    Notes: Abstract All archaeal histones studied to date have similar lengths, 66 to 69 amino acid residues that form three α-helices separated by two β-strand loop regions which together constitute a histone fold. In contrast, the eukaryal nucleosome core histones are larger, 102 to 135 residues in length, with N-terminal and C-terminal extensions flanking the histone fold that participate in gene regulation and higher-order chromatin assembly. In the Methanococcus jannaschii genome, MJ1647 was annotated as an open reading frame predicted to encode an archaeal histone with an approximately 27-amino-acid C-terminal extension, and we here document the DNA binding and assembly properties and thermodynamic stability parameters of the recombinant product of MJ1647 synthesized in Escherichia coli with (rMJ1647) and without (rMJ1647Δ) the C-terminal extension. The presence of the C-terminal extension did not prevent homodimer formation or inhibit DNA binding, but the complexes formed by rMJ1647, presumably archaeal nucleosomes containing a (rMJ1647)4 tetramer, were apparently less stable than those formed by (rMJ1647Δ)4. The presence of the C-terminal extension increased the thermostability of rMJ1647 when compared with rMJ1647Δ in 0.2 M KCl at pH 4 but not in the absence of KCl at pH 1. Based on thermal unfolding transitions, rMJ1647 and rHAfB generated by expression of AF0337 cloned from the genome of the related hyperthermophile Archaeoglobus fulgidus in E. coli were found to have higher thermodynamic stabilities than all previously studied archaeal histones.

    Notes: Abstract Phylogenetic analysis of conservative nucleotide substitutions in 18 complete sequences of the mitochondrial cytochrome β gene of Phocidae (true seals), Odobenidae (walruses), and Otariidae (sea lions and fur seals), plus three ursid and three felid sequences, identified the pinnipeds as monophyletic with Otariidae and Odobenidae on a common evolutionary branch. Analysis of total nucleotide differences separated the evolutionary lineages of northern and southern phocids. Both lineages are distinct from the most ancestral phocid genus, Monachus (monk seals), represented by the Hawaiian monk seal. The inclusion of the Hawaiian monk seal in the subfamily Monachinae makes the subfamily paraphyletic. Among the northern phocids, the hooded seal (genus Cystophora, chromosome number 2n = 34) is sister taxon to the Phoca complex. The Phoca complex, which is characterized by the chromosome number 2n = 32, includes genus Phoca and the monotypic genus Halichoerus (grey seal). The comparison does not support a generic distinction of Halichoerus within the Phoca complex. The present data suggest that Cystophora and Phoca separated ⩾6 million years ago. Among the southern phocids the close molecular relationship of the Weddell and leopard seals relative to their morphological distinction exemplifies rapid adaptation to different ecological niches. This result stands in contrast to the limited morphological differentiation relative to the pronounced molecular distinctions that may occur within the Phoca complex.

    Notes: Abstract The components of trail-laying and trailfollowing behaviour in the harvester termitesHodotermes mossambicus (Hagen) andTrinervitermes trinervoides (Sjöstedt) were investigated. In both species the trails are laid by pressing the third to sixth abdominal segments against the substratum. The trails are detected by olfactory receptors which are found on the antennae—principally on the distal one third.H. mossambicus trails andT. trinervoides trails laid on paper persisted for approximately 90 and 20 minutes respectively at 23° C±1° C. T. trinvervoides trail substances were steam volatile, but steam distillates ofH. mossambicus individuals were inactive. Ethanol or diethyl ether extracts ofH. mossambicus were, however, active. The volatile components of the secretions of the nasuteT. trinervoides soldiers were collected by drawing an air shtream over fighting colonies and passing it through gas scrubbing bottles containing appropriate solvents.

    Notes: Abstract The control of caste formation in the harvester termiteHodotermes mossambicus (Hagen) was investigated. Normal laboratory colonies contained a primary reproductive pair, between 6.59 and 24.98% workers, 0.39 and 3.45% soldiers and 74.64 and 92.06% larvae. Supplementary reproductives did not develop in colonies deprived of their king and queen. Continuous removal of soldiers stimulated soldier production. We have suggested that a pheromone produced by the soldiers plays a major role in controlling the soldier population of a colony. A similar phermone system appreared to control the production of workers. Evidence was also obtained that the primary reproductive pair influenced the production of workers and soldiers.

