    Notes: Abstract A new and sensitive procedure for the graphite furnace-atomic absorption spectrophotometric (GF-AAS) determination of Pd in soil at nanogram level is described. The method is based on prior separation and enrichment of the metal as Pd(II)-SnCl3 –-N-butylacetamide (BAA) complex into 1-pentanol (PN) by solvent extraction method. The value of the molar absorptivity of the complex in three solvents, i.e. ethyl acetate, 1-pentanol, chloroform, lie in the range of (0.70–2.75) × 104 L mol–1 cm–1 at λmax 360–440 nm. The metal could be enriched into organic solvent, i.e. PN, up to 10-folds. The sensitivity (A = 0.0044) of the method in the term of the peak height was 0.5 ng Pd/mL of the aqueous solution at an enrichment factor (EF) of 5. Optimization of analytical variables during enrichment and GF-AAS determination of the metal are discussed. The method has been applied for the analysis of Pd to soil samples derived from roads and highways in Germany.

    Notes: Abstract Detection of DNA synthesizing cells may often be achieved by immunocytochemical detection of bromodeoxyuridine (BrdU), which is rapid and appears to give similar results to those found using tritiated thymidine. However, the methodology for detection of BrdU involves a denaturation or digestion step to allow access of the antibody to BrdU incorporated into single- rather than double-stranded DNA. We wished to determine if microwave treatment could be used to enhance the detection of BrdU without the need for any other digestion/denaturation steps. An important consideration was to investigate whether such treatment produces a similar quantitative result, since BrdU detection is usually assessed on the basis of cell number rather than topographical distribution. We have found that microwave treatment can allow considerably lower antibody concentrations and eliminates the need for any other denaturation step. It also reduces the non-specific background staining found when using monoclonal antibodies on mouse tissue. We have performed cell counts and found that the number of BrdU positive cells remains constant for a range of different immunocytochemical parameters. We also report conditions where immunopositivity is adversely affected by changes in technique and describe the optimised conditions for obtaining reproducible results.

    Notes: Summary Cytochemical detection of ascorbic acid in cultured root tips of Zea mays shows that dividing cells accumulate ascorbic acid in the cytoplasm. The localization pattern alters in the root tip as the cells begin to elongate. In elongating cells ascorbic acid is distinctly localized on cell walls. Ascorbic acid content per cell inreases with the onset of cell elongation. Fully elongated cells contain fivefold more ascorbic acid than meristematic cells. Cytophotometric analysis reveals a sharp and positive correlation (r=+0.93) between percentage increase in content of ascorbic acid per cell and corresponding increase in cell size at different phases of cell elongation. IAA treatment to the roots raises the content of ascorbic acid per cell with a parallel increase in size of cell. Involvement of ascorbic acid in IAA induced cell elongation is discussed.

    Notes: Abstract A new extraction-spectrophotometric method for the determination of indium at ppb level in zinc effluent is described. It is based on quantitative extraction (51.2–99.2%) of the tetrabromoindate complex with toluene solution of N,N′-diphenylbenzamidine and its nine analogous from sulphuric acid solutions, which allowed the extract to react with basic dye (i.e. brilliant green, malachite green, crystal violet) solution in sequence. Among the basic dyes tested, brilliant green gave the most sensitive colour reaction. The value of molar absorptivity of the amidine bromoindate extracts with brilliant green lie in the range of (0.53–1.03)×105 l mole-1 cm-1 at absorption maximum, 630–640. With the most sensitive compound, N-(2-methyl-phenyl)-N-phenylbenzamidine(APPBA), the detection limit of the method is 13 ppb. The method is free from interferences of almost all ions associated with indium. The method has been applied for the determination of indium to zinc-effluent.

    Notes: Abstract Haemolysis by Sendal virus, α-toxin, and activated complement is inhibited by high concentrations of divalent cations. In Daudi cells, sublytic amounts of these agents induce the following changes: collapse of surface membrane potential, uptake of Na+ and loss of K+ from cells, and leakage of phosphorylated metabo-tites from cells. The changes induced by Sendal virus and complement are sensitive to physiological concentrations of extracellular Ca2+. It is concluded that fluctuations in plasma Ca2+ concentration may affect the damaging action of certain pore-forming agents on susceptible cells.

    Notes: Abstract A simple, selective, and sensitive field procedure for the spectrophotometric determination of thallium in water is described. The preconcentration is carried out by extracting the chlorothallate ion-pair complex of a cationic surfactant, i.e. CTAB or CPC, with toluene. The extract is reacted with a basic dye, i.e. brilliant green (BG). The molar absorptivity of the complexes is in the range of (1.63–1.70)×105 1 mole−1 cm−1 at the absorption maximum of 640 nm. The detection limit is 5 ng ml−1 Tl in water. None of the tested metal ions upto certain levels interfered with the determination. It is reproducibly applicable to the field analysis of Tl in water samples containing upto 20 ppb.
