    Description: Sulfatide (HSO 3 -3-galactosylceramide), which enriched in lipid rafts of plasma membranes in various epithelial cell lines, is a critical component of host cells for effective production of influenza A virus. However, the function of sulfatide in other virus infections targeting epithelial cells remains unknown. In this study, the effect of sulfatide on infection of human parainfluenza virus type 3 (hPIV3) was demonstrated by using genetically produced sulfatide-enriched cells and by treatment of hPIV3-infected cells with anti-sulfatide monoclonal antibody (GS-5) as well as by addition of sulfatide to the cells. hPIV3 was found to bind to sulfatide in a virus overlay assay and a solid-phase binding assay. Genetic expression of sulfatide in COS-7 cells defective in sulfatide suppressed initial hPIV3 infection and formation of multinucleate virus-infected cells. Treatment of virus-infected LLC-MK2 cells with GS-5 promoted formation of multinucleate cells. In contrast, exogenous addition of sulfatide to hPIV3-infected COS-7 cells and cells expressing the hPIV3- hemagglutinin-neuraminidase ( HN ) gene and fusion ( F ) gene conspicuously reduced the formation of multinucleate cells. The results suggest that sulfatide negatively regulates the fusion process of hPIV3, possibly through interaction with HN or F glycoprotein on the cell surface.

    Description: Late Oligocene to Middle Miocene adakitic andesites are found in the southern part of Okushiri Island, the northern Noto Peninsula and in the Toyama region in the present-day back-arc margin of the SW and NE Japan arcs. On Okushiri Island, adakitic andesite is accompanied by moderately alkaline basalt, whereas on the Noto Peninsula, adakitic andesite has been erupted along with high magnesian andesite (HMA), bronzite andesite and tholeiitic basalt. Adakitic andesites from all three locations are characterized by high Sr/Y and low Y, and have higher MgO contents than adakitic melts generated by experimental melting of metabasalt and amphibolite. They also have higher Ni and Cr contents than either Archaean tonalite–trondhjemite–granodiorite (TTG) suites or Early Cretaceous adakitic granites, which have been attributed to partial melting of subducted oceanic crust. The Noto Peninsula adakitic andesite has Sr and Nd isotopic compositions identical to normal mid-ocean ridge basalt (N-MORB), whereas the Okushiri Island and Toyama adakitic andesites are more isotopically primitive than N-MORB. The Noto Peninsula primary adakitic melt was derived from subducted oceanic N-MORB crust, whereas the Okushiri Island and Toyama primary adakites are interpreted as melts of subducted N-MORB and sediment that have subsequently interacted with the overlying mantle wedge peridotite. To explain the comagmatism of adakite, HMA and basalt, the following model is proposed. A hydrated adakitic diapir ascends from the subducting slab and is heated because it enters the overlying hot mantle wedge. The subsequent establishment of thermal and H 2 O gradients in the adakitic diapir and surrounding mantle wedge peridotite results in concurrent generation of adakitic andesite magma in the inner adakitic diapir region (low temperature and high H 2 O content), HMA and bronzite andesite magmas in the intermediate peridotite region (intermediate temperature and H 2 O content), and tholeiitic basalt magma in the outer peridotite region (high temperature and lower H 2 O content). Comagmatic adakite and mildly alkaline basalt are found in cooler and wetter adakitic diapirs and hotter and drier peridotite regions respectively. The most likely tectono-magmatic situation for the genesis of adakitic magmas in this example of a cool subduction zone involves upwelling of hot asthenosphere into the subcontinental lithosphere beneath the back-arc side of the NE Japan arc and northern end of the SW Japan arc, during the period spanning the pre-Japan Sea opening to syn-opening stages. The unusually high temperature conditions established in the mantle wedge owing to upwelling of hot asthenosphere caused partial melting of the relatively cool subducting Pacific plate, resulting in the generation of adakitic magmas.

    Description: As our cyber society develops and expands, the importance of cybersecurity operations is growing in response to cybersecurity threats coming from beyond national borders. Efficient cybersecurity operations require information exchanges that go beyond organizational borders. Various industry specifications defining information schemata for such exchanges are thus emerging. These specifications, however, define their own schemata since their objectives and the types of information they deal with differ, and desirable schemata differ depending on the purposes. They need to be organized and orchestrated so that individual organizations can fully exchange information and collaborate with one another. To establish the foundations of such orchestration and facilitate information exchanges, this paper proposes a reference ontology for cybersecurity operational information. The ontology structures cybersecurity information and orchestrates industry specifications. We built it from the standpoint of cybersecurity operations in close collaboration with cybersecurity organizations including security operation centers handling actual cybersecurity operations in the USA, Japan and South Korea. This paper demonstrates its usability by discussing the coverage of industry specifications. It then defines an extensible information structure that collaborates with such specifications by using the ontology and describes a prototype cybersecurity knowledge base we constructed that facilitates cybersecurity information exchanges among various parties. Finally, it discusses the usage scenarios of the ontology and knowledge base in cybersecurity operations. Through this work, we wish to contribute to the advancement of cybersecurity information exchanges.

