    Notes: Abstract The CFLP methodology was applied for Cleavase I site detection within ptDNA intergenic regions (atpB-rbcL and rps14-psaB) at both interspecific and intraspecific levels in the genus Phaseolus. Optimal Cleavase I reaction temperature was 55 °C and the semi-dry electrophoretic transfer was more efficient than the original capillary one. Cleavase reactions yield a high number of fragments as compared to PCR-RFLP and allowed differentiation within and between landraces and wild forms of the Lima bean (Phaseolus lunatus L.) originating from Andean and Mesoamerican regions of Latin America. From sequencing data and using stemloop program (GCG, Madison), congruent numbers of hairpins/fragments were identified within the sequences, highlighting the robustness of the Cleavase I. Our results pointed out the ubiquity of short conserved motifs amongst a geographically localized group of species. In the vicinity of these motifs, synapomorphic-like substitutions were frequently observed. A phylogenetic tree based on these sequences is congruent with the CFLP pattern as well as with the widely accepted phylogeny of the genus. The usefulness of this new tool as alternative and/or complementary to PCR-RFLP technology on ptDNA is suggested and discussed.

    Notes: Summary Computer-assisted metaphase analysis for prenatal diagnosis can be economically performed by a preexisting hardware-software system. The program can be run by technician-level (i.e., nonspecialist) personnel who can opt for the automatic operational mode for good quality metaphases or for the semimanual mode for problem metaphases (numerous superpositions, artifacts, etc.).     Description / Table of Contents: Résumé Les auteurs décrivent une nouvelle méthode de coloration dans le bloc de tissu avec différents sels métalliques (alun de fer, tétroxyde d'osmium, acétate d'uranyl et chromeosmium) et l'hématoxyline. Cette technique est utilisée sur des tissus qui sont ensuite inclus en Araldit et permet d'obtenir des coupes semi-fines déjà colorées. Les auteurs ont cherché à préciser la spécificité de fixation du fer dans le tissu au moyen de méthodes d'extraction et de blocage. D'après leurs observations, le fer se fixerait aux groupes tissulaires chargés négativement. La nature des liaisons semble être de type électrostatique et complexe. Les liaisons complexes semblent prédominer au niveau des structures contenant des acides nucléiques.
    Notes: Summary The authors describe a new method of block staining for semifine sections with various metal salts (ferriammonium-sulphate, osmium tetroxyde, uranyl acetate, chromeosmium) and hematoxylin. They investigated the specificity of iron-alaun-hematoxylin with several extractions and blocking methods. According to their observations the dissociated iron is bounded to the negatively charged tissular groups. The nature of binding is thought to be electrostatic and coordinativ (complex). The last one is probably predominant in nucleic acids containing structures. This method seems to be encouraging for electron-histochemical investigations.

    Notes: Abstract Aminopeptidase P was solubilized from bovine lung by sodium deoxycholate extraction of salt-washed, delipidated lung acetone powders. Hydrolysis of the standard aminopeptidase P substrate, Gly-Pro-Hyp, as well as cleavage of Arg-Pro-Pro and the Arg1-Pro2 bond of bradykinin, co-eluted from a Mono Q anion exchange column and demonstrated identical inhibitory profiles suggesting that all activities were functions of the same enzyme. The metal chelator, 1,10-phenanthroline, completely inhibited activity suggesting that aminopeptidase P is a metallopeptidase. 2-Mercaptoethanol was both a potent and specific inhibitor of the enzyme (at 4 mM). A variety of other peptidase inhibitors showed either no effect or failed to completely inhibit even at high concentrations. The inhibitory profile and substrate specificity differ considerably from previous reports claiming to study the properties of this enzyme. Evidence is provided that aminopeptidase P may have an important role in the pulmonary degradation of the potent vasoactive peptide, bradykinin.

    Notes: Abstract The pharmacokinetics of cefroxadin have been studied after the administration of single oral and intravenous doses to healthy volunteers. Cefroxadin was assayed by HPLC. The kinetics in plasma following i.v. administration were described by using a three-compartment model. An additional disposition phase was observed following oral administration that could not be detected after the low i.v. dose. The terminal half-life was 1.03 h. The apparent volume of distribution at the steady state was consistent with a diffusion of the antibiotic in all extracellular fluids. The AUCafter oral administration was linearly related to the dose. The urinary excretion amounted to 95% of the dose with virtually complete absorption of orally administered drug.

    Notes: Abstract Combined methodologies of enzyme-linked immunosorbent assay (ELISA), sodium dodecyl sulphate polyacrilamide gel electrophoresis (SDS-PAGE), immunoblotting, traditional myofibrillar ATPase (mATPase) histochemistry and immunocytochemistry of whole biopsied samples were used to study myosin heavy chain (MHC) isoforms in the equine gluteus medius muscle. The ELISA technique allowed the quantification of the three MHC isoforms known to be present in different horse muscles: slow (MHC-I) and two fast (termed MHC-IIA and MCH-IIX). The SDS-PAGE method resolved MHCs in three bands: MHC-I, MHC-IIX and MHC-IIA from the fastest to the slowest migrating band and a quantification by densitometry for each MHC isoform was also possible. The identity of these three MHCs was confirmed by immunoblots with specific monoclonal antibodies. Five fibre types were defined immunohistochemically according to their MHC content: I, I + IIA, IIA, the hybrid IIAX and IIX. When quantitative data obtained with the four different methodologies were combined and compared, they were consistent and, when considered together, showed significant correlation. Nevertheless, the percentage of MHC-IIA histochemically derived was underestimated, while that of MHC-IIX was overestimated in comparison with the immunocytochemical determination of these MHC isoforms. The percentage of MHC-I obtained by ELISA technique was underestimated. In short, these integrated methods for the analysis of MHCs at the protein level demonstrate that equine skeletal muscle does not express the MHC-IIB, so type II fibres have been misclassified in numerous previous studies based upon the very traditional mATPase histochemistry. They also offer new prospects for muscle fibre typing in equine experimental studies and veterinary medicine.

    Notes: Summary Different ratios of normal human plasma concentrations to fetal bovine serum concentrations have been tested on the in vitro proliferation of pluripotent hematopoietic progenitors. The slight modifications of the culture procedure described here resulted in significant enhancement of BFU-e formation whereas no significant differences in the formation of CFU-e, CFU-meg, CFU-gm, and CFU-mix were observed.
