    Description: Background: Severe shortage of liver donors and hepatocytes highlights urgent requirement of extra-liver and stem cell source of hepatocytes for treating liver-related diseases. Here we hypothesized that spermatogonial stem cells (SSCs) can directly transdifferentiate to hepatic stem-like cells capable of differentiating into mature hepatocyte-like cells in vitro without an intervening pluripotent state. Results: SSCs first changed into hepatic stem-like cells since they resembled hepatic oval cells in morphology and expressed Ck8, Ck18, Ck7, Ck19, OV6, and albumin. Importantly, they co-expressed CK8 and CK19 but not ES cell markers. Hepatic stem-like cells derived from SSCs could differentiate into small hepatocytes based upon their morphological features and expression of numerous hepatic cell markers but lacking of bile epithelial cell hallmarks. Small hepatocytes were further coaxed to differentiate into mature hepatocyte-like cells, as identified by their morphological traits and strong expression of Ck8, Ck18, Cyp7a1, Hnf3b, Alb, Tat, Ttr, albumin, and CYP1A2 but not Ck7 or CK19. Notably, these differentiated cells acquired functional attributes of hepatocyte-like cells because they secreted albumin, synthesized urea, and uptake and released indocyanine green. Moreover, phosphorylation of ERK1/2 and Smad2/3 rather than Akt was activated in hepatic stem cells and mature hepatocytes. Additionally, cyclin A, cyclin B and cyclin E transcripts and proteins but not cyclin D1 or CDK1 and CDK2 transcripts or proteins were reduced in mature hepatocyte-like cells or hepatic stem-like cells derived from SSCs compared to SSCs. Conclusions: SSCs can transdifferentiate to hepatic stem-like cells capable of differentiating into cells with morphological, phenotypic and functional characteristics of mature hepatocytes via the activation of ERK1/2 and Smad2/3 signaling pathways and the inactivation of cyclin A, cyclin B and cyclin E. This study thus provides an invaluable source of mature hepatocytes for treating liver-related diseases and drug toxicity screening and offers novel insights into mechanisms of liver development and cell reprogramming.

    Description: Haiyang-2A (HY-2A) has been working in-flight for over seven years, and the accuracy of HY-2A calibration microwave radiometer (CMR) data is extremely important for the wet troposphere delay correction (WTC) in sea surface height (SSH) determination. We present a comprehensive evaluation of the HY-2A CMR observation using the numerical weather model (NWM) for all the data available period from October 2011 to February 2018, including the WTC and the precipitable water vapor (PWV). The ERA(ECMWF Re-Analysis)-Interim products from European Centre for Medium-Range Weather Forecasts (ECMWF) are used for the validation of HY-2A WTC and PWV products. In general, a global agreement of root-mean-square (RMS) of 2.3 cm in WTC and 3.6 mm in PWV are demonstrated between HY-2A observation and ERA-Interim products. Systematic biases are revealed where before 2014 there was a positive WTC/PWV bias and after that, a negative one. Spatially, HY-2A CMR products show a larger bias in polar regions compared with mid-latitude regions and tropical regions and agree better in the Antarctic than in the Arctic with NWM. Moreover, HY-2A CMR products have larger biases in the coastal area, which are all caused by the brightness temperature (TB) contamination from land or sea ice. Temporally, the WTC/PWV biases increase from October 2011 to March 2014 with a systematic bias over 1 cm in WTC and 2 mm in PWV, and the maximum RMS values of 4.62 cm in WTC and 7.61 mm in PWV occur in August 2013, which is because of the unsuitable retrieval coefficients and systematic TB measurements biases from 37 GHz band. After April 2014, the TB bias is corrected, HY-2A CMR products agree very well with NWM from April 2014 to May 2017 with the average RMS of 1.68 cm in WTC and 2.65 mm in PWV. However, since June 2017, TB measurements from the 18.7 GHz band become unstable, which led to the huge differences between HY-2A CMR products and the NWM with an average RMS of 2.62 cm in WTC and 4.33 mm in PWV. HY-2A CMR shows high accuracy when three bands work normally and further calibration for HY-2A CMR is in urgent need. Furtherly, 137 global coastal radiosonde stations were used to validate HY-2A CMR. The validation based on radiosonde data shows the same variation trend in time of HY-2A CMR compared to the results from ECMWF, which verifies the results from ECMWF.

