    Notes: Summary Welwitschia mirabilis is a perennial desert plant with extremely large leaves (0.5–1.0 m broad, 1–2 m long). Leaf temperatures were measured in the field and the energy budget was calculated. The portions of the leaf which were kept above the ground had leaf temperatures which were only 4–6°C above air temperature. In the leaf portions which were in contact with the ground leaf temperatures were 6–12°C above air temperature (absolute maximum 51°C). The important feature in the energy budget ofWelwitschia mirabilis is its high reflectivity (38% of the global radiation). Only about 56% of the global radiation is absorbed by the thick leathery leaves. The energy loss due to convection is of the same order of magnitude as the reflection and it is abouy the same in the portions of leaf on and above the ground. The difference in leaf temperatures found in these portions is due to the loss of thermal radiation from the section of leaf above the ground to the cooler ground which is shaded by the leaf. The provision of a heat sink due to the large area of shade cast by these large leaves is of significance to the existence ofWelwitschia mirabilis in its arid habitats.

    Notes: Summary Increases in plant biomass are mainly a balance between growth of new leaves and growth of new roots, the new leaves having positive feedback upon the production process and the new roots having positive feedback upon the plant water status. Control of both opposing processes with respect to biomass production may be considered optimal whenever biomass of the plant reaches a maximum without adversely affecting plant water status. This occurs only if all carbohydrates are partitioned into growth of new leaves, unless water uptake is insufficient to meet the additional evaporative demand created by the newly grown leaf area without decreasing the water status of the plant. It is shown by theoretical considerations based upon optimization theory, especially by application of the Pontryagin Maximum Principle, that in this case carbohydrate partitioning is dependent upon the transpiration rate per leaf weight and upon the efficiency of the root at taking up water. Growth of Vigna unguiculata at two levels of air humidity and two levels of water uptake rate by the root was consistent with such a carbohydrate partitioning pattern. Growth of total biomass and its components (leaves, stems, and roots), whole plant transpiration, and the pattern of carbon partitioning were predicted and explained by applying the foregoing principles of optimization in a heuristic model for vegetative growth of an annual.

    Notes: Summary Tradescantia virginiana L. plants were cultivated under contrasting conditions of temperature, humidity, light quality and intensity, and nutrient status in order to investigate the effect of growth conditions on the water relations parameters of the leaf epidermal cells. Turgor pressure (P), volumetric elastic modulus (ɛ), half-time of water potential equilibration (T 1/2), hydraulic conductivity (L p ) were measured with the miniaturized pressure probe in single cells of the upper and lower epidermis of leaves. Turgor differed (range: 0.1 bar to 7.2 bar) between treatments with lowest values under warm and humid conditions and additional supply of fertilizer, and highest values under conditions of low air humidity and low nutrient supply. The volumetric elastic modulus changed by 2 orders of magnitude (range: 3.0 bar to 350 bar, 158 cells), but ɛ was only affected by the treatments, in as much as it was dependent on turgor. The turgor dependence of ɛ, measured on intact leaves of T. virginiana, was similar to that for cells of the isolated (peeled) lower epidermis, where ɛ as a function of turgor was linear over the whole range of turgors. This result has implications for the discussion of pressure/volume curves as measured by the pressure bomb where changes in “bulk leaf ɛ” are frequently discussed as “adaptations” to certain treatments. The measurements of the hydraulic conductivity indicate that this parameter varies between treatments (range of means: 2.4×10-6 cm s-1 bar-1 to 13.4×10-6 cm s-1 bar-1). There was a negative correlation for L p in cells of intact leaves as a function of turgor which was altered by the growing conditions. However, a correlation with turgor could not be found for cells from isolated epidermis or cells from a uniform population of plants. The large variation in L p from cell to cell observed in the present and in previous studies was accounted for in a study of 100 cells from a uniform population of plants by the propagation of measurement errors in calculating L p . The results suggest that in T. virginiana cellular water relations are changed mainly by the turgor dependence of ɛ.

