    Notes: Ethanol and cycloheximide inhibited the function of the ammonium transport system in growing cultures of Saccharomyces cerevisiae var. ellipsoideus measured as methylamine uptake. The effect was reversible with ethanol and irreversible with the antibiotic. The kinetic data are consistent with a reduction of the number of active carrier molecules located in the plasma membrane. In contrast, neither ethanol nor cycloheximide affected the specific rate of fructose uptake.

    Notes: Three white-rot fungi, Phanerochaete chrysosporium, Polyporus tulipiferae, and Polyporus sp. A336 were grown on 100-g amounts of chopped oat straw in gassed 4.5 L (diameter 16 cm, height 23 cm) solid-state reactors for two weeks. The different gas atmospheres were regulated by (1) air diffusion through foam plugs, (2) intermittent or continuous air flow, (3) intermittent oxygen, 50 or 100% continuous oxygen flow, and (4) continuous 10% carbon dioxide in oxygen flow. The fermented straw was analyzed for total weight loss, Klason lignin loss, and enzymatic (cellulase) hydrolysis. P. chrysosporium grown on straw in continuous oxygen at 35°C caused a 41% weight loss and 33.5% hydrolysis was obtained when the pretreated straw was hydrolyzed with cellulase enzyme. P. tulipiferae caused a 27% weight loss and 34.3% cellulase hydrolysis in the straw at 30°C. Polyporus sp. A336 selectively degraded lignin of the straw and under intermittent oxygen resulted in an 18% weight loss and 33.6% cellulase hydrolysis at 35°C. When the straw was supplemented with 10% xylose (straw basis) and was continuously gassed with 50% oxygen, Polyporus sp. A336 produced a 14.5% weight loss and 38.7% cellulase hydrolysis. Oxygen and carbon dioxide exchange rates were measured for some of these bench-scale fermentations.

    Notes: The evolution of the activity of the general amino acid permease and ethanol and glucose concentrations in the medium were studied in a mild fermentation process carried out by a wine strain of Saccharomyces cerevisiae var. ellipsoideus isolated from grape musts in spontaneous fermentation. The cells displayed a reduction in the activity of the general amino acid permease parallel to the increase of ethanol in the medium. This ethanol increase was not enough to promote a substantial inhibition on the total polypeptide synthesis measured as polyuridylic-acid-directed polyphenylalanine synthesis.

    Notes: A zymographic assay is described for the detection of peroxidase isoenzymes oxidizing 4-hydroxystilbene following isoelectric focusing. The assay is based on coupling intermediate products of the oxidation of 4-hydroxystilbene with 4-aminoantipyrine, with resultant formation of dye complexes. Control experiments in the absence of 4-hydroxystilbene and hydrogen peroxide demonstrate the peroxidative nature of the 4-hydroxystilbene-dependent dye reaction.

    Notes: Some polymorphic proteins (α1-antitrypsin, orosomucoid, transferrin, group specific component, plasminogen) and enzymes (phosphoglucomutase, acid phosphatase, estrase D) were determined in bloodstain extracts by isoelectric focusing with carrier ampholytes (CA) and with immobilized pH gradients (IPGs) rehydrated with CA. IPGs yield superior results for typing of genetics markers in bloodstains since phenotypes are better distinguished and the bands are straighter and sharper in the presence of contaminants. Also, the sensitivity of IPGs with CA is similar to isoelectric focusing (IEF) with CA. A new variant, ACP*B1, found in Negroid west African populations and not found in Caucasians is described. Such a variant can only be determined by IPGs since its isoelectric point (pI 5.95) is close to that of the ACP*B (pI 6.05) variant.

    Notes: Single point mutations in small DNA fragments and single unit differences in simple repetitive DNA can be detected as double-strand conformation polymorphisms using polyacrylamide gel electrophoresis, with and without sodium dodecyl sulfate, even at temperatures as low as 3°C. Changes in a single base are distinguished by means of the analysis of the heteroduplexes, and changes in more than two bases can be distinguished in both homoduplexes and heteroduplexes. Polymerase chain reaction (PCR) conditions can be designed not only to amplify homoduplexes, heteroduplexes and singlestrand DNA at the same time, but also to focus the analysis on either singlestrand conformation polymorphism (SSCP) or double-strand conformation polymorphism (DSCP). DSCP seems to be advantageous in typing DNA polymorphisms or mutations in loci with few variants, but, because it is necessary to have a simple pattern of all possible combinations of the alleles, it is not as advantageous in typing systems with many variants.
