood risk perception in four districts of Jingdezhen and examine the related influencing factors. A questionnaire survey of 719 randomly sampled respondents was conducted in 16 subdistricts of Jingdezhen. Analysis of variance was conducted to identify the correlations between the impact factors and public flood risk perception. Then, the flood risk perception differences between different groups under the same impact factor were compared. The results indicated that the socio-demographic characteristics of the respondents (except occupation), flood experience, flood knowledge education, flood protection responsibility, and trust in government were strongly correlated with flood risk perception. The findings will help decision makers to develop effective flood risk communication strategies and flood risk reduction policies.

Description: Project procurement method (PPM) selection influences the efficiency of project implementation. Owners are presented with different options for project delivery. However, selecting the appropriate PPM poses great challenges to owners, given the existence of ambiguous information. The interval neutrosophic set (INS) shows power to handle imprecise and ambiguous information. This paper aims to develop a PPM selection model under an interval neutrosophic environment for owners. The main contributions of this paper are as follows: (1) The similarity measure is innovatively introduced with interval neutrosophic information to handle the PPM selection problem. (2) The similarity measure based on minimum and maximum operators is applied to construct a decision-making model for PPM selection, through considering the truth, falsity, and indeterminacy memberships simultaneously. (3) This study establishes a PPM selection method with INS by applying similarity measures, that takes account into the determinacy, indeterminacy, and hesitation from the decision experts when giving an evaluation value. A case study on selecting PPM is made to show the applicability of the proposed approach. Finally, the results of the proposed method are compared with those of existing methods, which exhibit the superiority of the proposed PPM selection method.
