    Notes: Abstract Commercial polyvinylsilazane was crosslinked and then crushed to powder. The powder was compacted by cold isostatic pressing at 630 MPa and pyrolysed at 1050 °C in flowing argon. Crack-free Si–C–N material was obtained. Bulk density of the material was 1.95 Mg m−3. Open porosity was 9.6%. The material was amorphous as a result of X-ray diffraction analysis. Elastic modulus measured by pulse–echo method was 105 GPa. Vicker's hardness calculated from indentation at 98 MPa was 6.1 GPa. Fracture toughness measured by indentation fracture method was 2.1 MPa m1/2. Average bending strength was 118 MPa. The material shrank 1.9% in length during heating up to 1400 °C in nitrogen. The thermal expansion coefficient of the material heat treated up to 1400 °C increase from 3.08 × 10−6 °C−1 at 100 °C to 3.96 × 10−6 °C−1 at 1200 °C.

    Notes: Abstract Purpose. All transplanted solid organs experience some degree of ischemia-reperfusion (I-R) injury. This I-R injury can contribute to graft dysfunction which stems in part from the acute phase response and a resultant host of cytokines. Recent evidence suggests that organs remote to the site of I-R injury can be affected by circulating cytokines originating from these I-R injuries. Since many of these acute phase cytokines inhibit hepatic cytochrome P-450 (CYP) enzymes, we chose to investigate whether extrahepatic I-R injuries could influence hepatic oxidative drug metabolism. Methods. Fifteen dogs were divided into three surgical groups: (I) sham I-R; (II) bilateral normothermic renal I-R; and (III) normothermic intestinal I-R. Antipyrine (AP) was selected as a model substrate and administered intravenously at a dose of 10 mg/kg. AP serum concentrations were determined by HPLC and cytokine activity (IL-1, IL-6, and TNFα) was measured via bioassay. Serial AP clearance and serum cytokine concentrations were determined 3 days prior to and at 4 hr, 24 hr, 3 days and 7 days after surgery. Hematology and blood chemistries were monitored throughout the study period. Results. AP clearance was significantly reduced in groups II and III at 4 and 24 hrs post-I-R injury, while AP binding and apparent volume of distribution were unaffected. Peak levels of TNF and IL-6 activity occurred at 1 and 4 hours, respectively. IL-1 activity was not detected in any group. AP clearance correlated strongly to circulating levels of IL-6 (r = −0.789, p = 0.0002). Conclusions. Our findings indicate that extrahepatic I-R injury can affect hepatic oxidative drug metabolism and this effect is mediated in part by circulating cytokines.

    Notes: Abstract Genes encoding seven monoclonal antibody-defined cell-surface antigens have been regionally mapped on human chromosome 11, and compared to those of the AL complex defined by polyclonal antibodies using mutational analysis. MIC1, encoding W6/34 antigen, is probably identical to S1, previously mapped to 11pter-p13. MDU1 and MIC8, encoding 4F2 and TRA-1.10 antigens, respectively, are probably identical to S2(a4) and map to 11q13-q22. MIC9, which governs expression of 4D12 and 2E2 antigens, and maps to 11q22-qter, is not related to any of the five AL genes. MIC4 and MIC11, both mapping to 11pter-p13, may have some relationship to S3 and S1, respectively, but identity has not been proven.

    Notes: Abstract Purpose. Our laboratory has previously prepared gelatin/methotrexate (MTX) conjugates containing mixed conjugation sites and by-product crosslinking, both of which may alter conjugate effectiveness. In this study, we prepared and evaluated gelatin/MTX conjugates having specific conjugate bond sites and minimal by-product crosslinking. Methods. Opposite polarity conjugates were produced by coupling gelatin having blocked amino groups with MTX (G-MTX) and by coupling MTX having blocked amino groups with gelatin (M-GEL) using 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide HCl. Amino groups were blocked using citraconic anhydride and deblocked under acidic conditions. Gelatin and MTX contents were determined spectrophotometrically. The stability of each conjugate was determined by evaluating their in vitro release of MTX in isotonic buffer at pH 7.4 and 37°C for 7 days. Results. The G-MTX and M-GEL conjugates contained 21 and 1.2 mole MTX/mole gelatin and released 12 and 17% MTX by 7 days resulting in pseudo-first order release rate constants of 0.76×10−3 and 1.0×10−3 hr−1, respectively. Alternate MTX species (≤ 10%) were detected during the release study and were attributed to low molecular weight gelatin/MTX fragments and MTX polymers. Conclusions. Gelatin/MTX conjugates having opposite conjugate bond polarities and minimal by-product crosslinking have been produced and slowly released MTX by hydrolytic cleavage indicating good stability for future cell culture studies.

    Notes: Summary Dynamic secondary ion mass spectrometry (DSIMS) investigations have been carried out with Cr, Mn, Fe, Co, Mo, Rh, W, Re, Os and Ir under 4 mPa N2O, NO and 3 mPa NO2 as reactant gases. Results indicate similar behaviour in adsorption for Cr, Mn, Fe, Mo, W on the one hand and for Co, Rh, Os and Ir on the other. For the first group of metals the nitrogen oxide molecules are always totally destroyed in adsorption whereas the second group shows evidence for surface compounds such as MeNO (Me=metal) indicating only a partial dissociation in the case of N2O and NO2, and molecular adsorption under NO respectively. Re does not belong uniquely to either group because it reacts with N2O and NO2 dissociatively whereas under NO only partial dissociation is observed.
