    Notes: Electrophoretic separation of the water-soluble proteins from seven Acanthamoeba strains, followed by lactic dehydrogenase (LDH) visualization by the zymogram method, has indicated a single LDH activity in each extract. To account for this observation, three alternatives are considered: inadequate electrophoretic resolution, two gene-encodement of LDH with constraints relating to subunit assembly or oligomer stability, and one gene-LDH encodement.

    Notes: The feasibility of determining the ground-water contribution to stream flow during periods of storm runoff by continuous monitoring of a stream's electrical conductance was investigated. Methods involving graphical techniques or ground-water stage versus base flow rating curves are generally used to determine the ground-water contribution, but the ground-water contribution can also be estimated if stream flow and the conductance of surface water and of ground water are known. The conductance method was tested on a 95-square-mile basin in north-central Illinois. Continuous records of stream flow, stream-flow conductance, and ground-water stage within the basin were collected. A rating curve of base flow versus mean ground-water stage was constructed, and ground-water discharges determined from the rating curve were compared with those computed from conductance data. The conductance method normally gave a lower estimate of the ground-water contribution than did the rating-curve method. However, analyses of storms which were preceded by extended dry periods resulted in much lower estimates of ground-water discharge by the rating-curve method than by the conductance method. The conductance method offers a simple alternative for use in areas where observation wells are unavailable or where rating curves are difficult to construct.

    Notes: Illinois aquifers furnish approximately 233 mgd (10.2 m3/s) of water to 677 public-water supplies outside the six-county area of northeastern Illinois. Ground water is usually obtained from sand-and-gravel deposits in the glacial drift or from limestone or sandstone formations in the underlying bedrock. The most favorable ground-water conditions are found in the northern third and the southern tip of the State, while, elsewhere, major aquifers are sand-and-gravel deposits of the Mississippi, Illinois, buried Mahomet, Wabash, Ohio, Kaskaskia, and Embarrass valleys.A brief review was made of data and information in the State Water Survey files for each public ground-water supply, and an assessment was given as adequate, marginal, or deficient, in terms of present demands. Twenty-four supplies were studied in greater detail, including calculations of aquifer sustained yields. The study indicated that 39 supplies were marginal and four were judged deficient in meeting current demands. The majority of the marginal and deficient supplies are located in the central third of the State; but so are most of the supplies (outside of north-eastern Illinois).The study represents the first of a three-part plan to: (1) define problem areas and determine priorities for studies in greater detail, (2) conduct regional studies, including test drilling, in problem areas to determine how great the water resource is, and (3) determine the water resource alternatives available to public ground-water supplies that are found to be inadequate.

    Notes: In 1984, the Illinois Department of Energy and Natural Resources was required to assess the regulations and practices of the Illinois Underground Injection Control (UIC) program as it relates to Class I hazardous waste disposal wells. Nine injection wells, including two standbys (one inactive), are currently in operation at seven sites in the state. These wells range in depth from 1540 to 5524 feet (470 to 1683m; most inject wastes into porous carbonate formations (two wells inject into a thick sandstone). In 1984, approximately 300 million gallons (1.1 billion liters) of industrial wastes were disposed of in these wells. Acids were the most common waste disposed of, although water made up 70 to 95 percent of the wastes by volume. Illinois has been granted primacy in operating this program.The geologic environment, consisting of the unit accepting the waste and confining units lying above and below, has the capacity to accept the waste, to retain it, and to protect all underground sources of drinking water (USDW) from contamination by its injection. The geology of Illinois is relatively simple and includes disposal zones and associated confining units suitable for deep-well injection across the central two-thirds of the state.The regulatory structure for Class I injection wells is generally adequate in concept and scope to ensure containment of injected wastes and to safeguard underground sources of drinking water in Illinois. There is a need to update and strengthen selected portions of the regulatory practices in the areas of waste sampling protocol, chemical analysis of collected waste samples, and evaluation of injection well testing and monitoring data.A number of technologies exist that can treat and dispose of most hazardous and non-hazardous waste streams. Each of these technologies has associated with it economic, environmental and societal impacts.

    Notes: Overpumping of the Cambrian-Ordovician aquifer in the Chicago area has caused severe water-level declines in portions of Cook, DuPage, Kane, and Will Counties. Recent changes in the accounting procedure for diversion of Lake Michigan water have released more water for public supplies. As communities currently withdrawing water from deep wells receive allocations of Lake Michigan water, deep pumpage will decrease in those areas and water levels will partially recover. Major cones of depression will shift southward to Joliet and westward to the Fox River communities north of Aurora. Critically low water levels caused by pumpage in these areas will result in a loss of pumping capacity of as much as 19% by the year 2020. Despite lake allocations, deep pumpage will grow again and will still exceed the practical sustained yield (65 mgd/2.46 × 105 m3/day) of the aquifer. Continued growth in Wisconsin pumpage will also contribute an estimated additional decline of about 80 feet near the State line between 1980 and 2020.

    Notes: A spatial stochastic approach was adopted to estimate the capture zones of a welifield in eastern Netherlands near the town of Wierden. The main sources of uncertainty were found to be the hydraulic conductivity of the production aquifer and the spatial distribution and thickness of an impeding clay layer situated above the production aquifer. Both were modeled as random space functions and a Monte Carlo analysis was performed to assess the probability distributions of the capture zones for t = 25 years and t →∞. The distributions were compared with two deterministic capture zones to show how the stochastic approach preserves the discontinuous character of the impeding clay layer while the deterministic approach smooths out the discontinuities. Furthermore, it is shown how the stochastic approach reveals the uncertainty about the location of the capture zones while the deterministic approach obscures it.

    Notes: Abstract A fragment from Bacillus acidopullulyticus strain 294-16 encoding a pullunase activity has been cloned into Bacillus subtilis. The nucleotide sequence of the 3972 base pairs (bp) fragment has been determined and shown to include only one complete open reading frame (ORF) of 863 codons. The deduced amino acid sequence of this ORF, denoted pulB, shows homology to a number of amylolytic enzymes. Primary and secondary structure analysis indicates that the central region of the protein forms the catalytic domain in a characteristics (β/α)8 barrel. Three carboxylic acid residues essential for catalysis were identified. Regions within the catalytic domain proposed to be involved in substrate binding have been identified by homology.