    Notes: Abstract Rates of shoreline change and overwash penetration distances were calculated for barrier islands along the Louisiana, Mississippi, and Alabama coasts with the orthogonal grid mapping system (OGMS). Average rates of shoreline change are exceptionally high in Louisiana, being of the order −4.7 to −7.4 m yr−1. Mississippi and Alabama recession rates are lower and range from −2.0 to −3.1 m yr−1 over the period of record. Erosion rates along the shorelines of these islands have remained relatively constant over the period of study with five exceptions in coastal Louisiana and the Chandeleur-Breton Islands Arc, and two exceptions along the Mississippi-Alabama barrier islands where they have accelerated. Mean overwash penetration is greatest along Dauphin Island, Alabama, and Cat Island, Mississippi: 207.6 and 197.9 m, respectively. The Chandeleur-Brenton Islands Arc range from 88.1 m at the central barrier to 180.4 along the flanks. The Mississippi islands range from 105.2 m on Ship Island to 200.5 m along central Horn Island. Mean overwash penetration along the Louisiana barriers is highly variable: 46.3 to 211.4 m.

    Notes: Summary Analysis of cloned SalI fragments coding for the 26S rRNA in wheat mitochondria (Triticumaestivum L.) shows that the 26S gene is contained within the same basic structural unit RS (which is about 6.6 kbp long). This repeated sequence is flanked by four different sequences I, II, III, IV in the orientations: I-RS-III, I-RS-IV, II-RS-III and II-RS-IV. This organization of the 26S gene, similar to the organization found for the 5S and 18S gene, suggests again that reciprocal intra-and/or intermolecular recombination occurs within these repeated sequences. S1 mapping and R loop mapping indicate that the 26S gene is about 3.5 kbp and has no intron.

    Notes: Summary Mitochondrial DNA from etiolated seedling of male-fertile (F) and male-sterile (S) sunflower (Helianthus annuus L.) has been investigated for the first time in an isogenic couple HA89. 1. The mt-DNAs isolated from the S and F cytoplasms give a sharp and symmetrical band at 1.706 g/ml in an analytical CsCI gradient ultracentrifugation. 2. In polyacrylamide gel it is found that mitochondria from F cytoplasm contain a Low-Molecular-Weight (LMW) mt-DNA molecule of 1.45 Kilobase-pairs (kb) in addition to the major mt-DNA. On the contrary, mitochondria from S cytoplasm contain no LMW molecule. Treatments with DNAse, RNAse and Nuclease S1 show that this LMW mt-DNA molecule consists of a supercoiled circular DNA. The length and form of the LMW molecule are confirmed by electron microscopy observations. 3. The mt-DNAs from S and F cytoplasms have also been investigated using restriction endonuclease digestions (Sa11, Xho1, Bg11). The electrophoresis of the resulting fragments reveals several differences between the S and F cytoplasm. It is not known whether the differences observed between F and S mt-DNA are correlated with the cytoplasmic male sterility phenotype.

    Notes: Abstract Because of its key role in the metabolism of glycerol during fermentation, 1,3-propanediol dehydrogenase (EC 1.1.1.202) of Enterobacter agglomerans CNCM 1210 was purified to homogeneity and studied with respect to its sensitivity to pH and to nucleotide and 1,3-propanediol concentrations. Enzyme activity was optimal at pH 7.8. The enzyme was competitively inhibited by NAD+ (Ki of 0.29 mM), and 1,3-propanediol exerted a strong inhibitory effect according to a mixed-type inhibition with a Ki of 13.7 mM and an a-factor of 9.0. It is proposed that these dehydrogenase properties be extended to the dehydrogenases of Citrobacter freundii and Klebsiella pneumoniae, which exhibited numerous similar physical properties.

    Notes: Summary Blood clotting factors were investigated in a patient with trisomy 8 mosaicism, a patient with an r(13), and a patient with distal trisomy 13q. Results are compatible with the assignment of the structural genes of factors VII and X to 13q34 and the existence of a regulatory mechanism associated with chromosome 8 controlling the expression of factor VII alone.
