    Notes: A balloon-borne mass spectrometer system has been flown successfully to determine the chemical composition of polar stratospheric aerosols over northern Scandinavia. The experiment combines an aerodynamic lens which collimates the aerosols into a narrow beam, a small sphere in which they evaporate, and a mass spectrometer for gas analysis. High-speed differential pumping by two liquid helium pumps effectively lowers the presence of ambient gases without affecting the particles of the beam. Field measurements and aerosol studies inside a large cryo-chamber have shown that the concept of particle focusing, evaporation and subsequent mass spectrometric gas analysis is a reliable technique to determine the molecular composition of aerosols especially in polar stratospheric clouds. © 2002 American Institute of Physics.

    Notes: The adsorption dynamics of water on NaCl(100) is studied by molecular dynamics calculations as a function of coverage. We find that, starting from a critical coverage of about 1/2 monolayer, a coupling of the water dipoles sets in and the interaction between the water molecules wins over the interaction between adsorbate and substrate leading via percolation to formation of infinite cluster networks. This effect is confirmed qualitatively by surface sensitive optical second harmonic measurements with well-controlled water exposure. At a coverage of one bilayer, a two-dimensional "ice'' structure is found to be stable. Simulated low-energy electron diffraction (LEED) patterns for this configuration are in excellent agreement with recent observation of a c(4×2) overstructure.

    Notes: Atomic-force pictures reveal a heterogeneous microstructure at the surface of glassy layers which should be similar to one of the many equivalent microstructures a liquid is running through. These microstructures are described with the aid of a kinetic model of reversible aggregation which goes back to formulations as used in the description of living polymerization or aggregation of polymers in solution. Aggregates are considered as dynamic subsystems wherein collective modes of motions are excited. Fluctuations of the aggregates, densely packed in a disordered pattern, leads to a broad size distribution which happens to be controlled by Boltzmann's factor. The disordered structure within the aggregates themselves is optimized, reduced aggregate energy and entropy should be equal. Symmetries are deduced which elucidate many universal properties of the dynamic microstructure of liquids. Thermodynamic properties like the specific heats of aggregation in liquids or the dependence of the glass transition of homologues of linear atactic polystyrene are consistently described. © 1997 American Institute of Physics.

    Notes: Characteristics are presented for Yersiniu enterocoliticu-like organisms isolated from vacuum-packaged beef and lamb stored for 21-35 days at l-3°C. Isolation of this organism was more frequent after 28 days of storage under vacuum conditions than under nonvacuum conditions (leaker packages). A higher incidence of isolates was obtained from cuts packaged under high vacuum conditions.

    Notes: A comparison of three relatively rapid methods of determining free fatty acids concentrations in cheese was performed by correlating the results with a more laborious but accurate gas chromatographic (GC) technique. Acid Degree Values were found to correlate poorly with the GC data, particularly for short chain fatty acids. Values obtained using a Copper Soap Method correlated closely with the GC data for total and long chain fatty acids, but did not correlate as well with short chain fatty acids levels. The Extraction-Titration Method was found to be a simple and reliable technique that yielded values which correlate closely with short chain fatty acid concentrations.

    Notes: A qualitative screening revealed the occurrence of lipase, esterase, protease, amylase, endo-1,4-β-D-glucanase, xylanase, pectinmethylesterase, polygalacturonase, catalase, β-D-glucosidase and β-D-galactosidase activities in the technical Aspergillus niger enzyme under study (Lipase 2212 D, Röhm). The isolation and purification of lipolytic activities were performed by combination of DEAE-Trisacryl M ion exchange chromatography, Sephadex G 50 gel filtration and hydrophobic chromatography using Phenylsepharose CL-4B. The individual purification steps were checked by specific enzyme visualization in ultrathin agar gels after ultrathin-layer isoelectric focusing (UIEF). Two UIEF homogeneous lipase isoenzymes (I and II) were isolated and characterized by the following parameters: isoelectric points (I: 4.0; II. 3.5); molecular weights (I: 31000 daltons; II: 19000 daltons); carbohydrate contents (I: 6%; II: 9%) and compositions; pH optima (I, II: 5-6); substrate specificities and various effectors.

    Notes: : In the past, deliberate contamination of foods in the U.S. has been committed by individual criminals, disgruntled employees, or political activists with a narrow agenda. After September 11, 2001, every nation has had to consider a much wider range of food security issues. Food is a global commodity and nearly every country both exports and imports foods. Every nation must consider the security of its domestic production and look beyond its borders to its trading partners to assure the safety of its food supply. In the nearly 2 years since September 11, 2001, many steps have been taken to improve food security. New legislation has been enacted to strengthen the ability of government to respond to terrorist threats. New relationships have been forged between food security agencies, law enforcement, and the intelligence community. Virtually every segment of the domestic and imported food supply has come under scrutiny in an effort to identify points of weakness and appropriate protective measures. Gaps in our knowledge about specific agents and food processes have been identified and research to fill these gaps has been initiated. This effort will continue for the foreseeable future and could have a wide range of benefits, including improvements to food safety, a reduction in product counterfeiting, and a reduction in the “gray market” food trade. The scale and diversity of the U.S. food supply makes it an attractive target, but at the same time makes it very resilient and responsive to emerging threats.

    Notes: Uptake of [15N]-ammonia was more sensitive to UV-B exposure than the total 14CO2 fixation rate of Lithodesmium variabile Takano. Short-term UV-B radiation (15 min) had practically no effect on the kinetics of [15N]-ammonia, whereas there was an effect on [14C]-bicarbonate uptake rate. A significant reduction was found after 30 and 60 min UV-B stress. The time course of photosynthetic uptake of 15NH4Cl at several wavelengths was markedly depressed at shorter wavelengths (irradiation with WG 280). A short-term (11 min) exposure to ultraviolet radiation had no influence on the [14C]-labeled photosynthetic products. However, the [15N]-label of several amino acids and the ratio of [15N]-glutamine to [15N]-glutamic acid varied after irradiation with different ultraviolet wavebands. The results are discussed with reference to UV damage to the key enzymes of nitrogen metabolism.

    Notes: HIV encephalitis is the common pathologic correlate of HIV-dementia (HAD). HIV-infected brain mononuclear phagocytes (MP) (macrophages and microglia) are reservoirs for persistent viral infection. When activated, MP contribute to neuronal damage. Such activated and virus-infected macrophages secrete cellular and viral factors, triggering neural destructive immune responses. Our Center's laboratories have begun to decipher the molecular and biochemical pathways for MP-mediated neuronal damage in HAD. This review will discuss the salient clinical and pathological features of HAD and highlight the recent advances made, by our scientists and elsewhere, in unraveling disease mechanisms, including the role of chemokines and their receptors in the neuropathogenesis of HIV-1 encephalitis.
