    Notes: A simple method was developed for purifying Escherichia coli H 30 verocytotoxin (VT). The toxin, released from the cells by exposure to polymyxin B, was subjected to differential ammonium sulphate precipitation and sequential chromatography on hydroxylapatite, chromatofocussing, Cibachron blue, and Sephadex G-100 columns. The purified toxin, of 39 kDa by gel filtration and a pI of 6.72, resolved as a band which migrated at 32 kDa and another band, of less than 14 kDa, which migrated with the buffer front on reducing sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The purified preparation was relatively heat-stable, and had a specific activity of 3 × 109 CD50 units/mg protein in Vero cells, and LD50 values of 0.2, 9.0, and 40 μg proteindeadenylylation. Carbon-limited chemostat cultures showed low glutamine synthetase activity levels but the synthesis of the enzyme was derepressed when the cultures became N-limited. kg in rabbits, rats, and mice, respectively. Antiserum to the toxin specifically neutralized H 30 VT, Shiga toxin, and VT activity from some clinical isolates of VT+E. coli, but not that from a porcine oedema disease strain.

    Notes: Abstract Verocytotoxin 2 (VT2) was purified from Escherichia coli strain E32511 using, as starting material, cells harvested from a Penassay broth culture incubated for 6 h at 37°C in the presence of mitomycin C (0.2 μg/ml). A crude extract of VT2, obtained by polymyxin B treatment of cell pellets, was purified using differential ammonium sulphate precipitation, and sequential column chromatography. The purified toxin was estimated to have a pI of 6.5 by chromatofocusing and a molecular weight of 42 000 by gel filtration; it had a specific activity of 1.39 × 106 CD50 units/mg protein in Vero cells, and resolved as a major band of Mr 35 000 and another band of 〈14 000 which migrated with the buffer front on reducing SDS-polyacrylamide gel electrophoresis. The purified toxin was not neutralized by VT1 antisera and antisera prepared to this toxin in rabbits did not neutralize VT1, but completely neutralized the activity of the homologous toxin. Purified VT2, when administered intravenously, was lethal to rabbits with an LD50 of 30 μg/kg. Rabbits displayed symptoms of overt haemorrhagic caecitis and flaccid paralysis.

    Description: The behaviour of aquifers with karst porosity is dependent on hydrological conditions. This is due to the peculiar characteristics of the groundwater flow and dynamics of hydrological processes in karst. As a result, karst aquifers are especially vulnerable to the effects of environmental change. We assessed the long-term climatological and hydrological trends and the short-term effects of increasingly frequent extreme hydrological events (droughts) for the Mediterranean karst spring Rižana in SW Slovenia. The findings predict higher mean annual air temperatures of 0.34°C decade –1 , lower annual precipitation of c. 60 mm decade –1 and higher annual actual evapotranspiration (especially during spring and summer) of 32–49 mm decade –1 . As a consequence, we can expect a decrease in the mean annual discharge of the spring of c. 480 l s –1 decade –1 with prolonged dry summer periods. Detailed monitoring of the physical, chemical and microbiological parameters showed that the flood pulses caused by precipitation events after a long dry period cause a significant deterioration in water quality. In such situations, contaminants stored in the unsaturated zone are flushed out and counts of coliform bacteria can reach 〉1400 cfu (100 ml) –1 , with total Al and Fe concentrations up to 206 and 474 μg l –1 , respectively. These results suggest that there should be urgent adherence to water quality standards to protect karst water sources in view of the anticipated climatological stresses. Management strategies should promote monitoring and the rational use of karst water supplies.
