    Notes: A new electro-optically modulated optical solid-state (MOSS) interferometer has been constructed for the measurement of the low order spectral moments of line emission from optically thin radiant media. The instrument, which is based on the principle of the Fourier transform spectrometer, is rugged, compact, and inexpensive and offers a number of advantages over conventional grating based spectrometers. Most importantly, by employing electro-optical path-length modulation techniques, the spectral information is obtained using a single photomultiplier tube. Specifically, the zeroth moment (brightness) is given by the average signal level, the first moment (shift) by the modulation phase and the second moment (line width) by the modulation amplitude. In applications such as Zeeman spectroscopy and motional Stark effect (MSE), polarization modulation can be used to effect a modulation of the center frequency and/or coherence of the light which is then measured by the MOSS spectrometer. This article describes a number of applications, including Doppler and charge exchange recombination spectroscopy and polarization spectroscopy (Zeeman and MSE) for which the MOSS spectrometer is an inexpensive and powerful alternative to multichannel grating spectrometers. © 1999 American Institute of Physics.

    Notes: A quadrature polarimeter for measurement of plasma magnetic field and electron densities is described. The polarization ellipse tilt angle is encoded as a relative phase shift between orthogonally polarized beams. This phase can be recovered using standard quadrature interferometric methods. A reference wave is used to generate clock pulses for a digitizing instrument that measures directly the in-phase and quadrature components of the modulation.     Notes: Diffraction from plasma density irregularities can have significant consequences for the tomographic inversion of line-of-sight integrated measurements of the phase disturbance suffered by a probing beam. A new tomographic approach which takes account of diffraction from high-K refractive index structures is described. The relevance for multichannel interferometry measurements is demonstrated numerically and the technique is applied to the inversion of multichannel TEXT interferometer data.

    Notes: We report the successful construction and operation of a far-infrared (337 μm) interferometer based on a multiple-order diffraction grating wheel. On reflection from the wheel circumference, the incident laser beam is diffracted into a number (∼12) of approximately equal intensity high-order beams. Since the Doppler offset of the beams is proportional to the diffraction order, information encoded by the spatially distinct beams is multiplexed in the frequency domain. This allows simultaneous multichannel phase shift information to be obtained using only a single detector.

    Notes: A novel multichannel scanning interferometer designed for tomographically inferring contours of electron density in magnetically confined plasmas is described. The scanning element is a multisectored blazed rotating grating. The diffracted beam emerges at a different angle from each sector, giving rise to a fan array of discrete beams for each rotation of the grating. Signals from the probing chords are multiplexed in time enabling the use of a single detecting element for the extraction of many channels of line-integrated density information. An air-turbine-driven grating wheel assembly has been fabricated and initial tests performed. The proposed interferometer is to operate at 337 μm and will be installed on the H-1 helical axis stellarator currently under construction at the Australian National University.

    Notes: We have developed a three-dimensional Gaussian-beam ray-tracing program to aid in the design of infrared, far-infrared, and millimeter wave interferometer and polarimeter diagnostic systems for magnetic confinement fusion relevant plasma physics experiments. An overview of the program is presented along with a description of the ray-tracing algorithm. A model is developed for the case of diffraction of a Gaussian beam off a cylindrical grating and is shown to be in good agreement with experimental measurements. The program has been used to aid the design of the scanning-grating interferometer system for the H-1NF heliac experimental plasma device. The program is written in the Research Systems Inc. Interactive Data Language and, on a typical modern personal computer, is able to trace and render the ∼50 element three-view 44-beam H-1NF interferometer optical system in about one minute. © 2001 American Institute of Physics.