    Description: As an intrinsic property, elasticity of soft material is affected significantly by the externally applied alternating magnetic field. Magnetostrictive properties of the grain-oriented (GO) silicon steel under DC-biased and multisinusoidal magnetizations are measured by using a laser-based measuring system. Magnetostriction curves of the GO silicon steel sheet under different magnetizations are obtained and the influence of frequency and DC bias on the magnetostrictive property is observed and analyzed based on the measured data. In addition, the spectrum of magnetostriction under harmonic magnetization is obtained, and the acoustic noise level of the GO silicon steel sheet represented by the A-weighted decibel value caused by magnetostriction is measured under DC-biased and multisinusoidal magnetizations. The measurement results are applied to the simulation of the three-limb laminated core model, and the effects of DC bias and harmonics on magnetic flux density and displacement are analyzed.

    Description: An experimental wideband chaotic ground penetrating radar is proposed to locate underground pipes. A chaotic signal with a bandwidth of 1.56 GHz is utilized as the probe signal. The localization of the pipes is achieved by correlating the chaotic echo signal with its delayed duplicate and back-projection algorithm. Experimental results demonstrate that plastic pipe, metallic pipe, and multiple pipes can be located with a range resolution of 10 cm. Limited by the height of the sand, the detectable range is estimated to be 0.7 m for both the plastic pipes and the metallic pipes when the transmitting power is −12 dBm. The proposed system has the potential to detect buried pipes, and it is suitable for geological and civil engineering applications.

    Description: Agricultural emissions are crucial to regional air quality in the autumn and spring due to the intense agricultural activities in Northeast China. However, information on rural ambient particulate matter (PM) in Northeast China is rare, limiting the accurate estimation of agricultural atmospheric particulate matter emissions. In this study, we monitored hourly ambient PM2.5 (PM with a diameter of less than 2.5 μm) concentrations and analyzed daily chemical components (i.e., water-soluble ions, trace elements, organic carbon, and element carbon) at a rural site in Northeast China during the autumn and spring and assessed the impact of agricultural activities on atmospheric PM2.5 concentrations. The results showed that the daily average concentrations of PM2.5 were 143 ± 109 (range: 39–539) μg m−3 from 19 October to 23 November 2017 (i.e., typical harvesting month) and 241 ± 189 (range: 97–976) μg m−3 from 1 April to 13 May 2018 (i.e., typical tilling month). In autumn, the ambient PM2.5 concentrations were high with a Southwest wind, while a Southeast wind caused high PM2.5 concentrations during spring in the rural site. The concentrations of selected water-soluble ions, trace elements, and carbonaceous fractions accounted for 33%, 4%, and 26% of PM2.5 mass concentrations, respectively, in autumn and for 10%, 5%, and 3% of PM2.5 mass concentrations, respectively, in spring. On the basis of the component analysis, straw burning, agricultural machinery, and soil dust driven by wind and tilling were the main contributors to high rural PM2.5 concentrations. In addition, the increasing coal combustion around the rural site was another important source of PM2.5.

    Description: Embankments are usually constructed along rivers as a defense structure against flooding. Overtopping failure can cause devastating and fatal consequences to life and property of surrounding areas. This motivates researchers to study the formation, propagation, and destructive consequences of such hazards in risk analysis of hydraulic engineering. This paper reports a numerical simulation of failure processes in homogeneous embankments due to flow overtopping. The employed numerical approach is based on a double-point two-phase material point method (MPM) considering water–soil interaction and seepage effects. The simulated results are compared to available laboratory experiments in the literature. It was shown that the proposed method can predict the overtopping failure process of embankments with good accuracy. Furthermore, the effects of the cohesion, internal fiction angle, initial porosity, and maximum porosity of soil on the embankment failure are investigated.

