    Notes: Abstract Expression of structural genes of phospholipid biosynthesis in yeast is mediated by the inositol/choline-responsive element (ICRE). ICRE-dependent gene activation, requiring the regulatory genes INO2 and INO4, is repressed in the presence of the phospholipid precursors inositol and choline. INO2 and, to a less extent, INO4 are positively autoregulated by functional ICRE sequences in the respective upstream regions. However, an INO2 allele devoid of its ICRE functionally complemented an ino2 mutation and completely restored inositol/choline regulation of Ino2p-dependent reporter genes. Low-level expression of INO2 and INO4 genes, each under control of the heterologous MET25 promoter, did not alter the regulatory pattern of target genes. Thus, upstream regions of INO2 and INO4 are not crucial for transcriptional control of ICRE-dependent genes by inositol and choline. Interestingly, over-expression of INO2, but not of INO4, counteracted repression by phospholipid precursors. Possibly, a functional antagonism between INO2 and a negative regulator is the key event responsible for repression or de-repression.

    Notes: Abstract The ICL1 gene encoding the isocitrate lyase from Saccharomyces cerevisiae was cloned and sequenced. A reading frame of 557 amino acids showing significant similarity to isocitrate lyases from seven other species could be identified. Construction of icl1 null mutants led to growth defects on C2 carbon sources while utilization of sugars or C3 substrates remained unaffected. Using an ICL1-lacZ fusion integrated at the ICL1 locus, a more than 200-fold induction of β-galactosidase activity was observed after growth on ethanol when compared with glucose-repressed conditions. A preliminary analysis of the ICL1 upstream region identified a 364-bp fragment necessary and sufficient for this regulatory phenotype. Sequence motifs also present in the upstream regions of co-regulated genes were found within this region.

    Notes: Zusammenfassung Petrographische und chemische Untersuchungen an Dioriten und Graniten des Kyffhäuser und von Dessau lassen eine Ichor-Metamorphose, d. h. eine Alkali-Metasomatose von den Porenräumen aus, erkennen. In den metamorph-metablastischen Granitabkömmlingen wird die Ichor-Metamorphose deutlich an einer Glomeroblastenbildung und einer erneuten körnigen Texturentwicklung. Diese beweist eine Zufuhr von Albitmolekül. Sie wird in dem metamorph-metablastischen dioritischen Gestein neben der Verdrängung der primären Plagioklase aus der Alkalisierung und Enteisenung der Hornblende ersichtlich. Die magmatischen Ausgangsgesteine: Diorit, Quarzdiorit, Biotitdiorit, Gabbrodiorit (?) und Hornblendegabbro zeigen neben der Alkalisierung eine Muskovitisierung der basischen Plagioklase und eine Biotitisierung der Hornblende, woraus eine Zufuhr an Kali hervorgeht. Diese Stadien der Metamorphose durchlaufen den tektonischen Raum von der Grenze metamorph-nichtmetamorph bis hinab zur Grenze metamorph-magmatisch (Erdmannsdörffer) und rind in der Mitteldeutschen Schwelle zuerst aus dem tiefsten Stockwerk, im Südwesten im Odenwald-Spessart (Erdmannsdörffer) und nunmehr auch aus ihrer äußersten Haube erkannt worden, wo die Schwelle unter die Phyllithülle hinabtaucht.

    Notes: Abstract A procedure to estimate the failure probabilities of reinforced concrete (R/C) chimneys under severe wind loading which allows for considering realistic correlation values of concrete strength is developed. The results show that the assumption of full correlation of concrete strength within cross-sections, as suggested for example in present codes, may significantly overestimate the failure rates in present examples by two orders of magnitude. However, it is also shown that not only one but a number of cross-sections may contribute to the total failure rate of R/C chimneys. As a consequence, the failure estimates increase, in some cases by as much as two orders of magnitude. In the numerical examples as investigated here these counteracting effects neutralize each other. This fact, however, should not be generalized.

    Notes: Summary To identify the mildew resistance locus Mla in barley with molecular markers, closely linked genomic RFLP clones were selected with the help of near-isogenic lines having the ‘Pallas’ and ‘Siri’ background. Out of 22 polymorphic clones 3 were located around the Mla locus on chromosome 5 with a distance of 5.1 + 2.9 cM (MWG 1H068), 4.2±1.7 cM (MWG 1H060) and 0.7 ± 0.7 cM (MWG 1H036), respectively. The polymorphic clone MWG 1H036 displayed the same RFLP pattern in both ‘Pallas’ and ‘Siri’ near-isogenic lines and in different varieties digested with six restriction enzymes possessing the same mildew resistance gene. The alleles of the Mla locus were grouped in 11 classes according to their specific RFLP patterns; 3 of these groups contain the majority of Mla alleles already used in barley breeding programs in Europe.

    Notes: Abstract This work reports calculations and measurements on mode converters for the transformation of TEOn gyrotron mode mixtures (primarily TEO2) into the linearly polarized HE11 hybrid mode at 70 GHz. This mode is ideal for quasi-optical launching systems for ECRH of plasmas. Mode transducers with axisymmetric radius perturbations convert the gyrotron TEOn mode composition into the TEO1 mode. Proper matching of the phase differences between the various modes and of the perturbation amplitudes of the several converter sections is required. The TEO1 wave is used for long-distance transmission through smooth-walled overmoded waveguides. A mode converter with constant diameter and periodically perturbed curvature transfers the unpolarized TEO1 mode into the polarized TE11 wave. The experimentally determined ΣTEOn-to-TEO1 conversion efficiency is (98.5±1)% (99% predicted) while the TEO1-to-TE11 transformer has a (94±2)% efficiency (93% theoretically); ohmic losses are included. The “Gaussian-like” HE11 mode with axisymmetric power distribution and almost no cross polarization is produced in a circumferentially corrugated TE11-to-HE11 mode transducer with a measured conversion efficiency of (98.3±1.5)% (98.5% predicted). The overall efficiency of the complete mode converter system in the desired mode was determined to be (91±2.5)%. High-power operation (200kW, 100ms) has been successfully demonstrated.