    Description: We report on the first clear detection of transient Extremely-Low-Frequency (ELF) signal caused by an extremely intense cosmic gamma-ray flare. On 2004 December 27, the brightest gamma-ray flare ever recorded was observed by numerous satellites. A transient ELF emission observed at Moshiri and Onagawa in Japan exactly coincided with the peak time of the flare, and its wide pulse width of ∼40 ms disfavors the possibility of lightning origin. Furthermore, the two horizontal components of ELF magnetic field data recorded at Esrange in Sweden showed clear transient Schumann resonance waveforms. The source direction determined by the Lissajous method roughly corresponds to the subflare point. The chance probability that a sprite occurs within 30 ms of the peak flare time is ∼0.025%, which again clearly excludes the sprite origin. Thus, a bright cosmic gamma-ray flare is a new source of transient ELF radio signals observed on the Earth, although the emission mechanism needs to be clarified in future.

    Description: Small interfering RNA (siRNA)-based RNA interference (RNAi) is widely used for target gene silencing in various organisms. We previously showed that 8-nt-long 5' proximal nucleotides, which include seed sequence (positions 2–8 from the 5' end of guide strand), and the complementary sequence of the passenger strand are capable of being simultaneously replaced with cognate deoxyribonucleotides without any substantial loss of gene silencing. In the present study, examination was made of RNA requirements in the non-seed region of siRNA. The non-seed region of siRNA was found to be subdivided into four domains, in which two nucleotide pairs (positions 13 and 14) were replaceable with cognate deoxyribonucleotides without reducing RNAi activity. However, RNA sequences at positions 9-12 and 15-18 were essential for effective gene silencing, and these two double-stranded RNA cores are required for binding of the trans -activation response RNA-binding protein (TRBP). The terminal RNA (positions 19–21) provided Argonaute protein binding sites. Argonaute binding was enhanced by the presence of RNAs at positions 15–18. Knockdown experiments showed that, unlike Argonaute and TRBP, Dicer was dispensable for RNAi. Based on these observations, we discuss possible RNA/protein and protein/protein interactions in RNA-induced silencing complex formation.

    Description: Major and trace element and Sr–Nd–Pb isotope data for whole-rocks and major element data for minerals within basalt samples from the Chugaryeong volcano, an intra-plate back-arc volcanic centre in the central part of the Korean Peninsula, are used to address the process of magma genesis in the deep back-arc region of eastern Asia. There are two lava flow units at Chugaryeong volcano: the Chongok (0·50 Ma) and the Chatan (0·15 Ma) basalts. These basalts have similar MgO (9·1–10·4 wt %) but exhibit differences in their major and trace element and isotope compositions. The Chongok basalt has higher TiO 2 , Al 2 O 3 , Na 2 O, K 2 O, P 2 O 5 , Cr 2 O 3 , large ion lithophile elements (LILE), high field strength elements (HFSE), and rare earth elements (REE), and lower FeO*, SiO 2 , and CaO than the Chatan basalt. In addition, the Chongok basalt has more radiogenic 143 Nd/ 144 Nd and 206 Pb/ 204 Pb, and less radiogenic 87 Sr/ 86 Sr and 208 Pb/ 204 Pb than the Chatan basalt. Chi-square tests for the major elements indicate that crystal fractionation can explain the chemical variations within each basalt suite; intra-crustal processes, including crystal fractionation and assimilation of continental crust, cannot result in the formation of one basalt suite from the other. The Sr–Nd–Pb isotopic compositions of the Chongok and Chatan basalts plot on mixing hyperbolae between peridotite mantle xenoliths from the area and a fluid flux derived from a mixture of ancient and recent sediments. The trace element compositions of the estimated primary melts for the two basalt suites suggest different degrees of partial melting of a common enriched mantle source that was metasomatized by a Ba-, K-, Pb-, and Sr-rich fluid. The estimated degree of melting increased with time from ~7·5% for the Chongok basalt to ~10% for the Chatan basalt. The source mantle for the Chatan basalt is more enriched in Ba and Pb, indicating a greater fluid flux than for the Chongok basalt. This suggests that melting of the source mantle increased with time, sustained by an increased sediment-derived fluid flux from the deeper upper mantle.

    Description: Ultrafast outflows (UFOs) are seen in many AGN, giving a possible mode for AGN feedback on to the host galaxy. However, the mechanism(s) for the launch and acceleration of these outflows are currently unknown, with UV line driving apparently strongly disfavoured as the material along the line of sight is so highly ionized that it has no UV transitions. We revisit this issue using the Suzaku X-ray data from PDS 456, an AGN with the most powerful UFO seen in the local Universe. We explore conditions in the wind by developing a new 3D Monte Carlo code for radiation transport. The code only handles highly ionized ions, but the data show the ionization state of the wind is high enough that this is appropriate, and this restriction makes it fast enough to explore parameter space. We reproduce the results of earlier work, confirming that the mass-loss rate in the wind is around 30 per cent of the inferred inflow rate through the outer disc. We show for the first time that UV line driving is likely to be a major contribution to the wind acceleration. The mass-loss rate in the wind matches that predicted from a purely line driven system, and this UV absorption can take place out of the line of sight. Continuum driving should also play a role as the source is close to Eddington. This predicts that the most extreme outflows will be produced from the highest mass accretion rate flows on to high-mass black holes, as observed.

