Notes: SYNOPSIS.Under the growth conditions employed, the G1 macronucleus of Tetrahymena pyriformis HSM contains 7.4 × 10-12 g DNA, the G2 micronucleus 0.42 × 10-12 g. DNA content from the Tetrahymena thermophila macronucleus did not significantly differ from that of HSM, but the micronucleus contained about twice as much DNA as the micronucleus of the HSM cells. The T. thermophila macronucleus contained on average enough DNA for ˜ 35 haploid micronuclear copies. A new spreading technic allowed separation of macronuclear substructures from cells of late G2 to early G1. Photometric determination of DNA content of 345 individual structures suggested the existence of 5 different-sized macronuclear structures with a DNA content corresponding to 2, 4, 8, and 16 × the basic values. Comparison of the DNA content of these structures with (a) mitotic micronuclear chromosomes and (b) meiotic micronuclear chromosomes of T. thermophila cells suggests that the 5 basic values of macronuclear structures derive from structures of micronuclear chromosomes. The micronuclear chromosomes of T. pyriformis may be oligotenic. It is suggested that these results further our understanding of macronuclear organization.

Notes: Abstract. The large leaf rosettes of the so-called ‘giant rosette’ plants which inhabit the alpine region of tropical mountains are composed of a great number of adult leaves surrounding a central cone of developing leaves. Upon onset of the nocturnal frost period the adult leaves nyctinastically bend inwards and form a night-bud around the central leaf bud. The insulating effect of the night-bud was analysed in four species: Senecio keniodendron, Senecio brassica. Lobelia keniensis and Lobelia telekii which grow on Mt Kenya (Kenya). Freezing is avoided by a delay of cooling which is sufficient until rewarming by the next day's sunshine. A consequence of this delay is that the temperature in the nocturnal bud remains higher than that of the outer leaves which are often stiffly frozen after cold nights. Only one freezing point was detected on the leaf temperature recordings. Depending on the water state, the freezing points were in the range — 1°C to —4.2°C. Sucrose, amounting to 38% of the leaf dry weight, may act as a cryoprotectant for the cell membranes. Frost hardiness of the leaves, as determined with a laboratory method, was sufficient to enable the plants to survive during the nocturnal frost temperatures as measured in the field.

Notes: We identified by cloning and DNA sequence analysis the phosphate regulatory gene phoB of Bradyrhizobium japonicum. The deduced gene product displayed pronounced similarity to the PhoB protein of Sinorhizobium meliloti (71.4% identical amino acids), Escherichia coli (50.2%) and other bacterial species. Insertion of a kanamycin resistance cassette into phoB led to impaired growth of the B. japonicum mutant in media containing approximately 25 μM phosphate or less. A standard plant infection test using wild-type and phoB-defective B. japonicum strains showed that the phoB mutation had no effect on the symbiotic properties of B. japonicum with its soybean host plant.

Description / Table of Contents: Summary In 1984 and 1985 three pairs of wild Kestrels breeding in nest boxes on buildings were watched 10 752 hours during the reproductive season by using an automatic registering system. Each flight of the Kestrels from or to the nest could be determined and timed by photographing a clock simultaneously. Miniature cameras and Super-8-films were used. Advantages and disadventages of the cameras and of the automatic registering system are discussed. During the reproductive season the pellets of adult and juvenile Kestrels accumulate in the nest. Therefore the contents of the nest could be used for analyzing quality and quantity of food. An appropriate method had to be developed. Pellets of Kestrels living in cages contained 16,7 % of the mice resp. voles and 8,7 % of the birds fed. A comparison between the samples of photographic registering at the nest boxes and the nest contents revealed that 16,3 % of the mice resp. voles and 8,5 % of the birds brought by the adults could be found in the latter. These figures are identical with those from the caged Kestrels. Using ascertained factors of correction composition and quantity of prey can be calculated by the nest contents after the fledgings have left the nest.
