    Notes: A spinning cylindrical filter, known as a spinfilter, permits the mammalian cell bioreactor operation at high perfusion rates leading to very high cell densities (107 mL-1). Filter screens with openings (25 μm) slightly larger than the average cell size have been used to retain single cells in suspension over a long period of operation without clogging. We have previously shown why it is necessary to optimize the rotational speed of the spinfilter in order to achieve efficient cell retention and avoid potential screen clogging. Effects of bulk mixing and perfusion rate on screen fouling and cell retention were also investigated. Based on this analysis, in this article, we suggest strategies for scaleup of spinfilters. Experimental data from 12- and 175-L (working volume) bioreactors is shown in support of the scaleup analysis. © 1994 John Wiley & Sons, Inc.

    Notes: Some synthetic peptides increased the luminol-dependent chemiluminescence of mouse blood during phagocytosis. It is suggested that the levels of antimicrobial activity of the neutrophil peroxidase system can be raised very quickly (within some dozen of seconds) by these peptides. This raises the possibility of finding a new approach to the therapy for infectious diseases.

    Notes: Alcohol yields of 6.5% were obtained with Saccharomyces cerevisiae in lactasehydrolyzed acid whey permeate containing 30-35% total solids. Maximum alcohol yields obtained with Kluyveromyces fragilis were 4.5% in lactase-hydrolyzed acid whey permeate at a solids concentration of 20% and 3.7% in normal permeate at a solids concentration of 10%. Saccharomyces cerevisiae efficiently converted the glucose present in lactase-hydrolyzed whey permeates containing 5-30% total solids (2-13% glucose) to alcohol. However, the galactose, which comprised about half the available carbohydrate in lactase-hydrolyzed whey, was not utilized by S. cerevisiae, so that even though alcohol yields were higher when this organism was used, the process was wasteful in that a substantial proportion of the substrate was not fermented. For the process to become commercially feasible, an efficient means of rapidly converting both the galactose and glucose to alcohol must be found.

    Notes: A general model of the transient behaviour of an oxygen probe is derived on more general assumptions than those previously considered. The present state of the art is shorly discussed in its relation to the newly derived equations: (1) the reducibility of the three-layer diffusional model to two-layer ones is discussed, and the two-layer model considering the membrane and the liquid film is verified experimentally, (2) experimental and theoretical treatments of transient probe characteristics determination are examined, and (3) the influence of chemical reactions of the first order as well as of zero order (microorgaganism respiration) in measured media on transientd probe characteristics is quantified.

    Notes: Whole cells of Escherichia coli containing the enzyme penicillinamidohydrolase EC 3.5.1.11 were immobilized on the surface of modified macroporous copolymers of glycidylmethacrylate with ethylenedimethacrylate and of copolymers of methacrylaldehyde (MA) with divinylbenzene (DVB) by means of glutaraldehyde. These polymeric carriers were modified before cell binding by using ammonia or polyamines, especially ethylenediamine and hexamethylenediamine (HMDA). The highest specific activity and the largest yield in cell immobilization were achieved with the macroporous copolymer of MA and DVB modified with HMDA. The material thus obtained was used in repeated conversions of benzylpenicillin to 6-aminopenicillanic acid in a stirred batch reactor.

    Notes: Hexokinase (B.C. 2.7.1.1) activity as a marker enzyme during FMD viral infection has been observed spectrophotometrically in a system coupled with glucose-6-phosphate dehydrogenase, in supernatants of BHK21Cl13 suspension as well as anchored cell culture at a minimum of 104 infective virus particles/ml. Specific activity increased with virus concentration in culture supernatants and abruptly decreased with a fall in virus titer, as has been noted by TCID/50,146 S concentration, and enzyme-linked immunosorbent assay (ELISA) readings.

    Notes: Various experimental procedures for the determination of transient characteristics with significant liquid film effects were tested. A comparison between transient characteristics obtained experimentally and those calculated from rational models indicates that all procedures but one give highly inconsistent results. Recalculation of transient characteristics with no liquid film (easily measured in the gas phase) to that with liquid film (occurring in viscous liquids) is recommended as well as the selected experimental procedure which yields consistent results in the situations where the steady-state probe reading is decreased up to one-half due to the liquid film.