    Description: After the occurrence of the 2011 magnitude 9 Tohoku earthquake, the seismicity in the overriding plate changed. The seismicity appears to form distinct belts. From the spatiotemporal distribution of hypocenters, we can quantify the evolution of seismicity after the 2011 Tohoku earthquake. In some earthquake swarms near Sendai (Nagamachi-Rifu fault), Moriyoshi-zan volcano, Senya fault, and the Yamagata–Fukushima border (Aizu-Kitakata area, west of Azuma volcano), we can observe temporal expansion of the focal area. This temporal expansion is attributed to fluid diffusion. Observed diffusivity would correspond to the permeability of about 10 −15 (m 2 ). We can detect the area from which fluid migrates as a seismic low-velocity area. In the lower crust, we found seismic low-velocity areas, which appear to be elongated along N–S or NE–SW, the strike of the island arc. These seismic low-velocity areas are located not only beneath the volcanic front but also beneath the fore-arc region. Seismic activity in the upper crust tends to be high above these low-velocity areas in the lower crust. Most of the shallow earthquakes after the 2011 Tohoku earthquake are located above the seismic low-velocity areas. We thus suggest fluid pressure changes are responsible for the belts of seismicity. Temporal expansion of the focal area in some earthquake swarms near Sendai (Nagamachi-Rifu fault), Moriyoshi-zan volcano, Senya fault, and the Yamagata-Fukushima border (Aizu-Kitakata area, west of Azuma volcano) induced by the 2011 Tohoku-Oki earthquake was observed. This temporal expansion can be explained by fluid diffusion. We found seismic low-velocity areas, which is the possible areas with fluid, beneath the swarms. From the results, the induced earthquakes are thought to be affected by the possible fluid pressure change.

    Description: We developed a method for classifying hydrometeor particle types, including cloud and precipitation phase and ice crystal habit, by a synergistic use of CloudSat/Cloud Profiling Radar (CPR) and Cloud–Aerosol Lidar and Infrared Pathfinder Satellite Observations (CALIPSO)/Cloud-Aerosol LIdar with Orthogonal Polarization (CALIOP). We investigated how the cloud phase and ice crystal habit characterized by CALIOP globally relate with radar reflectivity and temperature. The global relationship thus identified was employed to develop an algorithm for hydrometeor type classification with CPR alone. The CPR-based type classification was then combined with CALIPSO-based type characterization to give CPR-CALIOP synergy classification. A unique aspect of this algorithm is to exploit and combine the lidar's sensitivity to thin ice clouds and the radar's ability to penetrate light precipitation to offer more complete picture of vertically resolved hydrometeor type classification than has been provided by previous studies. Given the complementary nature of radar and lidar detections of hydrometeors, our algorithm delivers thirteen hydrometeor types: warm water, supercooled water, randomly-oriented ice crystal (3D-ice), horizontally-oriented plate (2D-plate), 3D-ice+2D-plate, liquid drizzle, mixed-phase drizzle, rain, snow, mixed-phase cloud, water+liquid drizzle, water+rain and unknown. The global statistics of three-dimensional occurrence frequency of each hydrometeor type revealed that 3D-ice contributes the most to the total cloud occurrence frequency (53.8%), followed by supercooled water (14.3%), 2D-plate (9.2%), rain (5.9%), warm water (5.7%), snow (4.8%), mixed-phase drizzle (2.3%), and the remaining types (4.0%). This hydrometeor type classification provides useful observation-based information for climate model diagnostics in representation of cloud phase and their microphysical characteristics.

    Description: Centromeres consist of DNA repeats in many eukaryotes. Non-allelic homologous recombination (HR) between them can result in gross chromosomal rearrangements (GCRs). In fission yeast, Rad51 suppresses isochromosome formation that occurs between inverted repeats in the centromere. However, how the HR enzyme prevents homology-mediated GCRs remains unclear. Here, we provide evidence that Rad51 with the aid of the Swi/Snf-type motor protein Rad54 promotes non-crossover recombination between centromere repeats to prevent isochromosome formation. Mutations in Rad51 and Rad54 epistatically increased the rates of isochromosome formation and chromosome loss. In sharp contrast, these mutations decreased gene conversion between inverted repeats in the centromere. Remarkably, analysis of recombinant DNAs revealed that rad51 and rad54 increase the proportion of crossovers. In the absence of Rad51, deletion of the structure-specific endonuclease Mus81 decreased both crossovers and isochromosomes, while the cdc27/pol32-D1 mutation, which impairs break-induced replication, did not. We propose that Rad51 and Rad54 promote non-crossover recombination between centromere repeats on the same chromatid, thereby suppressing crossover between non-allelic repeats on sister chromatids that leads to chromosomal rearrangements. Furthermore, we found that Rad51 and Rad54 are required for gene silencing in centromeres, suggesting that HR also plays a role in the structure and function of centromeres.