    Notes: Summary The relationship of leaf biomass and leaf area to the conductive area of stems and branches was investigated in Picea abies. A total of 30 trees were harvested to determine if these relationships were different in different crown zones and in trees growing with and without competition for light. Two methods were compared. In the first, data were accumulated from crown zones situated at the top of trees to the bottom; in the second, data were used from individual crown zones. The results indicated that the latter method is much more sensitive in detecting differences in the relationship of leaf biomass or leaf area to conductive area. The analysis also indicated that ratios such as leaf area/sapwood area are frequently size-dependent. This size-dependency can in some cases result in the differences being abscured, but more often leads to the false impression that the relationship between the variables changes. The relationship between leaf biomass and leaf area and conductive area of stems or branches was different in different crown zones and under different growth conditions. The slopes of these regressions appear to increase with decreasing transpirational demand and decrease with increasing hydraulic conductivity. The intercepts are probably related to the amount of identified sapwood actually involved in water conductance.

    Notes: Summary Individual 33-year-old forest trees of the deciduous conifer speciesLarix decidua, Larix leptolepis andLarix decidua x leptolepis were investigated with respect to the phenomenon of stem heterosis in hybrid larch; the first part of this study compares the gas exchange responses of leaves. CO2 assimilation per leaf area was similar in the three larch species, but on a dry weight basis the nitrogen content of the needles and maximum CO2 assimilation rate (Amax) were slightly higher in the hybrid. This increase was accompanied by a higher protein content than in the Japanese and a lower specific leaf weight than in the European larch. All three species were similar in terms of the photosynthetic “nitrogen use” and stomatal conductance atA max. The similar slopes of the area-related steady-state responses of gas exchange against irradiance, evaporative demand and internal CO2 concentration led to similar rates of CO2 uptake under ambient conditions. The natural combinations and variability of the environmental factors also reduced the small dry weight-related difference inA max between hybrid larch and the parent species, such that all trees achieved similar daily carbon gains. Thus, the ecological significance of small interspecific differences in the metabolism of leaves has very little effect under the natural habitat conditions of a temperate climate. The second part of the study will investigate the effect of growth characteristics on the heterosis of hybrid larch.

    Notes: Summary Among 33-year-old forest trees ofLarix decidua, L. leptolepis andL. decidua x leptolepis, the hybrid possessed an above-ground biomass which was three times greater, although all larches displayed similar relative distributions of biomass. At a “relative growth rate” slightly lower than in the parent species, hybrid larch achieved twice the annual carbon gain, increment in stem length and above-ground production, and its foliage-related stem growth was higher than in European (L. decidua) but similar to Japanese (L. leptolepis) larch. A similar “relative growth efficiency” and foliage-related total above-ground production in all trees did reflect the similarity of photosynthetic capacity of the hybrid found at the leaf level. While the lengths of lateral twigs on hybrid branches were intermediate between the European larch with short, and the Japanese larch with large, twigs the hybrid possessed the longest branches with the highest needle biomass. This resulted in a crown structure of the hybrid crown similar to the Japanese larch together with a high needle density on branches as in the European larch. In total, the foliage biomass per crown length was about 30% higher in hybrid larch than in both of the parent species. Thus, the high carbon input for the stem heterosis was based on a “complementation principle” of advantageous parent features at the crown level. Similar slopes of foliage against sapwood area of stem and branches did not indicate a special need for a thick hybrid stem with respect to water transport.

    Notes: Summary Uptake of water and magnesium chloride solution was investigated through the outer surface of twigs of Picea abies (L.) Karst. Water uptake was determined by using pressure/volume (P/V) curves of the twigs as a basis for calculation to avoid problems of superficial extraneous water. When water was sprayed on bark and needles of 3- to 7-year-old twigs at a xylem water potential of -1.00 MPa, they absorbed as much as 80 mm3 water in 200 min/g twig dry weight as the twig water potential recovered to -0.15 MPa. With fluorescent dyes, pathways for absorption of water and solutes through the twig bark were found, particularly through the radially orientated ray tissue. In addition to uptake by mass flow, magnesium could also diffuse along a concentration gradient from the twig surface into the xylem. In the field, the magnitude of these uptake processes would depend on the concentration of elements deposited by atmospheric precipitation, the concentration gradient between the plant surface and the xylem sap, the xylem water potential and the intensity and duration of each precipitation event.