    Notes: Abstract “Planning” occurs as a part of governmental operations wherever “decision-making” happens. For US metropolitan areas, the locuses of decision-making are multiple, ranging in scale from very small jurisdictions up through the hierarchy to state and federal levels, and in function from general governments to many special-purpose agencies — transportation, health care, education, etc. Almost all might affect or be affected by urban transportation decisions and actions. Since no one of these units of government is comprehensive in authority and activity, there is no single, centralized planning operation that is truly “comprehensive.” “Pluralistic planning” is increasingly trying to foresee and to accommodate the interactions among the various levels and functions. Instead of fragmenting, with the fragments pulled apart and insulated, we need to move toward partitioning, not merely to delimit boundaries but also to identify interfaces. This movement is hampered by the differentials in the development of the “state-of-the-art” of the technical planning process now used by the several levels and functional units of government. This is most advanced, and most effective, for small, homogeneous suburban jurisdictions primarily concerned with guiding and controlling physical development; it is in disarray in central cities trying to cope with social and economic problems as well as with physical deterioration; at the metropolitan scale it is highly developed technically but not very influential. There is a trend toward a network of planning activities that recognizes and facilitates interrelationships and interactions, both vertically among functional boundaries and horizontally across geographical-scale distinctions — a trend toward the “comprehensive” —but we have a very long way to go.

    Notes: Abstract Transgenic tobacco plants and progeny carrying coding sequences for neomycin phosphotransferase II (NPTII) and beta-glucuronidase (GUS) were recovered following microprojectile bombardment of tobacco leaves. Transgenic plants were regenerated from bombarded leaf pieces of tobacco cvs. ‘Xanthi’ and ‘Ky 17’ which were cultured in the presence of 100 or 200 μg/ml kanamycin for six to eight weeks. Among 160 putative transgenic plants from at least 16 independent transformation events 76% expressed NPTII, and 50% expressed GUS. Southern analysis of plants expressing either one or both of the enzymes indicated DNA in high molecular weight DNA in 8 of 9 independent transformants analyzed. Two independent transformants and their progeny were analyzed in detail. Analysis of progeny for quantitative enzyme levels of NPTII and GUS, and Southern analysis of parents and progeny clearly demonstrated that the genes were transmitted to progeny. One transformant demonstrated Mendelian ratios for seed germination on kanamycin-containing medium while the other transformant had non-Mendelian ratios. DNA analysis of progeny indicate complex integration of the plasmid DNA, and suggest that rearrangements of this DNA has occurred. These results are consistent with other methods of direct DNA uptake into cells, and verify that the microprojectile bombardment method is capable of DNA delivery into intact plant cells which can give rise to transgenic plants and progeny.

    Notes: Abstract We have generated transgenic maize seed containing β-glucuronidase(GUS) for commercial production. While many other investigators have demonstrated the expression of GUS as a scoreable marker, this is one of the first cases where a detailed characterization of the transgenic plants and the protein were performed which are necessary to use this as a commercial source of GUS. The recombinant β-glucuronidase was expressed at levels up to 0.7% of water-soluble protein from populations of dry seed, representing one of the highest levels of heterologous proteins reported for maize. Southern blot analysis revealed that one copy of the gene was present in the transformant with the highest level of expression. In seeds, the majority of recombinant protein was present in the embryo, and subcellular localization indicated that the protein was dispersed throughout the cytoplasm. The purified recombinant β-glucuronidase (GUS) was compared to native β-glucuronidase using SDS-PAGE and western blot analysis. The molecular mass of both the recombinant and native enzymes was 68 000 Da. N-terminal amino acid sequence of the recombinant protein was similar to the sequence predicted from the cloned Escherichia coli gene except that the initial methionine was cleaved from the recombinant GUS. The recombinant and native GUS proteins had isoelectric points (pI) from 4.8 to 5.0. The purified proteins were stable for 30 min at 25, 37, and 50 ° C. Kinetic analysis of the recombinant and native GUS enzymes using 4-methylumbelliferyl glucuronide (MUG) as the substrate was performed. Scatchard analysis of these data demonstrated that the recombinant enzyme had a Km of 0.20 mM and a Vmax of 0.29 mM MUG per hour, and the native enzyme had a Km and Vmax of 0.21 mM and 0.22 mM/h respectively. Using D-saccharic acid 1,4-lactone, which is an inhibitor of β-glucuronidase, the Ki of the native and recombinant enzymes was determined to be 0.13 mM. Thus, these data demonstrate that recombinant GUS is functionally equivalent to native GUS. We have demonstrated the expression of high levels of GUS can be maintained in stable germlines and have used an efficient recovery system where the final protein product, GUS, has been successfully purified. We describe one of the first model systems for the commercial production of a foreign protein which relies on plants as the bioreactor.
