The infraciliatures of three Chesapeake Bay species of Eutintinnus conforming in lorica morphology to E. angustatus, E. pectinis, and E. tenuis were compared following Protargol silver impregnation. The kinetome of these species had a number of shared attributes including: 1) a right and left field that were of similar length and kinetal spacing; 2) two, rarely three, long dorsal kineties composed of monokinetids; 3) a ventral kinety; and 4) the absence of a true posterior kinety. Nonetheless, the specific organization of infraciliary components varied considerably among species. For example, the organization of the ventral kinety and several kineties along the border of the left field was different in all three species. In addition, the dorsal kineties of E. angustatus differed positionally from those of E. pectinis and E. tenuis. Some divergence in oral infraciliature was also noted, with the arrangement of infundibular oral polykinetids in E. pectinis and E. tenuis being distinct from that of E. angustatus. The amount of variation in the infraciliature of Eutintinnus spp. was much greater than previously noted for congeneric species of tintinnines and supports the separation E. angustatus, E. pectinis, and E. tenuis at the generic or subgeneric level.

    Novobiocin-Induced Ultrastructural Changes and Antagonism of DNA Synthesis in Trypanosoma cruzi Amastigotes Growing in Cell-Free Medium (1989)

    Notes: The antimicrobial agent novobiocin, an inhibitor of the bacterial enzyme topoisomerase II (DNA gyrase), is known to antagonize Trypanosoma cruzi amastigotes growing in cell-free medium. To determine sites of antagonism of novobiocin, the effects of drug on parasite ultrastructure and incorporation of radiolabeled precursors of DNA, RNA and protein into macromolecules were determined. The predominant ultrastructural abnormality seen after exposure to 0.40 mM novobiocin for 24 h was the presence of electron-dense clumps in the mitochondrion-kinetoplast organelle in 95 of 257 (37%) of cells, in comparison to no clumps seen in 110 drug-free cells. In addition, in the nucleus, the karyosome was less distinct than in control cells and appeared to merge with the chromatin. In the radiolabcling studies, incorporation of thymidine was inhibited in a dose-dependent fashion by novobiocin (0.16–0.80 mM) in a range of drug concentrations that also inhibited parasite growth. For 0.16 and 0.24 mM novobiocin, incorporation of thymidine was inhibited up to 65% relative to drug-free control cells while uptake of uridine and leucine was unaltered. We interpret these ultrastructure and precursor-incorporation studies as suggesting that (i) the mitochondrion-kinetoplast and possibly the nucleus are sites of novobiocin antagonism of T. cruzi amastigotes and (ii) that novobiocin appears to antagonize DNA synthesis within these organisms. Whether the drug target is topoisomerase II, however, is as yet unknown.

The in vitro activities of a series of quassinoids against Plasmodium falciparum, Entamoeba histolytica, Giardia intestinalis and Toxoplasma gondii have been compared with their in vitro cytotoxic effects against KB cells (human epidermoid carcinoma of the nasopharynx). All of the compounds tested were more toxic to KB cells than to G. intestinalis, but four (ailanthinone, bruceine D, brusatol and glaucarubinone) were slightly less toxic to KB cells than to E. histolytica. Glaucarubinone was similarly more toxic to intracellular T. gondii than to KB cells but ailanthinone was more selective (36 times more toxic to T. gondii than to KB cells). All of the compounds were more toxic to P. falciparum than to KB cells; the most selective quassinoids—glaucarubinone, bruceine D, ailanthinone and brusatoi—were found to have toxicity/activity ratios of 285, 76, 48 and 32 respectively. These results suggest that quassinoids have a selective action on P. falciparum. Further studies to elucidate the basis for this are in progress.

    Notes: The paper studies export pricing to market (PTM) in a “small-country” context using a panel of disaggregated exports from Hong Kong since 1992. Conventional wisdom is that PTM is commonplace—except for US exports. This study provides a benchmark by which to interpret the puzzling behavior of US export prices. Empirically, Hong Kong's export price behavior is comparable to that of the US. This similarity reinforces the idea that PTM behavior is also a function of home market conditions and the ability to price discriminate across markets. There is little evidence of differences in PTM across Hong Kong's export destinations.

    Notes: The total intensity, sweetness, and acidity of sucrose/citric acid mixtures were judged by two types of taste panel: experienced assessors, most of whom had had many years of experience in sensory evaluation; and novice assessors, none of whom had previously taken part in a taste experiment. In other respects the experimental conditions remained almost constant. There was good correspondence between the two panels, particularly for judgments of total intensity, indicating that novice and experienced assessors evaluate taste mixtures in the same way. However, there was also an indication that experience on sensory panels may attenuate taste suppression, the suppression of acidity by sweetness being less pronounced for the experienced panel than for the novice panel. The implications for mixture perception are noted.

    Notes: A method is presented for determining steady-state capture zones in three dimensions around horizontal drains and vertical wells in homogeneous, anisotropic aquifers in a uniform flow field. Equations are presented for determining drawdown and velocity vector components in three dimensions around drains and wells. Using these equations, a second-order Runge-Kutta particle tracking algorithm is applied to trace streamlines in three dimensions. By tracking a large number of particles, it is possible to determine areas where capture occurs and areas where particles escape capture. The resulting 3D capture zones are diagrammed as both 2D (section view) plots and 3D plots.

    Notes: The ground waters of Runnels County, Texas, are highly contaminated with nitrate. The average nitrate concentration of 230 water samples was 250 mg/I NO3.The natural variations of the stable nitrogen isotopes N14 and N15 identified natural soil nitrate as the predominant source. Nitrate from animal wastes was of minor importance. The δN15 range of natural soil nitrate was +2 to +8% whereas the δN15 range of animal waste nitrate was +10 to +20%- (Atmospheric nitrogen was used as a standard for mass spectrometric analysis. Experimental error for sample preparation and isotopic analysis was ±1 %.) More than 66 percent of the ground-water nitrates analyzed were in the δN15 range of natural soil nitrates.Dryland farming since 1900 has caused the oxidation of the organic nitrogen in the soil to nitrate. Minimal fertilizer has been used because of the lack of suitable water for irrigation. During the period 1900-1950, nitrate was leached below the root zone but not to the water table. Extensive terracing after the drought in the early 1950's has raised the water table approximately 6 meters and has leached the nitrate into the ground water. Tritium dates indicate that the ground water is less than 20 years old.