    Description: Cellular automata (CA) is a bottom-up self-organizing modeling tool for simulating contagion-like phenomena such as complex land-use change and urban growth. It is not known how CA modeling responds to changes in spatial observation scale when a larger-scale study area is partitioned into subregions, each with its own CA model. We examined the impact of changing observation scale on a model of urban growth at UA-Shanghai (a region within a one-hour high-speed rail distance from Shanghai) using particle swarm optimization-based CA (PSO-CA) modeling. Our models were calibrated with data from 1995 to 2005 and validated with data from 2005 to 2015 on spatial scales: (1) Regional-scale: UA-Shanghai was considered as a single study area; (2) meso-scale: UA-Shanghai was partitioned into three terrain-based subregions; and (3) city-scale: UA-Shanghai was partitioned into six cities based on administrative boundaries. All three scales yielded simulations averaging about 87% accuracy with an average Figure-of-Merit (FOM) of about 32%. Overall accuracy was reduced from calibration and validation. The regional-scale model yielded less accurate simulations as compared with the meso- and city-scales for both calibration and validation. Simulation success in different subregions is independent at the city-scale, when compared with regional- and meso-scale. Our observations indicate that observation scale is important in CA modeling and that smaller scales probably lead to more accurate simulations. We suggest smaller partitions, smaller observation scales and the construction of one CA model for each subregion to better reflect spatial variability and to produce more reliable simulations. This approach should be especially useful for large-scale areas such as huge urban agglomerations and entire nations.

    Description: Ecosystem services (ESs) in rapidly urbanizing agricultural basins are vulnerable to environmental changes. Adequately understanding the driving forces and the dynamics of ESs related to water quantity and quality can provide a basis for making sound management decisions on the development of basins. Here, we explored the impacts of future land use and climate changes on four ESs: nitrogen and phosphorous purification, water supply, and soil retention services in the Taihu Basin region of eastern China. Spatially explicit methods, a cellular automata-Markov (CA-Markov) model and the delta downscaling method were used to quantify the ESs, simulate land use changes, and project future climate changes, respectively. We built a business-as-usual land use scenario, representative concentration pathways (RCPs) scenarios for climate change, as well as a combined land use and climate change scenario to analyze the changes in the drivers and the responses of ESs. The results showed the following: (1) future land use changes would significantly enhance the nitrogen purification service while reducing the phosphorus purification service compared to other services; (2) climate change would have substantial effects on water supply and soil retention, but these impacts would vary with different RCPs scenarios during three future periods; and (3) the combined scenarios of both drivers would obviously influence all ESs and lead to a nitrogen purification service that was different from the other three services. Moreover, the policy implications of the results were discussed. The findings can help guide the creation of policies for land structure and patterns, climate change adaptation, and ecosystem-based management to promote the sustainable development of watersheds at the regional scale.

    Description: Previous studies suggest that vegetable cultivation increases soil organic carbon (SOC) storage. However, how stable the SOC increment is, and how greenhouse cultivation contributes to the SOC increment in terms of quantity and stability, remains unclear. Soil samples were taken from three typical vegetable cultivation pattern fields: open field (OF), seasonal greenhouse (SG), and permanent greenhouse (PG), as well as adjacent non-vegetable fields. Three conceptual SOC pools, including active (Ca), slow (Cs), and resistant (Cr) pools were fractionated to evaluate SOC sequestration and its stability in vegetable cultivation. The results indicate that vegetable cultivation is associated with greater stored SOC compared with non-vegetable cultivation (SOC increased by 57.9% on average). Using non-vegetable fields as a reference, SOC increments by vegetable cultivation were associated with a higher proportion of Ca (3.7–6.6%) than the reference fields (1.0–2.0%), indicating that the SOC increments might be easily decomposed. Among the three vegetable cultivation patterns, SG, with a higher increase in Cr, is recommended due to its relatively more stable SOC sequestration. Overall, vegetable cultivation could enhance the quantity of SOC, but the stability of the SOC increment is affected by the vegetable cultivation pattern.

    Description: The diagnosis of damage in a bridge superstructure using quasi-static strain influence lines (ILs) is promising. However, it is challenging to accurately localize the damage in a bridge superstructure due to limited numbers of strain IL measurement points and inconsistencies between the loading conditions before and after damage. To address the above issues, the Brillouin optical time domain analysis (BOTDA) technique is first applied to bridge damage localization using quasi-static strain ILs, and the number of strain IL measurement points is substantially increased. Additionally, a damage localization index based on quasi-static strain ILs that is independent of differences in the loading conditions before and after damage is proposed to localize damage in the superstructure of a beam bridge. Finally, the effectiveness of the proposed method is demonstrated through both numerical analysis and measured data from a quasi-static test of a model bridge.
