    Notes: Undrawn nylon was found to be susceptible within a few days to degradation in wet or humid conditions at temperatures between 50 and 90°C. Model N-substituted amides, although not affected by these conditions, were attacked by hydrogen peroxide solutions. The degradation proceeded principally by rupture of the bond between the nitrogen of the amide and the carbon of its α-methylene group, consequent upon oxidation at this carbon. Unsubstituted primary amide, carboxyl, and aldehyde endgroups were formed, together with ammonia by hydrolysis of the oxidized amide. The bond between the α and β carbons of an N-methylene chain also suffered breakage, giving formic acid and carbon dioxide. The former could also arise by rupture of any initially produced N-formyl or N-formoyl amides. The appearance of lactic acid from butylacetamide showed that oxidation could proceed along the N-alkyl chain. Fragments of suitable size broken from the main chain of nylon became water-soluble and relatively stable to further attack by air or oxygen. These pieces essentially retained the nylon structure, but possessed two carboxylic ends and an average molecular weight of less than 600. A maximum oxygen uptake of about 30 moles per mole of original amide was calculated, corresponding to complete conversion into these soluble fragments. Significant degradation did not occur for many months at 60°C. in dry conditions or in solutions of 8-hydroxyquinoline, a process which has been patented. The latter was considered to function by chelating heavy metals into an inactive form and inhibiting peroxide formation.

    Notes: The exocellular polysaccharide fermented from glucose in good yield by Xanthomonas campestrìs NRRL B-1459, has been characterized. The general aspects of chemical constitution have been established, as well as the physical properties related to practical applicability. This macromolecular polysaccharide is composed of D-mannose, D-glucose, D-glucuronic acid (as the potassium salt), and a small proportion of acetyl groups. It can be produced on an industrial scale and is stable is storage. Analytical fractionation indicates fairly sharp molecular distribution for the native polysaccharide. The polysaccharide forms homogeneous dispersions in water which show plastic rheological properties and viscosity comparable with that of high-grade plant gums. Outstanding characteristics of practical significance are the atypical insensitivity of solution viscosity to salt effects and to heat, especially when salt is present. Solutions of low concentration show a restricted viscosity decrease upon salt addition; those of higher concentrations show substantial increases. Viscosity is enhanced still further by monovalent cations at basic pH and by divalent cations at neutral or slightly basic pH. Salt moderates or eliminates any viscosity decrease due to heat and, in somewhat higher concentrations, it increases the viscosity of heated solutions. Heating or deacetylating Polysaccharide B-1459 causes no impairment of its properties, but actual improvement. The constitutional basis for these unusual properties is discussed.

    Notes: A process has been developed giving over-all yields of about 50%: which could be increased to about 80% with higher material costs and processing time. The preferred method of recovering nucleotides from the cytoplasm has been thermal extraction by means of a heat exchanger followed by filtration, protein precipitation, and carbon adsorption. The concentrated eluate from the carbon is absorbed onto Dowex 1 ion-exchange resin, and groups of nucleotides are removed by pH-defined eluents and recovered by a second carbon adsorption. Finally, standard ion-exchange resolutions have been used to recover individual nucleotides. Other biochemicals in the extract have been investigated, and preliminary work has been done on the combination of nucleotide extraction with the recovery of nucleic acids from the residual cells.

    Notes: The design of a cabinet for the aseptic manipulation of material is described. The interior surfaces of the cabinet are sterilized by swabbing with a germicide. Germ-free conditions are maintained during operation by means of a positive pressure of sterile air. A fan and filter unit attached to the cabinet is used to blow sterile air into the cabinet in sufficient quantity so as to flow out of the hand ports at a velocity of 50 ft./min. In the design, careful consideration was given to the ease of manipulations in the cabinet.

    Notes: The formation of a solid phase as the result of isothermally increasing pressure on a binary solution is analyzed from a thermodynamic point of view. Exact thermodynamic equations are presented for the case in which there are no more than one liquid and one solid phase in equilibrium for the entire range of compositions. Approximate relations incorporating the additional assumption of ideality in both solid and liquid phases are also presented. Experimental data for the system ethylene bromide-ethylene chloride are reported and compared with predicted values. Incomplete data for the system benzene-n-heptane are also reported.

    Notes: In experimental investigations of the transient behavior of batch thermogravitational thermal diffusion columns it is expedient to obtain more than one sample from the column during a single run. Such sampling modifies the true batch nature of the column and thereby influences the experimental measurements to some degree. A combined theoretical and experimental investigation was carried out to determine the extent of the influence of sampling rate and thereby provide a means of predetermining a suitable sampling rate.The theoretical analysis was made by approximating the intermittent sampling as a continuous flow through the column. The transport equation of Furry, Jones, and Onsager was applied in the analysis, and an analytical solution was obtained for small separations of equicomposition binary solutions. The series solution is presented in the form of a graph. Experimental data were obtained in two thermogravitational columns with different plate spacings to test the theory. The effect of sampling rate was investigated, and theory and experiments were found to be in good agreement. It is concluded that the theory is entirely adequate to permit the prediction of sampling rates which will yield the maximum number of samples without disturbing the true batch behavior of the column.

    Notes: A study was made of the effects the surface tension of aqueous solutions of surface-active agents had on the bubble-formation characteristics of these solutions. Bubbles were formed by two processes: by passing air through a horizontal orifice submerged in the solution and by nucleate boiling at an electrically heated surface. The experimental measurements taken with the air-bubble system were used to determine the nature of the surface-tension effect on air-bubble formation. This knowledge was then applied to the interpretation of boiling measurements made with the same solutions. The boiling coefficients of the Tergitol-water solution were found to vary inversely with dynamic surface tension. Boiling coefficients for Aerosol-water solutions, however, varied in a manner which could not be related to surface-tension effects.

    Notes: The effects of liquid superheat and surface roughness on boiling coefficients were investigated in a series of experiments in which water, acetone, n-hexane, carbon tetrachloride, and carbon disulfide were boiled on a flat plate. In addition to the usual thermal measurements, the number of active boiling centers was determined, whenever possible, by visual means, and a quantitative measure of surface roughness was made. It was found that the number of active boiling centers on the plate increased with increasing surface roughness and that the calculated boiling coefficients were proportional to the one-third power of the number of bubble columns rising from the heated surface.An equation has been derived relating boiling coefficients to fluid properties and the number of active boiling centers on a surface. A second equation, based on the theory of thermal fluctuations, has been proposed to relate the number of active boiling centers to the independent variables of surface-roughness and temperature-difference driving force. The limited data available have been found to follow this proposed relation.The results of this work suggest a quantitative method of relating the boiling coefficient to the character of the surface which may explain the discrepancies observed in the slopes of boiling curves reported in the literature and in the actual values reported for the boiling coefficients measured on different surfaces.
