    Notes: Acinetobacter calcoaceticus RJE74 contains a large transmissible catabolic plasmid, pWW174, of about 200 kb, which encodes its ability to grow on benzene (Bzn+). pWW174 was unstable in Acinetobacter hosts and was lost at high frequency in the absence of selection for Bzn*. The catabolic pathway appeared to be via benzene cis-glycol, catechol and the β-ketoadipate (ortho) pathway. pWW174 encodes a catechol 1,2-oxygenase which is significantly more thermolabile than the chromosomally determined enzyme. pWW174 was able to complement all cat mutants (catechol to central metabolites) of A. calcoaceticus ADP1 (BD413) tested. Two regions of the plasmid were cloned, one carrying catA, the gene for catechol 1,2-oxygenase, and another carrying catBCDE, the subsequent four enzymes of the β-ketoadipate pathway: these two regions appeared to be separated by at least 10 kbp. Hybridization indicated homology between the plasmid cat genes and the corresponding chromosomal genes of ADPL.

    Notes: In Pseudomonas aeruginosa, the production of many virulence factors and secondary metabolites is regulated in concert with cell density through quorum sensing. Two quorum-sensing regulons have been identified in which the LuxR homologues LasR and RhIR are activated by N-(3-oxododecanoyl)-l-homo-serine lactone (OdDHL) and N-butanoyl-l-homoserine lactone (BHL) respectively. The lasR and rhIR genes are linked to the luxl homologues last and rhll, which are responsible for synthesis of OdDHL and BHL, respectively. As lasRI and rhlRI are both involved in regulating synthesis of exoenzymes such as elastase, we sought to determine the nature of their interrelationship. By using lacZ transcriptional fusions in both homologous (P. aeruginosa) and heterologous (Escherichia coli) genetic backgrounds we provide evidence that (i) lasR is expressed constitutively throughout the growth cycle, (ii) rhIR expression is regulated by LasR/OdDHL, and (iii) that RhIR/BHL regulates rhll. We also show that expression of the stationary-phase sigma factor gene rpoS is abolished in a P. aeruginosa lasR mutant and in the pleiotropic BHL-negative mutant PAN067. Furthermore, our data reveal that in E. coli, an rpoS-lacZ fusion is regulated directly by RhIR/BHL. Taken together, these results indicate that P. aeruginosa employs a multilayered hierarchical quorum-sensing cascade involving RhIR/BHL and LasR/OdDHL, interlinked via RpoS, to integrate the regulation of virulence determinants and secondary metabolites with adaptation and survival in the stationary phase.

    Notes: In recent years it has become clear that the production of N-acyl homoserine lactones (N-AHLs) is widespread in Gram-negative bacteria. These molecules act as diffusible chemical communication signals (bacterial pheromones) which regulate diverse physiological processes including bioluminescence, antibiotic production, piasmid conjugal transfer and synthesis of exoenzyme virulence factors in plant and animal pathogens. The paradigm for N-AHL production is in the bioluminescence (lux) phenotype of Photobacterium fischeri (formerly classified as Vibrio fischeri) where the signalling molecule N-(3-oxohexanoyl)-L-homoserine lactone (OHHL) is synthesized by the action of the Luxl protein. OHHL is thought to bind to the LuxR protein, allowing it to act as a positive transcriptional activator in an autoinduction process that physiologically couples cell density (and growth phase) to the expression of the bioluminescence genes. Based on the growing information on Luxl and LuxR homologues in other N-AHL-producing bacterial species such as Erwinia carotovora, Pseudomonas aeruginosa, Yersinia enterocolitica, Agrobacterium tumefaciens and Rhizobium legumino-sarum, it seems that analogues of the P. fischeri lux autoinducer sensing system are widely distributed in bacteria. The general physiological function of these simple chemical signalling systems appears to be the modulation of discrete and diverse metabolic processes in concert with cell density. In an evolutionary sense, the elaboration and action of these bacterial pheromones can be viewed as an example of multi-cellularity in prokaryotic populations.