    Notes: Abstract  Interactions between the extracellular matrix (ECM) and chondrocytes are of great importance for structure and function of cartilage. The present study was undertaken to answer the question whether caveolins take part in integrin-mediated cell–ECM interactions in the human cartilage. In samples of human knee joint cartilage, we detected the caveolin subtypes -1, -2, and -3 by immunohistochemical methods. Double-label experiments revealed a colocalization of caveolin with β1-integrin. Results of immunoprecipitation and immunoblotting assays show that β1-integrins associate with all three caveolin subtypes in human chondrocytes and indicate that they are part of the same complexes. Furthermore, immunoelectron microscopy shows the localization of β1-integrin in caveolae-like structures of the cell membrane. The data stimulate further investigations on the role of the caveolin–integrin complex for integrin-mediated signaling pathways in chondrocytes.

    Notes: Abstract  Until now, many extracellular matrix proteins, e.g. osteopontin and osteonectin, have been used to determine a cell’s osteogenic maturation. The disadvantage in evaluation of these proteins is their relative wide-ranging appearance throughout the osteogenic differentiation process. Thus, the aim of this study was to establish an immunohistochemical setup using E11, a marker that binds selectively to cells of the late osteogenic cell lineage. In addition, the histochemical expression of the bone matrix proteins osteonectin, osteopontin and fibronectin was compared to that of E11 using monoclonal antibodies. For light microscopical detection of osteogenic markers in cultured cells we developed a simple paraffin technique using a fibrin glue as embedding medium. This allows the handling of cultured cells such as a tissue sample and includes the use of stored biological specimens for further immunohistochemical experiments. We used newborn rat calvariae for whole tissue preparations and for isolation and cultivation of bone cells. In addition, we included the rat osteosarcoma cell line ROS 17/2.8 in this study. For the first time, we have localised E11 in osteocytes of rat calvaria preparations at the electron microscopical level. E11 was detected at plasma membranes of osteocytes and their processes, but not at those of osteoblasts. Accompanying experiments with cultured newborn rat calvaria cells and ROS 17/2.8 cells revealed E11 reactivity on a subset of cells. The results obtained confirm the suitability of the differentiation marker E11 as a sensitive instrument for the characterisation of bone cell culture systems.

    Notes: Summary Experiments with Prunus armeniaca were carried out under conditions of constant temperature but varying air humidity. Experiments were also contucted with a constant water vapor difference between the evaporating sites in a leaf and the air, but with varying leaf temperature. These served as a basis for predicting the daily course of total diffusion resistance under the natural climatic conditions of a desert. For the simulation, the rsults of the experiments at constant conditions with only one variable factor are fitted with empirical equations which serve as “calibration curves” to predict the change in diffusion resistance caused by a change in humidity and temperature calculated from the meteorological data of a desert day. The simulation shows that for P. armeniaca humidity and temperature are the dominating factors in controlling the daily course of diffusion resistance. For meteorologically very different days the simulation allows the increase in diffusion resistance in the morning to be predicted with an accuracy of 90%–105% as compared to directly observed measurements. In the afternoon, especially after extreme climatic conditions during the morning, the deviation between predicted and observed values of diffusion resistance may be greater, but not more than -20% to -30%. This possibly indicates the existence of an additional factor of significance which was not included in the simulation. The two peaked curves of net photosynthesis and transpiration characteristic of plants living under arid conditions can be explained in this species by the humidity-and temperature-controlled stomatal response. This stomatal regulation leads to a decreasing total daily transpirational water loss on a dry day as compared to a moist one. The significance of this controlling mechanism for the primary production and the water relations of P. armeniaca is discussed.
