    Notes: Abstract— An important aspect in the development of any aquaculture industry is the maximization of juvenile somatic growth (i.e., body growth) to reduce production time and increase the size of the final product. In this study, green sea urchins Strongylocentrotus clroebachiensis were fed a prepared diet from 4 December 1998 to 10 September 1999 (i.e., 280 d) in a laboratory to investigate the effect of protein source (soybean andlor fish), protein concentration (20,30,40, and 50% dry mass) and juvenile size (4-8 mm and 12-20 mm initial test diameter) on somatic growth. A natural diet of Laminwia longicrurus (i.e., kelp) was used as a reference. There was no difference in initial size among the treatments for either the smaller cohort 1 or the larger cohort 2 sea urchins (6.3 mm and 13.8 mm initial average test diameter, respectively) (P 〉 0.05 for all tests). After 280 d, the sea urchins fed kelp had an average size of 20.7 mm and 24.5 mm (cohort 1 and cohort 2, respectively). The final average sizes of the sea urchins fed the prepared diets, which did not relate to dietary protein concentrations andlor protein source, ranged from 13.2 mm to 16.2 mm (cohort 1) and from 20.4 mm to 22.9 mm (cohort 2), and were significantly smaller than the kelpfed sea urchins (P 〈 0.05 and P 〈 0.001, cohort 1 and cohort 2, respectively). All treatments experienced 95% survivorship or greater. Sea urchin size appears to affect growth rate when optimal conditions for growth are available (i.e., diet and water temperature). As water temperatures increased during the summer of 1999, the sea urchins in cohort 1 fed kelp had a significantly higher growth rate (0.069 dd) than the cohort 2 kelpfed sea urchins (0.052 dd) (P 〈 0.05). However, within each cohort, there were no significant difference in growth rate (P 〉 0.05) among the sea urchins fed prepared diets, suggesting sea urchins do not require high concentrations of dietary protein for superior growth, and that plant protein can substitute fish protein in sea urchin diets. Furthermore, the sea urchins fed the prepared diets had poorer test quality and larger gonad yields (13-22%) compared to the kelpfed sea urchins (4.2%) and a wild sample of sea urchins of similar size (4.0%). The results suggest that the sea urchins fed the prepared diets allocated more energy to gonad production, whereas those fed a natural diet allocated more energy toward test production. To address this gap, more research is required to identify the nutritional components required for test growth that were present in kelp, but appeared to be deficient in the prepared diets.

    Notes: The development of efficient feed management strategies in shrimp farming involves the investigation of aspects related to time of feed distribution, feeding rates, feeding frequency, and feed dispersal methods. This work examined the feeding levels of Penaeus subtilis in response to food dispersal method under semi-intensive culture conditions. The study consisted of two treatments: 1) a uniform feed dispersal over the culture area, and 2) concentration of feed in 0.2-m2 circular feeding trays. For each, three 105-m2 rectangular enclosures were installed in a single shrimp grow-out pond and stocked with P. subtilis at a density of 14.29 postlarvae/m2. Shrimp were fed daily at 0600, 0930 and 1430 h and sampled 30 min after feed distribution at 12-d intervals. After collection, shrimp weight was determined, their stomachs dissected and weighed. A total of 2,160 stomachs of P. subtilis were collected during a 89-d rearing cycle. Relative occurrence of artificial versus natural food in stomach contents was determined by stable carbon isotope mass spectrometry. Density of polychaetes in the substrate was monitored at each sampling period. After harvest, sediment samples were collected for physical and chemical analyses. In general, differences in shrimp growth between feeding methods were not significant. Final shrimp survival rates were between 69.0 ± 6.9% and 71.9 ± 3.7%. Stomach content weight of P. subtilis was significantly higher when feed was broadcast than for feed concentration. Average stomach content weight remained constant throughout the rearing cycle, although progressive increases in feed Occurrence were detected in P. subtilis diet. No periodicity in food intake along sampling periods could be related to increases in shrimp carapace length. Food intake was significantly lower at 0600 h than at 1430 h and at 0930 h, respectively. In both treatments, over half of the food contained in the stomach contents of shrimp was derived from artificial food. Sediment chemical analysis indicated statistically lower levels of sodium in the pond bottom of concentrated treatment. However, no short-term detrimental effects were detected on sediment chemical quality. Results indicated that feed broadcasting resulted in a greater access and a higher consumption of food among the cultured shrimp population, a lower number of empty stomachs, and a greater occurrence of artificial food in P. subtilis stomach contents.

    Notes: Using H-2 recombinant and mutant mice, the Rfv-1 gene influencing spontaneous recovery from Friend retrovirus (FV)-induced leukaemia was mapped in the D locus. Two D b alleles were required for full recovery, and a single Dd transgene did not convey increased susceptibility to FV in the presence of homozygous Db/b genotype. The results suggest that an increase in the expression of Db may lead to more effective stimulation of FV-specific CTL.

