    Description: The key event in the choice of repair pathways for DNA double-strand breaks (DSBs) is the initial processing of ends. Non-homologous end joining (NHEJ) involves limited processing, but homology-dependent repair (HDR) requires extensive resection of the 5' strand. How cells decide if an end is channeled to resection or NHEJ is not well understood. We hypothesize that the structure of ends is a major determinant and tested this hypothesis with model DNA substrates in Xenopus egg extracts. While ends with normal nucleotides are efficiently channeled to NHEJ, ends with damaged nucleotides or bulky adducts are channeled to resection. Resection is dependent on Mre11, but its nuclease activity is critical only for ends with 5' bulky adducts. CtIP is absolutely required for activating the nuclease-dependent mechanism of Mre11 but not the nuclease-independent mechanism. Together, these findings suggest that the structure of ends is a major determinant for the pathway choice of DSB repair and the Mre11 nuclease dependency of resection.

    Description: We report the effect of relaxation-type self-induced temperature oscillations in the system of two parallel plates of SiO 2 and VO 2 which exchange heat by thermal radiation in vacuum. The non-linear feedback in the self-oscillating system is provided by metal-insulator transition in VO 2 . Using the method of fluctuational electrodynamics, we show that under the action of an external laser of a constant power, the temperature of VO 2 plate oscillates around its phase transition value. The period and amplitude of oscillations depend on the geometry of the structure. We found that at 500 nm vacuum gap separating bulk SiO 2 plate and 50 nm thick VO 2 plate, the period of self-oscillations is 2 s and the amplitude is 4 K, which is determined by phase switching at threshold temperatures of phase transition.

    Description: Many genes involved in brain development have been associated with human neurodevelopmental disorders, but underlying pathophysiological mechanisms remain undefined. Human genetic and mouse behavioral analyses suggest that ENGRAILED-2 ( EN2 ) contributes to neurodevelopmental disorders, especially autism spectrum disorder. In mouse, En2 exhibits dynamic spatiotemporal expression in embryonic mid-hindbrain regions where monoamine neurons emerge. Considering their importance in neuropsychiatric disorders, we characterized monoamine systems in relation to forebrain neurogenesis in En2 -knockout ( En2 -KO) mice. Transmitter levels of serotonin, dopamine and norepinephrine (NE) were dysregulated from Postnatal day 7 (P7) to P21 in En2 -KO, though NE exhibited the greatest abnormalities. While NE levels were reduced ~35% in forebrain, they were increased 40 – 75% in hindbrain and cerebellum, and these patterns paralleled changes in locus coeruleus (LC) fiber innervation, respectively. Although En2 promoter was active in Embryonic day 14.5 – 15.5 LC neurons, expression diminished thereafter and gene deletion did not alter brainstem NE neuron numbers. Significantly, in parallel with reduced NE levels, En2 -KO forebrain regions exhibited reduced growth, particularly hippocampus, where P21 dentate gyrus granule neurons were decreased 16%, suggesting abnormal neurogenesis. Indeed, hippocampal neurogenic regions showed increased cell death (+77%) and unexpectedly, increased proliferation. Excess proliferation was restricted to early Sox2/Tbr2 progenitors whereas increased apoptosis occurred in differentiating (Dcx) neuroblasts, accompanied by reduced newborn neuron survival. Abnormal neurogenesis may reflect NE deficits because intra-hippocampal injections of β-adrenergic agonists reversed cell death. These studies suggest that disruption of hindbrain patterning genes can alter monoamine system development and thereby produce forebrain defects that are relevant to human neurodevelopmental disorders.

    Description: We present a micromagnetic study on the eigen excitations of an artificial skyrmion crystal, which has been experimentally confirmed to be stable at room temperature without the need of any Dzyaloshinsky-Moriya interaction (DMI). Three in-plane rotational modes and one breathing-type mode are identified. We find the intrinsic origin of the dynamics of skyrmion crystal is the nontrivial magnetic texture instead of DMI. And the rotational direction of a skyrmion is solely determined by the sign of the skyrmion number, irrespective of its circulation sense, evidencing the topological nature of the magnetic skyrmion.

