    Notes: It was shown in the course of developing the high-accuracy universal polarimeter (HAUP) that any polarimetric optical analyses cannot be free from a systematic error γ originating in the parasitic ellipticities of the constituent Nicol prisms. A method by which one can remove γ in the HAUP method is presented. This has been successfully applied to measurements of the gyration tensors and birefringence of two enantiomorphic crystals of α-quartz. It was found that the order of magnitude of this error for a polarizer is about 10−4 and consequently γ lies between 10−4 and 10−3 in typical optical systems. Our results, g11 = 5.7 ± 0.52 × 10−5 and g33 = −13.6 ± 0.52 × 10−5 with a wavelength of 6328 Å for laevorotatory quartz at 300 K are in good agreement with some of the previous reports.

    Notes: A volumetric heating method is applied to the pre-slit placed on the SPring-8 standard-undulator beamline front ends in order to handle the very intensive photon beam. A graphite block is used as the radiation-absorbing body, which is brazed to a cooling holder made of oxygen-free high-conductivity copper. Various experiments and calculations have been carried out on the strength of the brazed joint. Comparisons between the experiments and calculations show that static fracture of the joint would not occur even under the highest heat load. The outgassing properties of the graphite blocks are also presented here.

    Notes: SPring-8 front ends have a novel structure which makes it easy to rearrange them and exchange the components. The structure has a common support for all the components except X-ray beam-position monitors and lead collimators. The alignment of the common support as well as the components was carried out with an accuracy of 0.25 mm in the vertical and horizontal directions. Replaceable pumping systems have also been placed on the common support and have achieved a vacuum of 2 × 10−8 Pa at the upstream part of the front ends without synchrotron radiation. During the commissioning with synchrotron radiation, the pumping systems displayed good pumping-down characteristics. Commissioning has been successfully performed for four standard in-vacuum X-ray undulators and three bending-magnet-beamline front ends up to July 1997. Measurements of temperature rise show that absorber, pre-slits and XY slits can handle the anticipated maximum heat load at a ring current of 100 mA.

    Description: The formation, evolution and dispersal processes of protoplanetary discs are investigated and the disc lifetime is estimated. The gravitational collapse of a pre-stellar core forms both a central star and a protoplanetary disc. The central star grows by accretion from the disc and irradiation by the central star heats up the disc and generates a thermal wind, which results in the disc's dispersal. Using the one-dimensional diffusion equation, we calculate the evolution of protoplanetary discs numerically. To calculate the disc evolution from formation to dispersal, we add source and sink terms that represent gas accretion from pre-stellar cores and photoevaporation, respectively. We find that the disc lifetimes of typical pre-stellar cores are around 2–4 million years (Myr). A pre-stellar core with high angular momentum forms a larger disc with a long lifetime, while a disc around an X-ray-luminous star has a short lifetime. Integrating disc lifetimes under various masses and angular velocities of pre-stellar cores and X-ray luminosities of young stellar objects, we obtain the disc fraction at a given stellar age and mean lifetime of the disc. Our model indicates that the mean lifetime of a protoplanetary disc is 3.7 Myr, which is consistent with the observational estimate from young stellar clusters. We also find that the dispersion of X-ray luminosity is needed to reproduce the observed disc fraction.

    Description: Fragmentation of protoplanetary discs due to gravitational instabilities is a candidate of a formation mechanism of binary stars, brown dwarfs, and gaseous giant planets. The condition for the fragmentation has been thought that the disc cooling time-scale is comparable to its dynamical time-scale. However, some numerical simulations suggest that the fragmentation does not occur even if the cooling time is small enough, or the fragmentation can occur even when the cooling is inefficient. To reveal a realistic condition for fragmentation of self-gravitating discs, we perform two-dimensional numerical simulations that take into account the effect of the irradiation of the central star and radiation cooling of the disc, and precisely investigate the structure of the spiral arms formed in the protoplanetary discs. We show that the Toomre Q parameter in the spiral arms is an essential parameter for fragmentation. The spiral arms fragment only when Q 〈 0.6 in the spiral arms. We have further confirmed that this fragmentation condition observed in the numerical simulations can be obtained from the linear stability analysis for the self-gravitating spiral arms. These results indicate that the process of fragmentation of protoplanetary discs is divided into two stages: formation of the spiral arms in the discs; and fragmentation of the spiral arm. Our work reduces the condition for the fragmentation of the protoplanetary discs to the condition of the formation of the spiral arm that satisfies Q 〈 0.6.

    Description: The angular momentum distribution and its redistribution are of crucial importance in the formation and evolution of circumstellar discs. Many molecular line observations towards young stellar objects indicate that the radial distributions of the specific angular momentum j have a characteristic profile. In the inner region, typically R 100 au, the specific angular momenta distribute as j    r 1/2 , indicating the existence of a rotationally supported disc. In the outer regions, R 5000 au, j increases as the radius increases, and the slope is steeper than unity. This behaviour is assumed to reflect the original angular momentum distributions in the maternal molecular clouds. In the intermediate region, 100 au R 5000 au, the j -distribution appears to be almost flat. While this is often interpreted to be a consequence of the conservation of the specific angular momentum, the interpretation actually is insufficient and requires a stronger condition that the initial distribution of j must be spatially uniform. However, this requirement seems to be unrealistic and inconsistent with observations. In this work, we propose a simple alternative explanation: the apparently flat j profile is produced by strong elongation owing to the large velocity gradient in the accreting flow, no matter what the initial j -distribution is. In order to show this, we provide a simple analytic model for the gravitational collapse of molecular clouds. We also propose a method to estimate the ages of protostars using only the observed rotation profile. We demonstrate the validity of this method in comparison with hydrodynamic simulations, and apply the model to the young stellar objects L1527 IRS, TMC-1A and B335.

