    Notes: It was shown in the course of developing the high-accuracy universal polarimeter (HAUP) that any polarimetric optical analyses cannot be free from a systematic error γ originating in the parasitic ellipticities of the constituent Nicol prisms. A method by which one can remove γ in the HAUP method is presented. This has been successfully applied to measurements of the gyration tensors and birefringence of two enantiomorphic crystals of α-quartz. It was found that the order of magnitude of this error for a polarizer is about 10−4 and consequently γ lies between 10−4 and 10−3 in typical optical systems. Our results, g11 = 5.7 ± 0.52 × 10−5 and g33 = −13.6 ± 0.52 × 10−5 with a wavelength of 6328 Å for laevorotatory quartz at 300 K are in good agreement with some of the previous reports.

    Notes: Abstract A method of measuring short circuit current has been established for high power and low internal resistance batteries. The battery was forcibly discharged at a constant resistance much smaller than its internal resistance by a newly constructed short circuit switch. Current and voltage were then monitored by a digital memory scope. Pasted-type and tubular-type lead-acid batteries were used as samples in this work. Short circuit current was also observed for the batteries in various states-of-charge.

    Notes: Abstract Capacity and cycle life have been measured for commercially available lead-acid batteries by superimposing an a.c. upon the charge and discharge d.c. to clarify the influence of an a.c. invasion into the d.c. system on battery performance in an electric power storage system. The current was controlled to beI=I 0(1+sinωt) in all the experiments. The value ofI 0 corresponded to 5 or 8 HR and the frequency range was 0.1 to 4000 Hz. No capacity change was observed for the a.c. superimposition on the charge current in this frequency range. When an a.c. was superimposed upon the discharge current the capacity of the battery increased by less than 1%. No effect on the cycle life caused by the a.c. superimposition on the charge and the discharge current was observed, as the inherent distribution of the cycle life of the batteries used was much greater than the change caused by the a.c. superimposition. Thus, it was clarified that the influence of the a.c. superimposition on battery capacity and cycle life is practically negligible for lead-acid batteries.

    Notes: Abstract Initial short circuit currents have been observed using our electronic short circuit switch and also predicted from terminal voltage and ohmic resistance according to Ohm's law for several kinds of lead-acid batteries in various states-of-charge. Ohmic resistance was measured by the d.c. step and the a.c. impedance methods. The predicted and the observed values have been compared in order to establish a prediction method for initial short circuit current. A good agreement was obtained, the root mean square percentage deviation of the predicted value from that observed being only 2∼19%, which confirms the validity of the prediction method for the initial short circuit current.

    Notes: Abstract. Butyltin compounds (BTs), including tributyltin (TBT) and its breakdown products, di- (DBT) and monobutyltin (MBT), were determined in sea water, sediment, and biota at various trophic levels in the food chain collected from Otsuchi Bay, Japan, for understanding distribution and bioaccumulation of BTs in natural marine ecosystems. BT residues were detected in all the compartments analyzed, although their concentrations appeared to be less than those in polluted areas. Concentrations of BTs in sea water were higher in locations near Otsuchi Port, indicating that maritime activities in the harbor has been a major source of BTs in this bay. A specific peak in BT residue levels was found in sediment cores at an estimated depth that dated to be from the 1980s. Lack of significant variation in the composition of BTs at different depths of cores suggests slow degradation rate of these compounds in sediments. BTs were accumulated in plankton and other organisms up to ∼70,000 times higher than in sea water. However, no considerable biomagnification was observed for BTs through the food chain. Relatively high concentrations were found in caprellids and smaller fish, such as gunnels. These organisms accumulated TBT as the predominant compound among BT derivatives and showed higher bioconcentration factors for TBT than in other species reported so far. Our results suggest that certain organisms in the food chain may have a less capacity to degrade TBT, and therefore may accumulate BTs at elevated levels.

    Notes: Abstract  A total of 400 yeast strains were examined for the ability to reduce ethyl 4-chloroacetoacetate (COBE) to ethyl 4-chloro-3-hydroxybutyrate (CHBE) by using acetone-dried cells in the presence of a coenzyme-recycling system in water/n-butyl acetate. We discovered some yeast strains that reduced COBE to (S)-CHBE. Heating of acetone-dried cells of the selected yeast strains increased the optical purity of the product. There may be several enzymes that can reduce COBE stereoselectively in the same yeast cells. The cultured broth of Candida magnoliae accumulated 90 g/l (S)-CHBE (96.6% enantiomeric excess, e.e.) in the presence of glucose, NADP and glucose dehydrogenase in n-butyl acetate. When these cells were heated, the stereoselectivity of the reduction increased to 99% e.e. (S)-CHBE is one of the useful chiral building blocks applicable to the synthesis of some pharmaceuticals. We expect that the cheap and industrial production of this important chiral compound will follow the discovery of this yeast strain.

    Notes: Abstract A mixture of rProROL having the full-length prosequence (97 amino acids) for a recombinant lipase of Rhizopus oryzae (rROL) and r28ROL having 28 amino acids of the same prosequence has been produced as active forms by Saccharomyces cerevisiae [Takahashi et al. (1998) J Ferment Bioeng 86: 164–168]. However, the separation of rProROL and r28ROL has not been successful due to their identical behavior on column chromatographs, presumably because of the similarity of their surface properties. The independent production of two different molecular forms of rROL was carried out using KEX2-engineered strains of S. cerevisiae, since r28ROL was predicted to be a product from rProROL by a Kex2-like protease. rProROL was successfully obtained by expression of the ROL gene in the S. cerevisiae kex2 strain in which the KEX2 gene encoding Kex2p was disrupted, while r28ROL was obtained by co-expression of the gene (KEX2Δ613) encoding the soluble form of the C-terminal truncated Kex2 protease (sKex2p). The specific lipase activities of rProROL and r28ROL were 92.9 U/mg and 140 U/mg, respectively. rProROL was stable at pH 2.2–8.0, and showed the optimal reaction temperature to be 30–35 °C with a T 50 of 55 °C (T 50 is the temperature resulting in 50% loss of activity). The values for r28ROL were pH 3.0–10.0, 25–30 °C, and 40 °C, respectively. rProROL was an N-linked glycosylated form, but r28ROL was not. The enhanced thermostability of rProROL did not seem to be due to the N-linked glycosylation, as judged by the results of the Endo H treatment. rProROL had the highest esterase activity toward p-nitrophenyl laurate (C12), whereas r28ROL had the highest esterase activity toward p-nitrophenyl caprylate (C8) and stearate (C18). These results suggest that the distinct properties of these two forms of lipase are caused by the different length of the ROL prosequence.
