    Description: Background: Sequencing of the genome of Propionibacterium acnes produced a catalogue of genes many of which enable this organism to colonise skin and survive exposure to the elements. Despite this platform, there was little understanding of the gene regulation that gives rise to an organism that has a major impact on human health and wellbeing and causes infections beyond the skin. To address this situation, we have undertaken a genome--wide study of gene regulation using a combination of improved differential and global RNA-sequencing and an analytical approach that takes into account the inherent noise within the data. Results: We have produced nucleotide-resolution transcriptome maps that identify and differentiate sites of transcription initiation from sites of stable RNA processing and mRNA cleavage. Moreover, analysis of these maps provides strong evidence for 'pervasive' transcription and shows that contrary to initial indications it is not biased towards the production of antisense RNAs. In addition, the maps reveal an extensive array of riboswitches, leaderless mRNAs and small non-protein-coding RNAs alongside vegetative promoters and post-transcriptional events, which includes unusual tRNA processing. The identification of such features will inform models of complex gene regulation, as illustrated here for ribonucleotide reductases and a potential quorum-sensing, two-component system. Conclusions: The approach described here, which is transferable to any bacterial species, has produced a step increase in whole-cell knowledge of gene regulation in P. acnes. Continued expansion of our maps to include transcription associated with different growth conditions and genetic backgrounds will provide a new platform from which to computationally model the gene expression that determines the physiology of P. acnes and its role in human disease.

    Description: Recuperation of braking energy offers great potential for reducing energy consumption in urban rail transit systems. The present paper develops a new control strategy with variable threshold for wayside energy storage systems (ESSs), which uses the supercapacitor as the energy storage device. First, the paper analyzes the braking curve of the train and the V-I characteristics of the substation. Then, the current-voltage dual-loop control method is used for ESSs. Next, in order to achieve the best energy-saving effect, the paper discusses the selection principle of the charge and discharge threshold. This paper proposes a control strategy for wayside supercapacitors integrated with dynamic threshold adjustment control on the basis of avoiding the onboard braking chopper’s operation. The proposed control strategy is very useful for obtaining good performance, while not wasting any energy in the braking resistor. Therefore, the control strategy has been verified through simulations, and experimental tests, have been implemented on the Batong Line of Beijing subway using the 200 kW wayside supercapacitor energy storage prototype. The experimental results show that the proposed control is capable of saving energy and considerably reducing energy consumption in the braking resistor during train braking.

    Description: The installation of stationary super-capacitor energy storage system (ESS) in metro systems can recycle the vehicle braking energy and improve the pantograph voltage profile. This paper aims to optimize the energy management, location, and size of stationary super-capacitor ESSes simultaneously and obtain the best economic efficiency and voltage profile of metro systems. Firstly, the simulation platform of an urban rail power supply system, which includes trains and super-capacitor energy storage systems, is established. Then, two evaluation functions from the perspectives of economic efficiency and voltage drop compensation are put forward. Ultimately, a novel optimization method that combines genetic algorithms and a simulation platform of urban rail power supply system is proposed, which can obtain the best energy management strategy, location, and size for ESSes simultaneously. With actual parameters of a Chinese metro line applied in the simulation comparison, certain optimal scheme of ESSes’ energy management strategy, location, and size obtained by a novel optimization method can achieve much better performance of metro systems from the perspectives of two evaluation functions. The simulation result shows that with the increase of weight coefficient, the optimal energy management strategy, locations and size of ESSes appear certain regularities, and the best compromise between economic efficiency and voltage drop compensation can be obtained by a novel optimization method, which can provide a valuable reference to subway company.

    Description: The application of a stationary ultra-capacitor energy storage system (ESS) in urban rail transit allows for the recuperation of vehicle braking energy for increasing energy savings as well as for a better vehicle voltage profile. This paper aims to obtain the best energy savings and voltage profile by optimizing the location and size of ultra-capacitors. This paper firstly raises the optimization objective functions from the perspectives of energy savings, regenerative braking cancellation and installation cost, respectively. Then, proper mathematical models of the DC (direct current) traction power supply system are established to simulate the electrical load-flow of the traction supply network, and the optimization objections are evaluated in the example of a Chinese metro line. Ultimately, a methodology for optimal ultra-capacitor energy storage system locating and sizing is put forward based on the improved genetic algorithm. The optimized result shows that certain preferable and compromised schemes of ESSs’ location and size can be obtained, acting as a compromise between satisfying better energy savings, voltage profile and lower installation cost.