    Description: d -Aspartate oxidase (DDO) catalyzes the oxidative deamination of acidic d -amino acids, whereas neutral and basic d -amino acids are substrates of d -amino acid oxidase (DAO). DDO of the yeast Cryptococcus humicola (ChDDO) has much higher substrate specificity to d -aspartate, but the structural features that confer this specificity have not been elucidated. A three-dimensional model of ChDDO suggested that a histidine residue (His56) in the active site might be involved in the unique substrate specificity, possibly through the interaction with the substrate side chain in the active site. His56 mutants with several different amino acid residues (H56A, H56D, H56F, H56K and H56N) exhibited no significant activity toward acidic d -amino acids, but H56A and H56N mutants gained the ability to utilize neutral d -amino acids as substrates, such as d -methionine, d -phenylalanine and d -glutamine, showing the conversion of ChDDO to DAO by these mutations. This conversion was also demonstrated by the sensitivity of these mutants to competitive inhibitors of DAO. These results and kinetic properties of the mutants show that His56 is involved in the substrate specificity of ChDDO and possibly plays a role in the higher substrate specificity toward d -aspartate.

    Description: Hexagonal close-packed iron (hcp-Fe) is a main component of Earth’s inner core. The difference in density between hcp-Fe and the inner core in the Preliminary Reference Earth Model (PREM) shows a density deficit, which implies an existence of light elements in the core. Sound velocities then provide an important constraint on the amount and kind of light elements in the core. Although seismological observations provide density–sound velocity data of Earth’s core, there are few measurements in controlled laboratory conditions for comparison. We report the compressional sound velocity ( V P ) of hcp-Fe up to 163 GPa and 3000 K using inelastic x-ray scattering from a laser-heated sample in a diamond anvil cell. We propose a new high-temperature Birch’s law for hcp-Fe, which gives us the V P of pure hcp-Fe up to core conditions. We find that Earth’s inner core has a 4 to 5% smaller density and a 4 to 10% smaller V P than hcp-Fe. Our results demonstrate that components other than Fe in Earth’s core are required to explain Earth’s core density and velocity deficits compared to hcp-Fe. Assuming that the temperature effects on iron alloys are the same as those on hcp-Fe, we narrow down light elements in the inner core in terms of the velocity deficit. Hydrogen is a good candidate; thus, Earth’s core may be a hidden hydrogen reservoir. Silicon and sulfur are also possible candidates and could show good agreement with PREM if we consider the presence of some melt in the inner core, anelasticity, and/or a premelting effect.

    Description: Reactive oxygen species (ROS) produced by plant NADPH oxidases (NOXes) are important in plant innate immunity. The Oryza sativa respiratory burst oxidase homologue B ( OsRbohB ) gene encodes a NOX the regulatory mechanisms of which are largely unknown. Here, we used a heterologous expression system to demonstrate that OsRbohB shows ROS-producing activity. Treatment with ionomycin, a Ca 2+ ionophore, and calyculin A, a protein phosphatase inhibitor, activated ROS-producing activity; it was thus OsRbohB activated by both Ca 2+ and protein phosphorylation. Mutation analyses revealed that not only the first EF-hand motif but also the upstream amino-terminal region were necessary for Ca 2+ -dependent activation, while these regions are not required for phosphorylation-induced ROS production.

    Description: Smad7 is an inhibitory molecule induced by members of the transforming growth factor-β (TGF-β) family, including TGF-β, activin, nodal and bone morphogenetic proteins (BMPs). To elucidate the in vivo functions of Smad7, we generated conditional Smad7 -knockout mice in which the Mad homology 2 (MH2) domain and the poly (A) signal sequence were flanked with loxP sites (floxed). The Smad7 -floxed mice exhibited no obvious phenotype. Smad7 total-null mice on a C57BL/6 background died within a few days of birth, whereas mice with an ICR background developed to adulthood but were significantly smaller than wild-type mice. Unexpectedly, phospho-Smad2 and phospho-Smad3 were decreased in Smad7 -deficient mouse embryonic fibroblast (MEF) cells, whereas phospho-Smad1/5/8 was similarly expressed in wild-type and Smad7 -deficient MEF cells. Moreover, expression levels of TGF-β type I receptor (ALK5) were higher in Smad7 -deficient MEF cells than in wild-type MEF cells. Plasminogen activator inhibitor-1 (PAI-1) and inhibitor of differentiation-1 (Id-1) mRNA were similarly expressed in wild-type and Smad7 -deficient MEF cells. Some differences were observed in mitogen-activated protein kinase (MAPK)-signalling between wild-type and Smad7 -deficient MEF cells. We demonstrated that Smad7 plays an important role in normal mouse growth and provide a useful tool for analysing Smad7 functions in vivo .