    Description: The aerosol mass spectrometer (AMS), combined with statistical methods such as positive matrix factorization (PMF), has greatly advanced the quantification of primary organic aerosol (POA) sources and total secondary organic aerosol (SOA) mass. However, the use of thermal vaporization and electron ionization yields extensive thermal decomposition and ionization-induced fragmentation, which limit chemical information needed for SOA source apportionment. The recently developed extractive electrospray ionization time-of-flight mass spectrometer (EESI-TOF) provides mass spectra of the organic aerosol fraction with a linear response to mass and no thermal decomposition or ionization-induced fragmentation. However, the costs and operational requirements of online instruments make their use impractical for long-term or spatially dense monitoring applications. This challenge was overcome for AMS measurements by measuring re-nebulized water extracts from ambient filter samples. Here, we apply the same strategy for EESI-TOF measurements of 1 year of 24 h filter samples collected approximately every fourth day throughout 2013 at an urban site. The nebulized water extracts were measured simultaneously with an AMS. The application of positive matrix factorization (PMF) to EESI-TOF spectra resolved seven factors, which describe water-soluble OA: less and more aged biomass burning aerosol (LABBEESI and MABBEESI, respectively), cigarette-smoke-related organic aerosol, primary biological organic aerosol, biogenic secondary organic aerosol, and a summer mixed oxygenated organic aerosol factor. Seasonal trends and relative contributions of the EESI-TOF OA sources were compared with AMS source apportionment factors, measured water-soluble ions, cellulose, and meteorological data. Cluster analysis was utilized to identify key factor-specific ions based on PMF. Both LABB and MABB contribute strongly during winter. LABB is distinguished by very high signals from C6H10O5 (levoglucosan and isomers) and C8H12O6, whereas MABB is characterized by a large number of CxHyOz and CxHyOzN species of two distinct populations: one with low H:C and high O:C and the other with high H:C and low O:C. Two oxygenated summertime SOA sources were attributed to terpene-derived biogenic SOA, a major summertime aerosol source in central Europe. Furthermore, a primary biological organic aerosol factor was identified, which was dominated by plant-derived fatty acids and correlated with free cellulose. The cigarette-smoke-related factor contained a high contribution of nicotine and high abundance of organic nitrate ions with low m∕z.

    Description: This study assessed the impact of a PRRSV (porcine reproductive and respiratory syndrome virus) recombinant strain (Horsens strain) on the reproductive performance of naïve pregnant sows in the last third of gestation. Fifteen sows were included: four negative reproductive controls (NTX), five infected with a PRRSV-1 field strain (Olot/91, T01), and six infected with the recombinant PRRSV-1 strain (Horsens strain, T02). Piglets were monitored until weaning. Reproductive performance was the primary variable. In sows, viremia and nasal shedding (T01 and T02 groups), and, in piglets, viral load in blood and in lungs, as well as macroscopic lung lesions (T01 and T02 groups), were the secondary variables. The reproductive performance results were numerically different between the two challenged groups. Moreover, viral loads in blood were 1.83 × 106 ± 9.05 × 106 copies/mL at farrowing, 1.05 × 107 ± 2.21 × 107 copies/mL at weaning from piglets born from T01 animals and 1.64 × 103 ± 7.62 × 103 copies/mL at farrowing, 1.95 × 103 ± 1.17 × 104 copies/mL at weaning from piglets born from T02 sows. Overall, 68.8% of T01 piglets and 38.1% of T02 piglets presented mild lung lesions. In conclusion, the results suggest that Horsens strain is less virulent than the field strain Olot/91 under these experimental conditions.

    Description: Characteristics and sources of volatile organic compounds (VOCs) were investigated with highly time-resolved simultaneous measurements by two proton-transfer-reaction time-of-flight mass spectrometers (PTR-ToF-MS) at an urban and a suburban site in New Delhi, India, from January to March 2018. During the measurement period, high mixing ratios of VOCs and trace gases were observed, with high nocturnal mixing ratios and strong day–night variations. The positive matrix factorization (PMF) receptor model was applied separately to the two sites, and six major factors of VOCs were identified at both sites, i.e., two factors related to traffic emissions, two to solid fuel combustion, and two secondary factors. At the urban site, traffic-related emissions comprising mostly mono-aromatic compounds were the dominant sources, contributing 56.6 % of the total mixing ratio, compared to 36.0 % at the suburban site. Emissions from various solid fuel combustion processes, particularly in the night, were identified as a significant source of aromatics, phenols and furans at both sites. The secondary factors accounted for 15.9 % of the total VOC concentration at the urban site and for 33.6 % at the suburban site. They were dominated by oxygenated VOCs and exhibited substantially higher contributions during daytime.