    Description: As the number of speech and video documents increases on the Internet and portable devices proliferate, speech summarization becomes increasingly essential. Relevant research in this domain has typically focused on broadcasts and news; however, the automatic summarization methods used in the past may not apply to other speech domains (e.g., speech in lectures). Therefore, this study explores the lecture speech domain. The features used in previous research were analyzed and suitable features were selected following experimentation; subsequently, a three-phase real-time speech summarizer for the learning of sustainability (RTSSLS) was proposed. Phase One involved selecting independent features (e.g., centrality, resemblance to the title, sentence length, term frequency, and thematic words) and calculating the independent feature scores; Phase Two involved calculating the dependent features, such as the position compared with the independent feature scores; and Phase Three involved comparing these feature scores to obtain weighted averages of the function-scores, determine the highest-scoring sentence, and provide a summary. In practical results, the accuracies of macro-average and micro-average for the RTSSLS were 70% and 73%, respectively. Therefore, using a RTSSLS can enable users to acquire key speech information for the learning of sustainability.

    Description: With its large capacity, the total urban rail transit energy consumption is very high; thus, energy saving operations are quite meaningful. The effective use of regenerative braking energy is the mainstream method for improving the efficiency of energy saving. This paper examines the optimization of train dwell time and builds a multiple train operation model for energy conservation of a power supply system. By changing the dwell time, the braking energy can be absorbed and utilized by other traction trains as efficiently as possible. The application of genetic algorithms is proposed for the optimization, based on the current schedule. Next, to validate the correctness and effectiveness of the optimization, a real case is studied. Actual data from the Beijing subway Yizhuang Line are employed to perform the simulation, and the results indicate that the optimization method of the dwell time is effective.

The hydrophobic image is firstly decomposed by the 2-level wavelet, along with the multi-Retinex algorithm in this paper. The processed low frequency sub-band and high frequency sub-band images are then reconstructed. The 3 × 3 Sobel operator is performed to measure the basic spatial gradient in four directions, including the horizontal direction, the diagonal direction, and then the vertical direction. The shape factor, the area ratio of the largest water droplet, and the coverage rate of the water droplet are selected as the feature parameters and input into the classification network that has been trained to do the hydrophobic level recognition. The effect of the different expansion speed on the desired learning results is discussed. The threshold plays a key role in image processing. Considering that the difference between the water droplet edge and the composite insulator surface is relatively small, the asymptotic semi-soft threshold function is used in pretreatment, whereas the adaptive two-dimensional Otsu’s method is used in image segmentation. The experimental results show that the proposed method has high recognition accuracy up to 94.8% for a diversity of images, and it is superior to the improved Shape Factor Method, the Multi-fractal Method, and the RBF Neural Network.

    Description: An intelligent tour service system including an augmented reality (AR) tour-sharing Application (APP) and a query-answering server was developed in this study to promote tourist attractions involving local Hakka culture in Thailand. Subsequently, use of this APP to navigate Hakka culture tourist attractions in Thailand was observed. The novel random neural networks (RNNs) were proposed to obtain query-answering services, and the practical experimental results showed that the accuracy of RNNs was 99.51%. This study also integrated the Technology Acceptance Model with Uses and Gratifications Theory to predict the gratification, usage intention, and user attitudes toward marketed attractions of the AR tour-sharing APP. A questionnaire survey was conducted, and 446 valid questionnaires were returned. The following results were obtained: (a) self-presentation and perceived usefulness (PU) directly influenced gratification; (b) perceived entertainment indirectly influenced gratification through perceived ease of use and PU, and information sharing indirectly influenced gratification through PU; and (c) gratification was significantly and positively related to usage intention and attitude toward attractions. Based on these results, suggestions that new technology marketing can be used to promote causes other than Hakka tourist attractions established in Thailand can be contrived. For example, the tour-sharing APP developed in this study could be applied to emphasize the characteristics of Thai Hakka culture; users’ fondness for self-presentation and information sharing can be used for word-of-mouth marketing to attract additional visitors. In addition, this research provides a reference for enterprises and marketers regarding the use of AR tour-sharing APPs to market tourist attractions, and also for future related studies.

The harmonic distribution caused by PWM is quite extensive, and increases the possibility of grid–train coupling resonance in the traction power supply system (TPSS). This paper first analyzes the mechanism of resonance, when the characteristic harmonic frequency of a four-quadrant converter (4QC) current that injects into the traction grid matches the resonant frequency of the traction grid, which may result in resonance in the system. To suppress resonance, this paper adopts specific harmonic elimination–pulse width modulation (SHE-PWM) technology combined with a transient direct current control strategy to eliminate the harmonics in the resonant frequency, which may suppress the grid–train coupling resonance. Due to the fact that the SHE-PWM process with multiple switching angles contains complex transcendental equations, the initial value is difficult to provide, and is difficult to solve using ordinary iterative algorithms. In this paper, an active-set secondary optimization method is used to solve the equation. The algorithm has the benefits of low dependence on initial values, fast convergence and high solution accuracy. Finally, the feasibility of the resonant suppression algorithm is verified by means of Matlab simulation.