    Notes: The use of NIR spectroscopy for the prediction of amino acid compositions in wheat and barley has been demonstrated. Using 2nd derivative mathematical treatments of log I/R data of ground wheat samples obtained on the Beltsville Universal Computerized Spectrophotometer, the concentration of lysine, threonine, tryptophan and methionine could be predicted with accuracies acceptable for screening purposes. With barley samples low standard errors of prediction (high accuracy) were obtained for I3 amino acids using a research NIR and a commercial bench-type instrument. For both of these instruments, use of 1st derivative mathematical treatments of spectral data gave the best accuracy. Results indicated that NIR techniques may be very useful in plant breeding programs and in the food and feed industry for the rapid and accurate analysis of amino acids.

    Notes: Abstract Collection and washing of cells by centrifugation is a time consuming and inconvenient component of many microbiological protocols, especially when sterility must be maintained. Filtration onto disposable membrane filters efficiently replaced the centrifugation steps in standard transformational competence protocols for Escherichia coli, Haemophilus influenzas, and Saccharomyces cerevisiae. With all three protocols filtered cells were competent sooner and showed transformation efficiencies comparable to or higher than those of cells prepared by centrifugation. The procedure can be easily adapted to collect cells for many other purposes, and should be especially useful in metabolic studies and in preparing cells for electroporation, as it facilitates both thorough washing and rapid but gentle resuspension of cells without compromising sterility.

    Notes: Abstract A series of isogenic mutants lacking either the O1 (O−:K66) or K66 (O1:K− antigens or both (O−:K−), some of which had additional defects in their LPS core polysaccharide was used to examine the interaction between polymorphonuclear leucocytes (PMNLs) and K. pneumoniae serotype O1:K66. In the absence of serum complement, only a O−:K− strain with a deep rough LPS chemotype elicited a PMNL-dependent chemiluminescent (CL) response. However, following opsonization of the non-capsulated strains by complement, the largest CL response was to the O1:K−1 mutant. This mutant also activated and bound more complement C3 than any of the other encapsulated or non-capsulated strains examined. Despite the surface exposure of smooth and rough LPS in the encapsulated parent and mutant strains, the K66 antigen reduced the binding of C3 and prevented PMNL activation. Both anti-LPS and anti-K66 antibodies, however, stimulated a PMNL-dependent CL response to the K66 bearing strains.

    Notes: Abstract The NH2-terminal amino sequence through the first 20 amino acids was obtained for transferrin-binding protein (TBP)1 from three strains of Neisseria meningitidis. These were identical except for a glutamine to a glycine substitution at residue 6 in one case. The sequences of the NH2-terminal 20 amino acids of TBP2 from the same three strains were also determined; one TBP2 had a Mr of 68 000 and the other two of 78 000. Sequences were identical up to residue 13 in all three proteins. Peptides based on the NH2-terminal sequences of TBP1 and 2 were synthesized, linked to Keyhole Limpet haemocyanin and used to raise antibodies in rabbits. Anti-peptide antibodies cross-reacted on immunoblotting with the respective TBPs from all meningococcal strains tested, as well as with those from N. gonorrhoeae suggesting that the NH2-terminals of these proteins are well conserved in the Neisseria. Neither anti-peptide serum reacted with the analogous TBP1 and 2 from Haemophilus influenzae, although common epitopes have previously been shown to exist.

    Notes: Abstract We describe experimental results on the extensional viscosity of mobile polymer solutions obtained from two instruments, the first being a commercial Spin Line Rheometer and the second a custom-built lubricated-die Converging Flow Rheometer. The interpretation of data in terms of Trouton ratios is facilitated by a simple analysis for the Generalized Newtonian Fluid model. Agreement between data from the two rheometers is satisfactory and we show that polymer solutions can be either tension stiffening or tension thinning. However, the Trouton ratios in both cases are greater than the Newtonian values and we anticipate that this will always be the case for polymer solutions.
