    Notes: . The microsporidian parasite known as Nosema helminthorum Moniez, 1887, parasitic in the tapeworm Moniezia expansa (Rudolphi, 1810), has been shown by electron microscopy to have two cycles of development, one with isolated nuclei, the other with paired nuclei (diplokarya). Both merogony and sporogony of the two separate sequences take place in direct contact with the host cell cytoplasm and ultimately give rise to unikaryotic and diplokaryotic sporoblasts. Sporogony is disporoblastic. The nuclear condition of the spores was not seen. The sequences, corresponding to those of the genera Unikaryon and Nosema, may be part of a single dimorphic life cycle and, if so, the species will have to be transferred to a new genus.

    Notes: SUMMARY. The structure and life history of a new species of eugregarine, Gregarina garnhami n.sp., is described from the intestinal caeca and mid-gut of Schistocerca gregaria Forsk. The parasite destroys considerable areas of the caecal epithelium and in cases of heavy infection, the masses of parasites present in the mid-gut result in the formation of localized barriers between the gut wall and the food material in the lumen. The cephalont and sporont stages of gregarines from Locusta migratoria migratorioides R. & F. and Anacrydium aegyptium Linn. are shown to be similar to those from Schistocerca gregaria and are believed to belong to the same species.

    Notes: Until recently, pansporoblastic microsporidia that produce a variable and large number of sporoblasts from a sporont have been included in a single genus, namely Pleistophora Gurley, 1893. Ultrastructural studies have been used to determine whether the resemblance of these species is fundamental or superficial. The results indicated that the multisporous pansporoblastic forms belong to at least three genera and, thus, that Pleistophora is a “composite genus.” The term pansporoblast was originally used for stages in myxosporidian development. The term sporophorous vesicle adopted from Gurley is suggested for the spore-containing vesicle in the Microspora. Three species were studied: Pleistophora typicalis, the type-species; Pleistophora culicis, for which a new genus Vavraia has already been proposed; and Pleistophora simulii. P. typicalis and V. culicis have isolated nuclei throughout their development, and the sporophorous vesicle wall enveloping the sporoblasts is derived from amorphous secretions laid down during merogony external to the plasmalemma. Pleistophora and Vavraia are differentiated principally in terms of the structure of the sporophorous vesicle wall and mode of division of the sporogonial plasmodium. The nuclei of young sporonts of P. simulii are in diplokaryon arrangement and undergo meiosis to give haploid nuclei in the sporoblasts. The sporophorous vesicle wall is membranoid and is laid down external to the plasmalemma at the onset of sporogony. A new genus, Polydispyrenia n. g., is suggested for this species, the affinities of which are closer to the dimorphic species of microsporidia than to Pleistophora or Vavraia. The terms “merontogenetic sporophorous vesicle” and “sporontogenetic sporophorous vesicle” are used to distinguish between the two groups.

    Notes: . A species of Encephalitozoon has been isolated from the urine of a patient with the acquired immunodeficiency syndrome and maintained in vitro in Madin Darby Canine Kidney cells. When examined by random amplified polymoprhic DNA polymerase chain reaction the new isolate was found to differ from E. hellem and to have amplified products in common with murine and canine E. cuniculi. However, it more closely resembled the canine than the murine isolate. Sodium dodecylsulphate polyacrylamide gel electrophoresis differentiated between all three isolates of E. cuniculi, with a band at 42–45 kDa present in the murine isolate only, bands at 52 kDa present in the canine and human isolates but not the murine, and a single band at 60 kDa (murine) and 65 kDa (canine) replaced by two bands at 55 and 70 kDa in the human isolate. The 55 kDa and 70 kDa antigens were also revealed as characteristic bands of the human isolate by Western blotting. The study has thus revealed that the species Encephalitozoon cuniculi is not a homogeneous entity.

    Notes: . Collections of Mansonia africana mosquito larvae were made at one site in N.E. Tanzania in 1985 and 1987 and from two additional sites, both within about 2 mi of the original one in 1987. An octosporous microsporidian, present at all three sites, was found in both years infecting from 7 to 22% of larvae. Spores (stained in Giemsa) measured 3.0 μm × 0.25 μm × 2.25 μm × 0.26 μm. Ultrastructurally, spores were seen to have an anterior rim surrounding a depressed area where the endospore was at its thinnest. In transmission electron microscopy section, the rim appeared as two processes into which all layers of the wall extended. At the posterior end all layers of the wall extended into a simple knob-like structure which could be interpreted as a section through a crest running longitudinally around the spore. The polar filament was anisofilar, with two anterior coils of greater diameter than the three posterior coils. Although most closely resembling the genera Amblyspora and Parathelohania in the family Thelohaniidae, the species in M. africana differs from the former, which has oval spores, broadly rounded at the ends, and from the latter, which has a prominent, ridged posterior extension to the spores. The new species has been placed in a new genus and the name Tricornia muhezae proposed.

A new genus, Vittaforma n. g. is proposed for the human microsporidium Nosema corneum Shadduck, Meccoli, Davis & Font, 1990, based on the ultrastructure of developmental stages in the liver of experimentally infected athymic mice. The diplokaryotic arrangement of the nuclei is the only character that conforms with the description of the genus Nosema. Sporogony is polysporoblastic, sporonts are ribbon-shaped, constricting to give rise to linear arrays of sporoblasts and each parasite is enveloped by a complete cistema of host endoplasmic reticulum. Comparison of N. corneum, with established genera revealed that there were none with the same combination of characters. Consequently it is proposed that Nosema corneum be placed in a new genus as Vittaforma corneae n. comb.

    Notes: . Pyrotheca hydropsycheae n. sp. is described from caddis fly larvae, Hydropsyche siltalai Döhler, 1963. All stages were found in oenocytes and fat body cells. Meronts were uni- or binucleate with simple surface membranes. The sporogonic stages were recognized ultrastructurally by the separation of an envelope, the sporophorous vesicle, from their surfaces. Mature sporogonial plasmodia were tetranucleate and gave rise by longitudinal fission to four uninucleate elongate sporoblasts with polar nuclei. Spores were lageniform with an inflated posterior end, containing the polar tube coils and the nucleus, and a narrow anterior section comprising two-thirds of the length, containing the polaroplast and straight part of the polar tube. The polaroplast consisted of an anterior region of loosely packed membranes arranged as partitions at angles to one another and a posterior region of increasingly closely packed parallel membranes. The spore wall consisted of an electron-dense exospore with a fuzzy coat and a thin electron-lucent endospore. All four spores derived from a sporont faced in the same direction in the sporophorous vesicle. Spores measured 8.7 μm long and extruded polar filaments were about 20 μm.

    Notes: . Surface plaque matrix (PQM) and a tubular arrangement of filaments border Trachipleistophora hominis parasites during growth within host muscle. The PQM at the parasite surface forms a network of processes which can be associated with filamentous tubules. Peroxidase tracer delineated the PQM and showed apparent connections with the tubules. The tubules at the interface of T. hominis-infected cells are structurally similar to the extrasporular tubules of the microsporidian, Ameson michaelis. The extrasporular tubules of A. michaelis and the proteins from T. hominis-infected muscle reacted to keratin antibodies, K8.13, K4 and K13. Conversely, antibodies produced to T. hominis-infected muscle, reacted with the extrasporular tubular proteins of A. michaelis. The PQM and tubular elements are thought to play an important role in affecting molecular traffic between the host and parasite.