    Description: Omega-3 (n-3) treatment may lower cardiovascular risk, yet its effects on the circulating lipidome and relation to cardiovascular risk biomarkers are unclear. We hypothesized that n-3 treatment is associated with favorable changes in downstream fatty acids (FAs), oxylipins, bioactive lipids, clinical lipid and inflammatory biomarkers. We examined these VITAL200, a nested substudy of 200 subjects balanced on demographics and treatment and randomly selected from the Vitamin D and Omega-3 Trial (VITAL). VITAL is a randomized double-blind trial of 840 mg/d eicosapentaenoic acid (EPA) + docosahexaenoic acid (DHA) vs. placebo among 25,871 individuals. Small polar bioactive lipid features, oxylipins and FAs from plasma and red blood cells were measured using three independent assaying techniques at baseline and one year. The Women’s Health Study (WHS) was used for replication with dietary n-3 intake. Randomized n-3 treatment led to changes in 143 FAs, oxylipins and bioactive lipids (False Discovery Rate (FDR) 〈 0.05 in VITAL200, validated (p-values 〈 0.05)) in WHS with increases in 95 including EPA, DHA, n-3 docosapentaenoic acid (DPA-n3), and decreases in 48 including DPA-n6, dihomo gamma linolenic (DGLA), adrenic and arachidonic acids. N-3 related changes in the bioactive lipidome were heterogeneously associated with changes in clinical lipid and inflammatory biomarkers. N-3 treatment significantly modulates the bioactive lipidome, which may contribute to its clinical benefits.

    Description: Improving the understanding of the health and climate impacts of aerosols remains challenging and is restricted by the limitations of current measurement techniques. Detailed investigation of secondary organic aerosol (SOA), which is typically the dominating fraction of the organic aerosol (OA), requires instrumentation capable of real-time, in situ measurements of molecular composition. In this study, we present the first ambient measurements by a novel extractive electrospray ionization time-of-flight mass spectrometer (EESI-TOF-MS). The EESI-TOF-MS was deployed along with a high-resolution time-of-flight aerosol mass spectrometer (HR-ToF-AMS) during summer 2016 at an urban location (Zurich, Switzerland). Positive matrix factorization (PMF), implemented within the Multilinear Engine (ME-2), was applied to the data from both instruments to quantify the primary and secondary contributions to OA. From the EESI-TOF-MS analysis, a six-factor solution was selected as the most representative and interpretable solution for the investigated dataset, including two primary and four secondary factors. The primary factors are dominated by cooking and cigarette smoke signatures while the secondary factors are discriminated according to their daytime (two factors) and night-time (two factors) chemistry. All four factors showed strong influence by biogenic emissions but exhibited significant day–night differences. Factors dominating during daytime showed predominantly ions characteristic of monoterpene and sesquiterpene oxidation while the night-time factors included less oxygenated terpene oxidation products, as well as organonitrates which were likely derived from NO3 radical oxidation of monoterpenes. Overall, the signal measured by the EESI-TOF-MS and AMS showed a good correlation. Further, the two instruments were in excellent agreement in terms of both the mass contribution apportioned to the sum of POA and SOA factors and the total SOA signal. However, while the oxygenated organic aerosol (OOA) factors separated by AMS analysis exhibited a flat diurnal pattern, the EESI-TOF-MS factors illustrated significant chemical variation throughout the day. The captured variability, inaccessible from AMS PMF analysis, was shown to be consistent with the variations in the physiochemical processes influencing chemical composition and SOA formation. The improved source separation and interpretability of EESI-TOF-MS results suggest it to be a promising approach to source apportionment and atmospheric composition research.

    Description: The Xact 625 Ambient Metals Monitor was tested during a 3-week field campaign at the rural, traffic-influenced site Härkingen in Switzerland during the summer of 2015. The field campaign encompassed the Swiss National Day fireworks event, providing increased concentrations and unique chemical signatures compared to non-fireworks (or background) periods. The objective was to evaluate the data quality by intercomparison with other independent measurements and test its applicability for aerosol source quantification. The Xact was configured to measure 24 elements in PM10 with 1 h time resolution. Data quality was evaluated for 10 24 h averages of Xact data by intercomparison with 24 h PM10 filter data analysed with ICP-OES for major elements, ICP-MS for trace elements, and gold amalgamation atomic absorption spectrometry for Hg. Ten elements (S, K, Ca, Ti, Mn, Fe, Cu, Zn, Ba, Pb) showed excellent correlation between the compared methods, with r2 values  ≥  0.95. However, the slopes of the regressions between Xact 625 and ICP data varied from 0.97 to 1.8 (average 1.28) and thus indicated generally higher Xact elemental concentrations than ICP for these elements. Possible reasons for these differences are discussed, but further investigations are needed. For the remaining elements no conclusions could be drawn about their quantification for various reasons, mainly detection limit issues. An indirect intercomparison of hourly values was performed for the fireworks peak, which brought good agreement of total masses when the Xact data were corrected with the regressions from the 24 h value intercomparison. The results demonstrate that multi-metal characterization at high-time-resolution capability of Xact is a valuable and practical tool for ambient monitoring.