    Description: : High-throughput sequencing technologies survey genetic variation at genome scale and are increasingly used to study the contribution of rare and low-frequency genetic variants to human traits. As part of the Cohorts arm of the UK10K project, genetic variants called from low-read depth (average 7 x ) whole genome sequencing of 3621 cohort individuals were analysed for statistical associations with 64 different phenotypic traits of biomedical importance. Here, we describe a novel genome browser based on the Biodalliance platform developed to provide interactive access to the association results of the project.     Description: A type of metal-dielectric multilayered structures is investigated theoretically and experimentally for achieving optical transparency with a high electrical conductivity. The structure in our demonstrated case comprises of two coupled metal-dielectric-metal planar optical resonators with metal-layer thicknesses near to its skin depth. Simulations show that the maximum transmittance for visible light can easily reach 90% for silver-based structures. Experimentally, the sample fabricated exhibits a transmission window with a bandwidth of 150 nm and a maximum transmittance of 76% around 643 nm wavelength at normal incidence. Its sheet resistance is measured to be less than 10Ω/□, much smaller than that of common indium-tin-oxide films. Transparent conductors functioning for blue light and even for the whole visible light are also shown to be theoretically possible. Owing to their simple fabrication procedure as well as design flexibility, such a layered structure can serve as a compelling alternative as transparent conductors for optoelectronic devices, especially for liquid-crystal displays and light-emitting diodes.

    Description: HLX-1, currently the best intermediate-mass black hole candidate, has undergone seven violent outbursts, each with a peak X-ray luminosity of L peak,X  ~ 10 42  erg s –1 . Interestingly, the properties of the HLX-1 outbursts evolve with time. In this work, we aim to constrain the physical parameters of the central engine of the HLX-1 outbursts in the framework of the black hole accretion. We find that the physical properties of the HLX-1 outbursts are consistent with being driven by the radiation pressure instability. This scenario can explain the evolution of the recurrent time-scales of the HLX-1 outbursts as a function of the durations.

    Description: We study the dependence of star formation quenching on galaxy mass and environment, in the Sloan Digital Sky Survey (SDSS; z  ~ 0.1) and the All-Wavelength Extended Groth Strip International Survey (AEGIS; z  ~ 1). It is crucial that we define quenching by low star formation rate rather than by red colour, given that one-third of the red galaxies are star forming. We address stellar mass M * , halo mass M h , density over the nearest N neighbours N and distance to the halo centre D . The fraction of quenched galaxies appears more strongly correlated with M h at fixed M * than with M * at fixed M h , while for satellites quenching also depends on D . We present the M * – M h relation for centrals at z  ~ 1. At z  ~ 1, the dependence of quenching on M * at fixed M h is somewhat more pronounced than at z  ~ 0, but the quenched fraction is low (10 per cent) and the haloes are less massive. For satellites, M * -dependent quenching is noticeable at high D , suggesting a quenching dependence on subhalo mass for recently captured satellites. At small D , where satellites likely fell in more than a few Gyr ago, quenching strongly depends on M h and not on M * . The M h dependence of quenching is consistent with theoretical wisdom where virial shock heating in massive haloes shuts down accretion and triggers ram-pressure stripping, causing quenching. The interpretation of N is complicated by the fact that it depends on the number of observed group members compared to N , motivating the use of D as a better measure of local environment.

    Description: The CRISPR-Cas9 system uses guide RNAs to direct the Cas9 endonuclease to cleave target sequences. It can, in theory, target essentially any sequence in a genome, but the efficiency of the predicted guide RNAs varies dramatically. If no targeted cells are obtained, it is also difficult to know why the experiment fails. We have developed a transient transfection based method to enrich successfully targeted cells by co-targeting the hypoxanthine phosphoribosyltransferase (HPRT) gene. Cells are transfected with two guide RNAs that target respectively HPRT and the gene of interest. HPRT targeted cells are selected by resistance to 6-thioguanine (6-TG) and then examined for potential alterations to the gene targeted by the co-transfected guide RNA. Alterations of many genes, such as AAVS1, Exo1 and Trex1, are highly enriched in the 6-TG resistant cells. This method works in both HCT116 cells and U2OS cells and can easily be scaled up to process multiple guide RNAs. When co-targeting fails, it is straightforward to determine whether the target gene is essential or the guide RNA is ineffective. HPRT co-targeting thus provides a simple, efficient and scalable way to enrich gene targeting events and to identify the cause of failure.

    Description: Magnetic, dielectric, and magnetoelectric properties have been investigated in the polycrystalline Mn 4 Nb 2 O 9 . Under zero magnetic fields, no dielectric anomaly and electric polarization are observed in this compound. When the sample is exposed in magnetic field, finite dielectric peaks and electric polarization are induced, which increase with increasing magnetic field, showing magneto-dielectric and magnetoelectric effects. The origin of magnetoelectric coupling of this compound has been discussed.
