    Notes: Cleavage and blastoderm formation in Coelopa frigida are extremely rapid developmental processes. In short (6-7 minutes) successive cell cycles, nuclei multiply and spread out through the egg. The movement seems to be aided by endoplasmic vesicles and cisternae which are in direct contact with the nuclear membrane. The first cells to separate from the egg plasmodium in early superficial cleavage stages are the pole cells.Precursor material from multivesicular bodies forms the pole cell membranes. The primary nuclei from the posterior pole region are removed from the blastoderm by the pole cell segregation. Blastoderm nuclei from the regions adjacent to the posterior pole migrate into the residual periplasm after pole cell segregation has been completed and constitute the blastoderm nuclei in that region of the egg. Nucleoli are not revealed during internal cleavage. They appear in pole cells shortly after their segregation. The generation time of the blastoderm nuclei increases after the twelfth cleavage. Concurrently, nucleoli form in the blastoderm nuclei and permanent cell membranes separate individual blastoderm cells. After blastoderm cells have been separated from each other, they remain in contact with the interior yolk sac by means of cytoplasmic canals. This contact is maintained at least during the early phases of blastokinesis. Observations on nuclear migration and rapid membrane formation are discussed as examples of protein assembly from subunits as an alternative to de novo protein synthesis in early stages of development.

    Notes: ---Solid-phase extraction, on-line derivatization, and measurement by ion trap mass spectrometry (ITD-MS) were used to investigate the biological degradation of pharmaceutical residues (clofibric acid, ibuprofen, diclofenac). The results of the single steps of sample pretreatment and analytical determination are reported. MS/MS measurements were performed on an ITD-MS by selecting collision induced dissociation of the molecular ions (M+) as parent ions to defined daughter ions. A pilot sewage plant and biofilm reactors operating under oxic and anoxic conditions were run as model systems with synthetic sewage water containing 10 to 50 mg/L dissolved organic carbon (DOC) and pharmaceuticals in concentrations of 10 μg/L. Clofibric acid displayed its persistent character in all cases. The pilot sewage plant and the oxic biofilm reactor showed comparable results for diclofenac and ibuprofen, which both were partly degraded. These results can explain the occurrence of these substances in sewage effluents and in the aquatic environment. A high degree of degradation was found especially for ibuprofen in the oxic biofilm reactor, which was attributed to adaptation of the biofilm to the residue. Two metabolites of ibuprofen could be identified on the basis of their mass spectra and comparison with literature data, viz. hydroxyibuprofen and carboxyibuprofen.

    Notes: Very good separations of non-ionic organic compounds have been achieved by capillary zone electrophoresis (CZE) using a tetrahep-tylammonium salt as an additive in aqueous acetonitrile as solvent. A systematic study was undertaken to determine the effect of experimental parameters on electroosmotic mobility and electro-phoretic mobility. It was found that pH, acetonitrile concentration, and the type and concentration of quaternary ammonium salt are important experimental variables. Under appropriate conditions, the separation window was enlarged and a broad range of electrically neutral organics, including very hydrophobic compounds (e. g. poly-cyclic aromatic hydrocarbons) and fairly hydrophilic compounds were separated in a relatively short time. By adjusting the separation conditions, high resolution CZE of a specific group of neutral organic compounds could be achieved. A method for calculation of capacity factor was proposed and capacity factors for a variety of non-lonic organic compounds were calculated.

    Notes: A molecular sieve known as Silicalite was used as the column packing for HPLC. Silicalite contains channels (or cavities) approximately 6 Å in diameter but, unlike most other molecular sieves, Silicalite is hydrophobic. The retention times of methyl ketones and substituted phenols containing n-alkyl groups increase with increasing chain length of the substituent. However, phenols with very bulky substituents appear to be excluded from the Silicalite channels and elute very quickly. Excellent separations were obtained for a number of compounds with only slight differences in chemical structure. These include phenol isomers with a primary- or secondary alkyl group, position isomers of substituted phenols, and aliphatic cis-trans isomers.

    Notes: Methods for the detectionn of metal cations under acidic conditions, near PH 2, in capillary electrophoresis (CE) were investigatged. Conditions for direct UV detection of UV absorbing metal cations such as Cr3+, Cu2+, Fe3+, UO22+, VO2+, and VO2+ were established With aqueous HCl or HClO4 as the electroyte carrier. The speciation of vanadium(IV) and vanadium(V) at PH 2.3 by CE was achieved with direct detection at 185 nm. With the strong absorbance at 185 nm, no complexation was needed to detect the metal cations. An indirect UV detrection scheme for acidic conditions was also investigated. Several background carrier electolytes (BCES) were studied including 4-methylbenzylamince, nicontinamide, pyridazine, guanidine, 3-picoline, and chromium (III) to determine their effectivencess under very acidic conditions. The efect of ioni c surfactants and the nonionic surfactant, Trition X-100, on the peak heights and N Values was also studied.

    Notes: Smooth muscle cells from spontaneously hypertensive rats (SHR) elaborated extracellular matrix (ECM) material in culture that was more stimulatory to growth of cells from normotensive (WKY) animals than their own matrix. Both cell types elaborated ECMs consisting of glycoconjugate material (proteoglycans, glycopeptides) elastin, and collagens, but there were differences in the relative proportions of the compounds synthesized. Cells from SHR produced an ECM richer in elastin than that synthesized by WKY derived cells (∼19% vs. 11%, respectively). However, the latter elaborated ECMs containing more (∼45% for WKY vs. 29% for SHR) glycoconjugate material than the former. The lysyloxidase-mediated crosslinking of elastin was more rapid in cultured cells from SHR animals than from their normotensive counterparts and may be as a consequence of increased substrate (tropoelastin) availability in ECMs from SHR animals. The relative proportions and sulphate levels of the glycosaminoglycans associated with matrix material elaborated by the two cell types were similar. Radiolabelled glycoconjugate material was degraded by cells (SHR/WKY) when they were plated upon pre-formed ECMs, and their patterns of synthesis of new matrix was markedly altered under such conditions. New matrix material elaborated by cells plated upon ECM-coated dishes consisted predominantly of glycopeptide and proteoglycan matrix components. Epidermal growth factor promoted the incorporation of [3H]-thymidine into DNA by quiescent cells, and this was also markedly stimulated when cells were plated onto ECM-coated plasticware rather than onto plastic substratum.
